Seed-Mediated Synthesis of Gold Nanorods at Low Concentrations of CTAB.

ABSTRACT: Although gold nanorods capped with hexadecyltrimethylammonium bromide (CTAB) have been prepared through the seed-mediated method for their use in diagnostics and therapeutics, the toxicity of AuNRs@CTAB limits their practical applications in the biomedical field. In this work, the synthesis and tuning of gold nanorods at very low concentrations of CTAB (as low as 0.008 M) was successfully achieved by using the seed-mediated method. Furthermore, we managed to optimize the growth conditions by changing the amount of seeds, AgNO₃, and/or HCl. At low CTAB concentrations, gold nanorods with tunable size and aspect ratio, high monodispersity, and high purity were obtained and studied by UV-vis spectroscopy, transmission electron microscopy, and Mie-Gans theoretical calculations. This work revealed a method of nanoparticle growth that may be extended to synthesize other nanomaterials such as Ag, Cu, Pd, and Pt at such low CTAB concentrations.

BSA modification to reduce CTAB induced nonspecificity and cytotoxicity of aptamer-conjugated gold nanorods.

Project description:Aptamer-conjugated gold nanorods (AuNRs) are excellent candidates for targeted hyperthermia therapy of cancer cells. However, in high concentrations of AuNRs, aptamer conjugation alone fails to result in highly cell-specific AuNRs due to the presence of positively charged cetyltrimethylammonium bromide (CTAB) as a templating surfactant. Besides causing nonspecific electrostatic interactions with the cell surfaces, CTAB can also be cytotoxic, leading to uncontrolled cell death. To avoid the nonspecific interactions and cytotoxicity triggered by CTAB, we report the further biologically inspired modification of aptamer-conjugated AuNRs with bovine serum albumin (BSA) protein. Following this modification, interaction between CTAB and the cell surface was efficiently blocked, thereby dramatically reducing the side effects of CTAB. This approach may provide a general and simple method to avoid one of the most serious issues in biomedical applications of nanomaterials: nonspecific binding of the nanomaterials with biological cells.

Time Optimization of Seed-Mediated Gold Nanotriangle Synthesis Based on Kinetic Studies.

Project description:The synthesis of shape-anisotropic plasmonic nanoparticles such as gold nanotriangles is of increasing interest. These particles have a high potential for applications due to their notable optical properties. A key challenge of the synthesis is usually the low reproducibility. Even the optimized seed-based methods often lack in the synthesis yield or are labor- and time-consuming. In this work, a seed-mediated synthesis with high reproducibility is replicated in order to determine the necessary reaction time for each step. Online monitoring of the reaction mixtures by UV-VIS spectroscopy is used as a powerful tool to track the evolution of the synthesis. The kinetics of the individual stages is elucidated by real-time investigations. As a consequence, the complete synthesis could be optimized and can now be realized in a single day instead of three without any loss in the resulting sample quality.

Gold nanorods derivatized with CTAB and hydroquinone or ascorbic acid: spectroscopic investigation of anisotropic nanoparticles of different shapes and sizes.

Project description:Gold nanorods stabilized by binary ligand mixtures of cetyltrimethylammonium bromide (CTAB, primary ligand) and ascorbic acid or hydroquinone were investigated by complementary synchrotron radiation-induced spectroscopies and microscopies, with the aim to find evidence of the influence of the secondary ligand molecular and chemical structure on the nanorod shapes and size ratios. Indeed, as it is well known that the CTAB interaction with Ag(i) ions at the NR surface plays a key role in directing the anisotropic growth of nanorods, the possibility to finely control the NR shape and dimension by opportunely selecting the secondary ligands opens new perspectives in the design and synthesis of anisotropic nanoparticles.

