Antioxidant Capacity Assessment of Plant Extracts for Green Synthesis of Nanoparticles.

ABSTRACT: In this work, water extracts from different bio-based products of plant origin were studied to evaluate their antioxidant capacity and their potential to form metal nanoparticles from aqueous solutions. Two traditional tests, the Folin-Ciocalteu assay and the DPPH radical scavenging capacity method were compared with a more recent one, SNPAC, based on the formation of silver nanoparticles. The silver nanoparticle antioxidant capacity method (SNPAC) was optimized for its application in the characterization of the extracts selected in this work; kinetic studies and extract concentration were also evaluated. The extracts were obtained from leaves of oak, eucalyptus, green tea, white and common thyme, white cedar, mint, rosemary, bay, lemon, and the seaweed Sargassum muticum. The results demonstrate that any of these three methods can be used as a quick test to identify an extract to be employed for nanoparticle formation. Additionally, we studied the synthesis of Cu, Fe, Pb, Ni, and Ag nanoparticles using eucalyptus extracts demonstrating the efficiency of this plant extract to form metallic nanoparticles from aqueous metal salt solutions. Metal nanoparticles were characterized by transmission electron microscopy and dynamic light scattering techniques.

Project description:Cancer and diabetes represent significant challenges in the field of biomedicine, with major and global impacts on public health. Acacia nilotica, commonly called 'gum arabic tree,' is recognized for its unique biomedical properties. The current study aimed to investigate the pharmacological potential of A. nilotica-based zinc-oxide nanoparticles (ZnO-NPs) in comparison to the ethanol and methanol-based extracts against cancer, diabetes, and oxidative stress. Green synthesis of ZnO-NPs was performed using barks of Acacia nilotica. Different techniques for the characterization of ZnO-NPs, including UV-Visible spectroscopy, Scanning Electron Microscopy, Fourier Transmission Infrared (FT-IR) spectroscopy, and X-ray Diffraction (XRD), were utilized. The morphological analysis of ZnO-NPs revealed that the fine NPs have mean particle sizes of 15 ± 1.5 nm. For the solvent based-extraction, leaves and barks were utilized and dissolved into ethanol and methanol for further processing. The MTT assay revealed that the optimum concentration of ZnO-NPs to inhibit the proliferation of liver cancer cell line HepG2 was 100 μg/mL where 67.0 % inhibition was observed; and both ethanol- and methanol-based extracts showed optimum inhibition at 100 μg/mL. The DPPH assay further demonstrated that 250 μg/mL of ZnO-NPs and 1000 μg/mL of both ethanol- and methanol-based extracts, as the optimum concentration for antioxidant activity (with 73.1 %, 68.9 % and 68.2 % inhibition respectively). The α-Glucosidase inhibition assay revealed that 250 μg/mL of ZnO-NPs and 10 μg/mL of both ethanol- and methanol-based extracts as the optimum concentration for antidiabetic activity (with 95 %, 93.7 % and 93.4 % inhibition respectively). The study provided interesting insights into the efficacy and reliability of ZnO-NPs for potential pharmacological application. Further research should be focused on examining specific pathways and the safety of ZnO-NPs in comparison to solvent-based extracts.

Project description:The research was focused on the synthesis of silver nanoparticles (AgNPs) using extracts from the "hairy" root cultures of Artemisia tilesii Ledeb. and Artemisia annua L. The effect of operational parameters such as type of solvent, temperature of extraction, flavonoids concentration, and reducing power of the wormwood "hairy" root extracts on the particle size and yield of the resultant nanoparticles is reported for the first time. From the studied solvents, a water-ethanol mixture with a concentration of 70 vol% was found to be the best for the extraction of flavonoids from all "hairy" root cultures. The total flavonoid contents in A. annua and A. tilesii "hairy" root extracts were up to 80.0 ± 0.9 and 108 ± 4.4 mg RuE per g DW, respectively. Identification of flavonoids was confirmed by UPLC-ESI-UHR-Qq-TOF-MS analysis. Luteolin-7-β-d-glucopyranosid, isorhamnetin 3-O-glucoside, baicalein-7-O-glucuronide, apigenin-7-O-glucoside, quercetin, sitosterol, caffeoylquinic, galic, chlorogenic and caffeic acids were founded in the extracts. These extracts demonstrated the high reducing activities. Spherical, oval and triangular nanoparticles with effective sizes of 5-100 nm were observed. The TEM data revealed great differences in the shapes of NPs, obtained from the extracts from different root clones. The clustered and irregular NPs were found in the case of using ethanol extracts, mostly aggregated and having the size of 10-50 nm. The sizes of AgNPs decreased to 10-30 nm in the case of using aqueous extracts obtained at 80 °C. Biosynthesized AgNPs showed surface plasmon resonance in the range of 400-450 nm. The antimicrobial activity of the as-produced AgNPs was studied by disc diffusion method on Gram-positive (Staphylococcus aureus ATCC 25923 (F-49)), Gram-negative (Pseudomonas aeruginosa ATCC 27853 (F-51), Escherichia coli ATCC 25922 (F-50)) and Candida albicans ATCC 88-653 strains. It was found that the nanoparticles in some cases possessed the greater ability to inhibit microorganism growth compared to 1 mM AgNO3 solution. The colloidal solutions of the obtained AgNPs were stable in the dark for 12 months at room temperature. Thus, the A. annua and A. tilesii "hairy" root extracts can be used for obtaining of bioactive AgNPs.

