Flexible polyfluorinated bis-diazirines as molecular adhesives.

ABSTRACT: Motivated by a desire to develop flexible covalent adhesives that afford some of the same malleability in the adhesive layer as traditional polymer-based adhesives, we designed and synthesized two flexible, highly fluorinated bis-diazirines. Both molecules are shown to function as effective crosslinkers for polymer materials, and to act as strong adhesives when painted between two polymer objects of low surface energy, prior to thermal activation. Data obtained from lap-shear experiments suggests that greater molecular flexibility is correlated with improved mechanical compliance in the adhesive layer.

Biocatalytic Carbene Transfer Using Diazirines.

Project description:Biocatalytic carbene transfer from diazo compounds is a versatile strategy in asymmetric synthesis. However, the limited pool of stable diazo compounds constrains the variety of accessible products. To overcome this restriction, we have engineered variants of Aeropyrum pernix protoglobin (ApePgb) that use diazirines as carbene precursors. While the enhanced stability of diazirines relative to their diazo isomers enables access to a diverse array of carbenes, they have previously resisted catalytic activation. Our engineered ApePgb variants represent the first example of catalysts for selective carbene transfer from these species at room temperature. The structure of an ApePgb variant, determined by microcrystal electron diffraction (MicroED), reveals that evolution has enhanced access to the heme active site to facilitate this new-to-nature catalysis. Using readily prepared aryl diazirines as model substrates, we demonstrate the application of these highly stable carbene precursors in biocatalytic cyclopropanation, N-H insertion, and Si-H insertion reactions.

Photocurable bioresorbable adhesives as functional interfaces between flexible bioelectronic devices and soft biological tissues.

Project description:Flexible electronic/optoelectronic systems that can intimately integrate onto the surfaces of vital organ systems have the potential to offer revolutionary diagnostic and therapeutic capabilities relevant to a wide spectrum of diseases and disorders. The critical interfaces between such technologies and living tissues must provide soft mechanical coupling and efficient optical/electrical/chemical exchange. Here, we introduce a functional adhesive bioelectronic-tissue interface material, in the forms of mechanically compliant, electrically conductive, and optically transparent encapsulating coatings, interfacial layers or supporting matrices. These materials strongly bond both to the surfaces of the devices and to those of different internal organs, with stable adhesion for several days to months, in chemistries that can be tailored to bioresorb at controlled rates. Experimental demonstrations in live animal models include device applications that range from battery-free optoelectronic systems for deep-brain optogenetics and subdermal phototherapy to wireless millimetre-scale pacemakers and flexible multielectrode epicardial arrays. These advances have immediate applicability across nearly all types of bioelectronic/optoelectronic system currently used in animal model studies, and they also have the potential for future treatment of life-threatening diseases and disorders in humans.

Labeling Preferences of Diazirines with Protein Biomolecules.

Project description:Diazirines are widely used in photoaffinity labeling (PAL) to trap noncovalent interactions with biomolecules. However, design and interpretation of PAL experiments is challenging without a molecular understanding of the reactivity of diazirines with protein biomolecules. Herein, we report a systematic evaluation of the labeling preferences of alkyl and aryl diazirines with individual amino acids, single proteins, and in the whole cell proteome. We find that alkyl diazirines exhibit preferential labeling of acidic amino acids in a pH-dependent manner that is characteristic of a reactive alkyl diazo intermediate, while the aryl-fluorodiazirine labeling pattern reflects reaction primarily through a carbene intermediate. From a survey of 32 alkyl diazirine probes, we use this reactivity profile to rationalize why alkyl diazirine probes preferentially enrich highly acidic proteins or those embedded in membranes and why probes with a net positive charge tend to produce higher labeling yields in cells and in vitro. These results indicate that alkyl diazirines are an especially effective chemistry for surveying the membrane proteome and will facilitate design and interpretation of biomolecular labeling experiments with diazirines.

Diazirines as Potential Molecular Imaging Tags: Probing the Requirements for Efficient and Long-Lived SABRE-Induced Hyperpolarization.