Project description:The immune system may respond to engineered nanomaterials (ENM) through inflammatory reactions. The NLRP3 inflammasome responds to a wide range of ENM, and its activation is associated with various inflammatory diseases. The objective of the study was to compare the effects of gold ENM of different shapes on NLRP3 inflammasome activation and related signalling pathways. Differentiated THP-1 cells (wildtype, ASC- or NLRP3-deficient), were exposed to PEGylated gold nanorods, nanostars, and nanospheres. Exposed cells were subjected to gene expression analysis. Nanorods, but not nanostars or nanospheres, showed NLRP3 inflammasome activation. ASC- or NLRP3-deficient cells did not show this effect. Gold nanorod-induced NLRP3 inflammasome activation was accompanied by downregulated sterol/cholesterol biosynthesis, oxidative phosphorylation, and purinergic receptor signalling. In conclusion, the shape and surface chemistry of gold nanoparticles determine NLRP3 inflammasome activation.

Wavelength-Dependent Plasmon-Mediated Coalescence of Two Gold Nanorods.

Project description:Plasmon-mediated coalescence of two nearby gold nanorods (NRs) suspended in water induced by the illumination of a linearly polarized (LP) light was studied theoretically. We analyzed the coupled optical forces and torques in terms of Maxwell's stress tensor upon two identical NRs irradiated by a LP plane wave using the multiple multipole method to estimate the optomechanical outcome. Numerical results show that the light-matter interaction can perform attraction or repulsion, depending on their initial configurations. For the attraction, the end-to-end or side-by-side coalescence of the two gold NRs could be caused by the LP light, depending on the wavelength. For example, the side-by-side coalescence of two adjacent NRs of r = 15 nm and L = 120 nm is most likely induced by 800-nm LP laser beam, whereas the end-to-end coalescence by 1064-nm or 1700-nm LP laser. These distinct phenomena are attributed to the perpendicular or parallel alignment of NR to the polarization of LP light in different wavelength ranges. The magnitude of optical force, proportional to the light's fluence, could be stronger than van der Waals force. The estimation based on quasi-static model without considering the fluid dynamics may provide an insight to optical manipulation on the self-assembly of gold colloid.

Large-Scale Synthesis of Gold Nanorods through Continuous Secondary Growth.

Project description:Gold nanorods (GNRs) exhibit a tunable longitudinal surface plasmon resonance (LSPR) that depends on the GNR aspect ratio (AR). Independently controlling the AR and size of GNRs remains challenging but is important because the scattering intensity strongly depends on the GNR size. Here, we report a secondary (seeded) growth procedure, wherein continuous addition of ascorbic acid (AA) to a stirring solution of GNRs, stabilized by cetyltrimethylammonium bromide (CTAB) and synthesized by a common GNR growth procedure, deposits the remaining (~70%) of the Au precursor onto the GNRs. The growth phase of GNR synthesis is often performed without stirring, since stirring has been believed to reduce the yield of rod-shaped nanoparticles, but we report that stirring coupled with continuous addition of AA during secondary growth allows improved control over the AR and size of GNRs. After a common primary GNR growth procedure, the LSPR of GNRs is ~820 nm, which can be tuned between ~700-880 nm during secondary growth by adjusting the rate of AA addition or adding benzyldimethylhexadecylammonium chloride hydrate (BDAC). This approach for secondary growth can also be used with primary GNRs of different ARs to achieve different LSPRs and can likely be extended to nanoparticles of different shapes and other metals.

Additive-Free Synthesis of (Chiral) Gold Bipyramids from Pentatwinned Nanorods.

Project description:The production of colloidal metal nanostructures with complex geometries usually involves shape-directing additives, such as metal ions or thiols, which stabilize high-index facets. These additives may however affect the nanoparticles' surface chemistry, hindering applications, e.g., in biology or catalysis. We report herein the preparation of gold bipyramids with no need for additives and shape yields up to 99%, using pentatwinned Au nanorods as seeds and cetyltrimethylammonium chloride as surfactant. For high-growth solution:seed ratios, the bipyramids exhibit an unusual "belted" structure. Three-dimensional electron microscopy revealed the presence of high-index {117}, {115}, and {113} side facets, with {113} and {112} facets at the belt. Belted bipyramids exhibit strong near-field enhancement and high extinction in the near-infrared, in agreement with electromagnetic simulations. These Ag-free bipyramids were used to seed chiral overgrowth using 1,1'-binaphthyl-2,2'-diamine as a chiral inducer, with g-factor up to 0.02, likely the highest reported for bipyramid seeds so far.

