The complete mitochondrial genome of Cephenemyia stimulator (Diptera: Oestridae).

ABSTRACT: Cephenemyia stimulator parasitizes roe deer (Capreolus capreolus) throughout its geographical distribution. The complete circular C. stimulator mitogenome was assembled, which is 16,407 bp in length, and encodes 13 protein-coding genes, 22 tRNA genes, and 2 rRNA genes. A phylogenetic tree was built with mitogenome sequences, including C. stimulator and 13 related Oestridae species, using Sarcophaga tuberosa as an outgroup.

The phylogenetic tree of O. ovis and 13 related Oestridae species, with Sarcophaga tuberosa as outgroup, was built.

Project description:Abstract Bradysia impatiens (Johannsen, 1912) is an important pest of various mushrooms and plant seedlings, while it also damages plants as vectors transmitting pathogens. The complete B. impatiens mitogenome is 16,479 bp in length, with an A + T content of 80.6%. The mitogenome had three overlapped regions; the longest overlap is 7 bp. Phylogenetic analysis reveals that B. impatiens is closely related to Bradysia amoena. The study provided a basis for future studies of species divergence in Bradysia.

Project description:The complete mitochondrial genome of Limnophyes minimus (Meigen 1818) was sequenced and annotated, and its general features and base composition were analyzed. The phylogenetic relationships of the families Chironomidae, Simuliidae, Sciaridae and Culicidae based on 25 metagenomes were reconstructed using maximum likelihood (ML) methods based on the concatenated nucleotide sequences, the phylogenetic analysis showed that L. minimus belongs family Chironomidae, which is consistent with the traditional morphological classification.

The complete mitochondrial genome of Sarcophaga brevicornis (Diptera: Sarcophagidae).

Project description:Sarcophaga brevicornis (Diptera: Sarcophagidae) was of utmost forensic importance due to their wide distribution, ubiquitous, and synanthropic nature. The complete mitochondrial genome (mitogenome) of S. brevicornis was first sequenced and assembled in this study. The length of circular mitogenome was 15,152?bp, which showed in a typical arthropod genome, including 13 protein-coding genes (13 PCGs), two ribosomal RNA (two rRNA) genes, 22 transfer RNA (22 tRNA) genes, and an AT-rich region. Its nucleotide composition was A 39.0%, C 12.7%, G 10.6%, and T 37.7%. Furthermore, phylogenetic relationships of S. brevicornis and the published sarcophagid species were evaluated based on 13 PCGs. The results indicated that S. brevicornis was clearly separated from the other sarcophagid species, but it was closed to the species of Sarcophaga similis. This study provided a significant database reference for genetic structure and phylogenetic analysis of Sarcophagidae.

The nucleotide composition biases toward A and T is 77.2% of the entirety. All PCGs start with ATN codons, except COI and ND1, and end with TAA or incomplete stop codon. Phylogenetic analyses based on 11 Diptera species supported the monophyly of superfamily Tephritoidea.

The circle DNA is composed of 13 protein-coding genes (13 PCGs), 2 ribosomal RNA (2 rRNAs), 22 transfer RNA (22 tRNAs) and a AT-rich region. It shows that arrangement of the genes is similar with the classical metazoan. The size of mitogenome is 15,236?bp containing A (40.0%), G (9.3%), T (36.6%), and C (14.1%). Moreover, phylogenetic analysis reveals that the branch of S. pterygota is clustered separately. This study enriches the mitogenome database of flesh flies and represents progress for analyzing of phylogenetic relationships.

It indicated that Opomyzoidea is assigned to be the sister of the clade of (Ephydroidea + (Tephritoidea?+?Lauxanioidea)).

All genes have the same location and coding strand as in other published species of Dolichopodidae. Nucleotide composition is biased toward A and T, which together made up 77.3% of the entire genome. Bayesian inference strongly supported the monophyly of Empidoidea, Empididae and Dolichopodidae, with the phylogenetic relationships within Empidoidea: (Dolichopodinae + Neurigoninae) + ((Empidinae + (Trichopezinae + Oreogetoninae)) + Ocydromiinae).

The full length of mitogenome was 14, 940 bp, consisting of 13 protein-coding genes (PCGs), two ribosomal RNA (rRNA), 22 transfer RNA (tRNA), and one AT-rich region. The nucleotide content of these flies was 40.0% A, 13.2% C, 9.1% G, and 37.6% T. This study illustrates that the arrangement of the genes was identical to classical metazoans. Besides, the phylogenetic analyses indicated that the branch of M. pascuorum was clustered separately from the common three Muscina spp in the tree. This genome provides an essential reference for understanding the phylogenetic relationships of Muscidae.
