Triblock-Diblock Composite Nanoassemblies with Sequentially Addressable Host-Guest Properties for Hydrophobics and Hydrophilics.

ABSTRACT: Noncovalent encapsulation of multiple guest molecules with distinct physiochemical properties, especially hydrophilic and hydrophobic guests, into a single polymeric nanoassembly is challenging, but would be profoundly beneficial to combination therapeutic delivery strategies. Here, we report a nanocomposite co-assembled by coating a layer of an amphiphilic block copolymer onto surface of a crosslinked inverse micelle driven by hydrophobic interactions. The hydrophobic coating layer and water-filled inverse micelles were utilized to encapsulate hydrophobic and hydrophilic cargos respectively. Rationally introducing the pH and redox responsive moieties into coating polymer and inverse micelles, allows sequentially addressing triggered release of the encapsulated cargos.

Construction of Chiral Nanoassemblies Based on Host-Guest Complexes and Their Responsive CD and CPL Properties: Chirality Transfer From 2,6-helic[6]arenes to a Stilbazolium Derivative.

Project description:A couple of water-soluble chiral 2,6-helic[6]arene derivatives P -H1 and M -H1 were synthesized, and they could form 1:1 stable complexes with 4-[(4'-N, N-diphenylamino)-styryl]-N-methylpyridinium iodide (G) in water. Compared with G, the host-guest complexes exhibited enhanced fluorescence, which might be attributed to the spatial confinement of G and restriction of aggregation-caused quenching (ACQ) effects. Based on the host-guest complexation, the first helic[6]arene-based chiral assemblies were then constructed, and they showed rectangular or hexagonal nanostructures by scanning electron microscopy (SEM) images. Interestingly, the assemblies showed clear mirror-image circular dichroism (CD) and circularly polarized luminescence (CPL) spectra in aqueous solution, revealing a consecutive chirality transfer from the chiral macrocyclic cavities of the hosts to G. Moreover, the supramolecular chirality of the assemblies could also show responsiveness to the pH values and temperatures of the system.

Host-Guest Carbon Dots for Enhanced Optical Properties and Beyond.

Project description:Carbon dots, generally small carbon nanoparticles with various forms of surface passivation, have achieved the performance level of semiconductor quantum dots in the green spectral region, but their absorption and fluorescence in red/near-IR are relatively weaker. Conceptually similar to endofullerenes, host-guest carbon dots were designed and prepared with red/near-IR dyes encapsulated as guest in the carbon nanoparticle core. Beyond the desired enhancement in optical properties, the host-guest configuration may significantly broaden the field of carbon dots.

Core-shell structured nanoassemblies based on ?-cyclodextrin containing block copolymer and poly(?-benzyl L-aspartate) via host-guest complexation.

Project description:Double hydrophilic copolymers (PEG-b-PCDs) with one PEG block and another block containing ?-cyclodextrin (?-CD) units were synthesized by macromolecular substitution reaction. Via a dialysis procedure, complex assemblies with a core-shell structure were prepared using PEG-b-PCDs in the presence of a hydrophobic homopolymer poly(?-benzyl L-aspartate) (PBLA). The hydrophobic PBLA resided preferably in the cores of assemblies, while the extending PEG chains acted as the outer shell. Host-guest interaction between ?-CD and hydrophobic benzyl group was found to mediate the formation of the assemblies, where PEG-b-PCD and PBLA served as the host and guest macromolecules, respectively. The particle size of the assemblies could be modulated by the composition of the host PEG-b-PCD copolymer. The molecular weight of the guest polymer also had a significant effect on the size of the assemblies. The assemblies prepared from the host and guest polymer pair were stable during a long-term storage. These assemblies could also be successfully reconstituted after freeze-drying. The assemblies may therefore be used as novel nanocarriers for the delivery of hydrophobic drugs.

Diblock, Triblock and Cyclic Amphiphilic Copolymers with CO2 Switchability: Effects of Topology.

Project description:The combination of topology and CO2 switchability could provide new options for amphiphilic copolymers. Cyclic molecules supply novel topologies, and CO2 switching provides stimulus responsiveness. Cyclic poly(2-(diethylamino)ethyl methacrylate)-b-poly(ethylene oxide) and their corresponding block copolymers were prepared from poly(ethylene oxide) and 2-(diethylamino)ethyl methacrylate via atom transfer radical polymerization and Keck allylation with a Hoveyda-Grubbs catalyst. Changes in conductivity, surface activity, and hydrodynamic size were examined to illustrate the switchability of the produced amphiphilic copolymers upon contact with CO2 in the presence of water. The reversible emulsification and switchable viscosity behaviors of the copolymers were also demonstrated.

Sequentially addressable dielectrophoretic array for high-throughput sorting of large-volume biological compartments.

Project description:Droplet microfluidics has become a powerful tool in precision medicine, green biotechnology, and cell therapy for single-cell analysis and selection by virtue of its ability to effectively confine cells. However, there remains a fundamental trade-off between droplet volume and sorting throughput, limiting the advantages of droplet microfluidics to small droplets (<10 pl) that are incompatible with long-term maintenance and growth of most cells. We present a sequentially addressable dielectrophoretic array (SADA) sorter to overcome this problem. The SADA sorter uses an on-chip array of electrodes activated and deactivated in a sequence synchronized to the speed and position of a passing target droplet to deliver an accumulated dielectrophoretic force and gently pull it in the direction of sorting in a high-speed flow. We use it to demonstrate large-droplet sorting with ~20-fold higher throughputs than conventional techniques and apply it to long-term single-cell analysis of Saccharomyces cerevisiae based on their growth rate.