Seed-Mediated Synthesis of Thin Gold Nanoplates with Tunable Edge Lengths and Optical Properties.

Project description:Thin Au nanoplates show intriguing localized surface plasmon resonance (LSPR) properties with potential applications in various fields. The conventional synthesis of Au nanoplates usually involves the formation of spherical nanoparticles or produces nanoplates with large thicknesses. Herein, we demonstrate a synthesis of uniform thin Au nanoplates by using Au-Ag alloy nanoframes obtained by the galvanic replacement of Ag nanoplates with HAuCl4 as the seeds and a sulfite (SO32-) as a ligand. The SO32- ligand not only complexes with the Au salt for the controlled reduction kinetics but also strongly adsorbs on Au {111} facets for effectively constraining the crystal growth on both basal sides of the Au nanoplates for controlled shape and reduced thicknesses. This seed-mediated synthesis affords Au nanoplates with a thickness of only 7.5 nm, although the thickness increases with the edge length. The edge length can be customizable in a range of 48-167 nm, leading to tunable LSPR bands in the range of 600-1000 nm. These thin Au nanoplates are applicable not only to surface-enhanced Raman spectroscopy with enhanced sensitivity and reliability but also to a broader range of LSPR-based applications.

The effect of gold nanorods on cell-mediated collagen remodeling.

Project description:Cardiac fibroblasts, the noncontractile cells of the heart, contribute to myocardial maintenance through the deposition, degradation, and organization of collagen. Adding polyelectrolyte-coated gold nanorods to three-dimensional constructs composed of collagen and cardiac fibroblasts reduced contraction and altered the expression of mRNAs encoding beta-actin, alpha-smooth muscle actin, and collagen type I. These data show that nanomaterials can modulate cell-mediated matrix remodeling and suggest that the targeted delivery of nanomaterials can be applied for antifibrotic therapies.

Bottom-Up Then Top-Down Synthesis of Gold Nanostructures Using Mesoporous Silica-Coated Gold Nanorods.

Project description:Gold nanostructures were synthesized by etching away gold from heat-treated mesoporous silica-coated gold nanorods (AuNR@mSiO2), providing an example of top-down modification of nanostructures made using bottom-up methodology. Twelve different types of nanostructures were made using this bottom-up-then-top-down synthesis (BUTTONS), of which the etching of the same starting nanomaterial of AuNR@mSiO2 was found to be controlled by how AuNR@mSiO2 were heat treated, the etchant concentration, and etching time. When the heat treatment occurred in smooth moving solutions in round-bottomed flasks, red-shifted longitudinal surface plasmon resonance (LSPR) was observed, on the order of 10-30 min, indicating increased aspect ratios of the gold nanostructures inside the mesoporous silica shells. When the heat treatment occurred in turbulent solutions in scintillation vials, a blue shift of the LSPR was obtained within a few minutes or less, resulting from reduced aspect ratios of the rods in the shells. The influence of the shape of the glassware, which may impact the flow patterns of the solution, on the heat treatment was investigated. One possible explanation is that the flow patterns affect the location of opened pores in the mesoporous shells, with the smooth flow of solution mainly removing CTAB surfactants from the pores along the cylindrical body of mSiO2, therefore increasing the aspect ratios after etching, and the turbulent solutions removing more surfactants from the pores of the two ends or tips of the silica shells, hence decreasing the aspect ratios after etching. These new stable gold nanostructures in silica shells, bare and without surfactant protection, may possess unique chemical properties and capabilities. Catalysis using heat-treated nanomaterials was studied as an example of potential applications of these nanostructures.
