The complete mitochondrial genome of Oestrus ovis (Linnaeus, 1758) (Diptera: Oestridae).

ABSTRACT: Larvae of the sheep bot fly, Oestrus ovis (Linnaeus, 1758), cause cavitary myiasis in domestic and wild hosts, including man. The complete circular O. ovis mitogenome was assembled, which is 16,584 bp in length and encodes 13 protein-coding genes, 22 tRNA genes, and two rRNA genes. The phylogenetic tree of O. ovis and 13 related Oestridae species, with Sarcophaga tuberosa as outgroup, was built.

Project description:In this study, the complete mitochondrial genome (mitogenome) of Melanostoma mellinum (Linnaeus, 1758) was sequenced using the-next generation sequencing technology. The assembled mitogenome of M. mellinum has a total length of 16,055bp and contains 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), and 2 ribosomal RNA genes (rRNAs). The results of phylogenetic reconstruction based on the combined mitochondrial gene dataset indicated that M. mellinum belongs to Melanostoma genus with a close relationship to Melanostoma orientale, but the monophyly of the tribe Bacchini is not well supported.

The complete mitochondrial genome of Syritta pipiens (Linnaeus, 1758) (Diptera: Syrphidae) and phylogenetic analysis.

Project description:The complete mitochondrial genome (mitogenome) of Syritta pipiens (Linnaeus, 1758) was sequenced with 15,745 bp in length including 37 genes and a non-coding region. The overall nucleotide composition showed a strong AT bias. Most protein-coding genes (PCGs) used ATN as the start codon while ATP6 and ND1 used TTG, and stopped by TAA or TAG but ND5 ended with an incomplete T. Phylogenetic trees were reconstructed based on the 24 complete mitochondrial sequences from Syrphidae using the methods of maximum-likelihood (ML) and Bayesian inference (BI), resulted in S. pipiens clustered into the clade of Eristalinae, which conformed to the traditional classification, but the trees did not support the monophyly of Eristalinae. More molecular data is needed for further study.

Complete mitochondrial genome of Trachycardium flavum (Linnaeus, 1758).

Project description:The complete mitochondrial genome of Trachycardium flavum from South China Sea was first determined in this study, which is 16,596 bp in length. The base composition of the mitogenome is a less biased (A, G, T, and C was 21.5%, 26.3%, 37.0%, and 15.2%, respectively) with A + T contents of 58%. It consists of 24 tRNA genes, two rRNA genes, and 12 protein-coding genes (PCGs). No typical control region was found in non-coding sequence. Interestingly, ATP8 genes was absence same as most shellfish. Nine PCGs use a special start codon TTG, and the other three genes use a normal initiation codon ATN. Except ND6 use a single base G as the stop codon, the others all end with the normal stop codon TAG. The phylogenetic tree showed that T. flavum was first clustered with Fulvia mutica, Cerastoderma edule, and Acanthocardia tuberculate, which all belong to Cardiidae.

Prevalence Rate and Molecular Characteristics of Oestrus ovis L. (Diptera, Oestridae) in Sheep and Goats from Riyadh, Saudi Arabia.

Project description:Heads of sheep (n = 600) and goats (n = 800) slaughtered at Al-Aziziah Abattoir in Riyadh, Saudi Arabia, were inspected for the presence of O. ovis larvae (L). Heads were split along the longitudinal axes, and larvae (L1, L2, and L3) were gathered. The infestation rate was significantly higher in goats (44.5%; 356/800) than that in sheep (22.3%; 134/600). Out of the 151 collected larvae from sheep, 0% were L1, 1.3% were L2, and 98.7% were L3. Out of the total of 468 larvae from goats, 0% were L1, 1.2% were L2, and 98.8% were L3. The infestation rate was significantly higher in males than that in females. Myiasis-causing larvae collected from Riyadh, Saudi Arabia, were authenticated as O. ovis, according to morphological characteristics. Polymerase chain reaction (PCR) amplification of a partial fragment (600 bp) of the mitochondrial cytochrome c oxidase subunit I (mtCOI) gene further confirmed the species. Phylogenetic analysis based on the partial mtCOI gene sequence demonstrated that 23 unique sequences showed high similarity based on nucleotide pairs of O. ovis accessions retrieved from GenBank.