Project description:Diazirines are an attractive class of potential molecular tags for magnetic resonance imaging owing to their biocompatibility and ease of incorporation into a large variety of molecules. As recently reported, 15 N2 -diazirine can be hyperpolarized by the SABRE-SHEATH method, sustaining both singlet and magnetization states, thus offering a path to long-lived polarization storage. Herein, we show the generality of this approach by illustrating that the diazirine tag alone is sufficient for achieving excellent signal enhancements with long-lasting polarization. Our investigations reveal the critical role of Lewis basic additives, including water, on achieving SABRE-promoted hyperpolarization. The application of this strategy to a 15 N2 -diazirine-containing choline derivative demonstrates the potential of 15 N2 -diazirines as molecular imaging tags for biomedical applications.

Molecular characterization of adhesives (glue lining pastes) used in restoration

Project description:The molecular characterization of samples from works of art can provide valuable insights into the composition of ancient restoration materials and their conservation state. Here, we present a novel experimental protocol for the molecular characterization of a specific adhesive used in historical painting restoration, known as "glue lining pastes." Due to the high molecular complexity of these adhesives, we propose a multi-step extraction protocol to simultaneously recover and fractionate from a single microsample the three main classes of biomolecules contained in glue pastes (lipids, polysaccharides, and proteins). High-performance separation coupled with high-resolution MS techniques were applied to the isolated fractions to identify specific components. The proposed method was optimized using test specimens of various traditional glue pastes applied to canvases and successfully applied to a historical glue paste sample from the 17th-century painting "La fuga in Egitto," part of the Pagliara collection at the University Suor Orsola Benincasa (Naples, Italy). The data collected in this work provide insights into the specific recipe used for adhesive preparation, supporting artistic and historical interpretations, and contributing to a broader understanding of old restoration practices.

Molecular architecture regulation for the design of instant and robust underwater adhesives.

Project description:Development of underwater adhesives with instant and robust adhesion to diverse substrates remains challenging. A strategy taking the structural advantage of phenylalanine derivative, N-acryloyl phenylalanine (APA), is proposed to facilely prepare a series of underwater polymeric glue-type adhesives (UPGAs) through one-pot radical polymerization with commonly used hydrophilic vinyl monomers. The adjacent phenyl and carboxyl groups in APA realize the synergy between interfacial interactions and cohesion strength, by which the UPGAs could achieve instant (~5 seconds) and robust wet tissue adhesion strength (173 kilopascal). The polymers with varied hydrophobicity and substitutional groups as well as carboxyl and phenyl groups in separated components are designed to investigate the underwater adhesion mechanism. The universality of APA for the construction of UPGAs is also verified by the copolymerization with different hydrophilic monomers, and the applications of the UPGAs have been validated in diverse hemorrhage models and distinct substrates. Our work may give a promising solution to design potent underwater adhesives.

Bulky Polyfluorinated Terphenyldiphenylboranes: Water Tolerant Lewis Acids.

Project description:Protocols for the synthesis of the bulky polyfluorinated triarylboranes 2,6-(C6 F5 )2 C6 F3 B(C6 F5 )2 (1), 2,6-(C6 F5 )2 C6 F3 B[3,5-(CF3 )2 C6 H3 ] (2), 2,4,6-(C6 F5 )3 C6 H2 B(C6 F5 )2 (3), 2,4,6-(C6 F5 )3 C6 H2 B[3,5-(CF3 )2 C6 H3 ] (4) were developed. All boranes are water tolerant and according to the Gutmann-Beckett method, 1-3 display Lewis acidities larger than that of the prominent B(C6 F5 )3 .

Insight into the Molecular Weight of Hydrophobic Starch Laurate-Based Adhesives for Paper.

Project description:Instead of using finite petroleum-based resources and harmful additives, starch can be used as a biodegradable, low-cost, and non-toxic ingredient for green adhesives. This work employs K3PO4 catalyzed transesterifications of cassava starch and methyl laurate at varying reaction times (1-10 h), resulting in the enhanced hydrophobicity of starch laurates. At longer reaction times, starch laurates having higher degrees of substitution (DS) were obtained. While starch laurates are the major products of transesterification, relatively low-molecular-weight byproducts (1%) were detected and could be hydrolyzed starches based on gel permeation chromatography results. Contact angle measurements confirmed the relatively high hydrophobicity of the modified starches compared with that of native starch. The modified starches were then employed to prepare water-based adhesives on paper (without any additional additives). Notably, the shear strength of the esterified starch adhesives appears to be independent of the DS of esterified samples, hence the transesterification reaction times. Additionally, the shear strength of water-based adhesives (0.67-0.73 MPa) for bonding to paper substrates is superior to that of two other commercially available glues by a factor of 10 to 80 percent.