Host-guest interaction and properties of cucurbit[8]uril with chloramphenicol.

Project description:The interaction between cucurbit[8]uril (Q[8]) and chloramphenicol (CPE) was investigated using single-crystal X-ray diffraction spectroscopy, isothermal titration calorimetry (ITC) and UV-vis, NMR and IR spectroscopy. The effects of Q[8] on the stability, in vitro release performance and antibacterial activity of CPE were also studied. The results showed that CPE and Q[8] formed a 1:1 inclusion complex (CPE@Q[8]) with an inclusion constant of 5.474 × 105 L/mol. The intervention of Q[8] did not affect the stability of CPE, but obviously reduced the release rate of CPE in artificial gastric and intestinal juice; Q[8] has a slow-release effect on CPE. The antibacterial results showed that the minimum inhibitory concentration (MIC) of CPE and CPE@Q[8] toward Escherichia coli (E. coli) was 1.5 × 10-3 and 1.0 × 10-3 mol/L, respectively, and toward Staphylococcus aureus (S. aureus), the MIC was 2.0 × 10-3 mol/L for both CPE and CPE@Q[8]. Therefore, Q[8] enhanced the inhibitory activity of CPE against E. coli.

Blending of diblock and triblock copolypeptide amphiphiles yields cell penetrating vesicles with low toxicity.

Project description:We prepared dual hydrophilic triblock copolypeptide vesicles that form both micron and nanometer scale vesicles in aqueous media. The incorporation of terminal homoarginine segments into methionine sulfoxide-based vesicles was found to significantly enhance their cellular uptake compared to a non-ionic control. We also demonstrated that diblock and triblock copolypeptides with similar hydrophobic domains were found to mix well and form vesicle populations with uniform compositions. Blending of amphiphiles in vesicle nanocarriers was found to impart these materials with many advantageous properties, including good cellular uptake while maintaining minimal toxicity, as well as biological responsiveness to promote vesicle disruption and release of encapsulated cargos.

Synthesis and 3D Printing of PEG-Poly(propylene fumarate) Diblock and Triblock Copolymer Hydrogels.

Project description:PEG-based hydrogels are used widely in exploratory tissue engineering applications but in general lack chemical and structural diversity. Additive manufacturing offers pathways to otherwise unattainable scaffold morphologies but has been applied sparingly to cross-linked hydrogels. Herein, mono methyl ether poly(ethylene glycol) (PEG) and PEG-diol were used to initiate the ring-opening copolymerization (ROCOP) of maleic anhydride and propylene oxide to yield well defined diblock and triblock copolymers of PEG-poly(propylene maleate) (PPM) and ultimately poly(propylene fumarate) (PPF) with different molecular mass PEG macroinitiators and block length ratios. Using continuous digital light processing (cDLP) hydrogels were photochemically printed from an aqueous solution which resulted in a 10-fold increase in elongation at break compared to traditional diethyl fumarate (DEF) based printing. Furthermore, PPF-PEG-PPF triblock hydrogels were also found to be biocompatible in vitro across a number of engineered MC3T3, NIH3T3, and primary Schwann cells.

Tuning MXene Properties through Cu Intercalation: Coupled Guest/Host Redox and Pseudocapacitance.

Project description:MXenes are 2D transition metal carbides, nitrides, and/or carbonitrides that can be intercalated with cations through chemical or electrochemical pathways. While the insertion of alkali and alkaline earth cations into Ti3C2Tx MXenes is well studied, understanding of the intercalation of redox-active transition metal ions into MXenes and its impact on their electronic and electrochemical properties is lacking. In this work, we investigate the intercalation of Cu ions into Ti3C2Tx MXene and its effect on its electronic and electrochemical properties. Using X-ray absorption spectroscopy (XAS) and ab initio molecular dynamics (AIMD), we observe an unusual phenomenon whereby Cu2+ ions undergo partial reduction upon intercalation from the solution into the MXene. Furthermore, using in situ XAS, we reveal changes in the oxidation states of intercalated Cu ions and Ti atoms during charging. We show that the pseudocapacitive response of Cu-MXene originates from the redox of both the Cu intercalant and Ti3C2Tx host. Despite highly reducing potentials, Cu ions inside the MXene show an excellent stability against full reduction upon charging. Our findings demonstrate how electronic coupling between Cu ions and Ti3C2Tx modifies electrochemical and electronic properties of the latter, providing the framework for the rational design and utilization of transition metal intercalants for tuning the properties of MXenes for various electrochemical systems.

Synthesis and Host-Guest Properties of Acyclic Pillar[<i>n</i>]naphthalenes.

Project description:Here we report a new class of synthetic receptors, acyclic pillar[n]naphthalene (n = 2-4, Dimer, Trimer, and Tetramer) oligomers, which are made up of 2,3-diethoxynaphthalene units linked by methylene bridges at the 1- and 4-positions. They can be synthesized through a one-step condensation of 2,3-diethoxynaphthalene monomer and paraformaldehyde in the presence of BF3•(Et)2O catalyst. The crystal structure of Tetramer has an interesting pseudo-cycle shaped structure in the solid state. Their complexation behaviors toward several organic ammonium cations (1 +-15 +) and electron-deficient neutral guests (16-17), were examined by means of 1H NMR spectroscopy. Tetramer shows good host-guest properties toward the ammonium guests, giving association constants (K a) in the magnitude of 102-104 M-1, which are comparable with those for some macrocyclic hosts.