Complete mitochondrial genome of the cockle Anadara antiquata (Linnaeus, 1758).

Project description:The complete mitochondrial genome of Anadara antiquata was first determined. With a length of 45,454 bp, it consists of 29 tRNA, 2 rRNA, and 17 protein-coding genes (PCGs). The non-coding region was large and atypical around the genome with total 24,162 bp long. The nucleotide composition is significantly biased with AT contents of 62.2%. PCGs have five types of start codon, and terminate with a complete stop codon TAA or TAG. 19 microsatellites (SSRs) were identified in mitogenome sequences. Phylogenetic analysis demonstrated that A. antiquata was first clustered with Anadara vellicate, then together with Tegillarca granosa.

Complete mitochondrial genome of tiger cowrie Cypraea tigris (Linnaeus, 1758).

Project description:The complete mitochondrial genome of Cypraea tigris from the South China Sea has been determined, which was the first report of complete mitogenome in the superfamily Cypraeoidea. It is 16,177 bp long and consists of 21 tRNA genes, 2 rRNA genes, 13 protein-coding genes (PCGs), and 1 control region. The base composition of C. tigris mitogenome is biased (A, G, T, and C were 28.8, 17.9, 37.1, and 16.3%, respectively) with A + T contents of 58.5%. All of the PCGs use a typical start codon (ATN) and a complete stop codon (TAA or TAG) as normal. The maximum-likelihood tree demonstrated that the Cypraeoidea was closer to the superfamily Tonnoidea, and further clarified the phylogenetic relationships of each superfamily in Littorinimorpha.

Project description:Tenebroides mauritanicus Linnaeus, 1758 (Coleoptera: Trogossitidae) is a storage pest that feeds mainly on soybean and corn. In this study, we sequenced the entire mitochondrial genome of Tenebroides mauritanicus (GenBank accession number: OM161967). The total length of the mitochondrial genome is 15,696 bp, GC content is 29.65%, and the contents of each base is 38.37% A, 18.35% C, 11.30% G and 31.98% T, respectively. The genome encodes 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs) and 2 ribosomal RNA genes (rRNAs). Phylogenetic analysis showed that Tenebroides mauritanicus is clustered with Byturus ochraceus. This study provides a piece of valuable genomic information for the population genetics, phylogeny, and molecular taxonomy of Tenebroides mauritanicus.

Characterization of complete mitochondrial genome of seven-eleven crab Carpilius maculatus (Linnaeus, 1758).

Project description:In this study, we first determined and characterised the complete mitochondrial genome of seven-eleven crab Carpilius maculatus from South China Sea. The C. maculatus mitogenome is 15,761 bp long, and consists of 22 tRNA genes, two rRNA genes, 13 protein-coding genes (PCGs), and one control region. The nucleotide composition of C. maculatus mitogenome is significantly biased (A, G, T, and C was 35.9%, 9.8%, 35.5%, and 18.9%, respectively) with A + T contents of 71.3%. Three PCGs used an unusual initiation codon, and four PCGs were terminated with an uncomplete or abnormal stop codon. Six microsatellites were identified in C. maculatus mitogenome sequences. Phylogenetic tree showed that C. maculatus formed monophyletic clade with other Heterotremata species.

The overall base composition\ of \the broad-nosed pipefish is 27.72% for A, 27.81% for C, 16.53% for G, 27.94% for T and show 87% identities to Syngnathus schlegeli (GenBank: AP012318). The complete mitogenome of the broad-nosed pipefish, provides essential and important DNA molecular data for further phylogeography and evolutionary analysis for seahorse family.