Project description:The current study investigates the phytochemical composition of coffee plant organs and their corresponding antioxidant capacities compared to green and roasted coffee beans. HPLC analysis indicated that the investigated compounds were present in all organs except mangiferin, which was absent in roots, stems and seeds, and caffeine, which was absent in stems and roots. Total phytochemicals were highest in the green beans (GB) at 9.70 mg g-1 dry weight (DW), while roasting caused a 66% decline in the roasted beans (RB). This decline resulted more from 5-CQA and sucrose decomposition by 68% and 97%, respectively, while caffeine and trigonelline were not significantly thermally affected. Roasting increased the total phenolic content (TPC) by 20.8% which was associated with an increase of 68.8%, 47.5% and 13.4% in the antioxidant capacity (TEAC) determined by 2,2-diphenyl-1-picryl hydrazyl radical (DPPH), 2,2-azino bis (3-ethyl benzothiazoline-6-sulphonic acid) radical (ABTS) and Ferric ion reducing antioxidant power (FRAP) assays, respectively. Amongst the leaves, the youngest (L1) contained the highest content at 8.23 mg g-1 DW, which gradually reduced with leaf age to 5.57 mg g-1 DW in the oldest (L6). Leaves also contained the highest TPC (over 60 mg g-1 GAE) and exhibited high TEAC, the latter being highest in L1 at 328.0, 345.7 and 1097.4, and least in L6 at 304.6, 294.5 and 755.1 µmol Trolox g-1 sample for the respective assays. Phytochemical accumulation, TPC and TEAC were least in woody stem (WS) at 1.42 mg g-1 DW; 8.7 mg g-1 GAE; 21.9, 24.9 and 110.0 µmol Trolox g-1 sample; while herbaceous stem (HS) contained up to 4.37 mg g-1 DW; 27.8 mg g-1 GAE; 110.9, 124.8 and 469.7 µmol Trolox g-1 sample, respectively. Roots contained up to 1.85 mg g-1 DW, 15.8 mg-1 GAE and TEAC of 36.8, 41.5 and 156.7 µmol Trolox g-1 sample. Amongst the organs, therefore, coffee leaves possessed higher values than roasted beans on the basis of phytochemicals, TPC and TEAC. Leaves also contain carotenoids and chlorophylls pigments with potent health benefits. With appropriate processing methods, a beverage prepared from leaves (coffee leaf tea) could be a rich source of phytochemicals and antioxidants with therapeutic and pharmacological values for human health.

Project description:Titanium dioxide (TiO2) nanoparticles were synthesized via a novel eco-friendly green chemistry approach using marine natural extracts of two red algae (Bostrychia tenella and Laurencia obtusa), a green alga (Halimeda tuna), and a brown alga (Sargassum filipendula) along with a marine sponge sample identified as Carteriospongia foliascens. X-ray diffraction (XRD), scanning electron microscope (SEM), UV-Vis, X-ray spectroscopy (EDS), X-ray photoelectron spectroscopy (XPS), and Fourier-transform infrared spectroscopy (FTIR) were employed to characterize the crystal structure, surface morphology, and optical properties of the synthesized nanoparticles. Each of the as-synthesized marine extract based TiO2 nanoparticles was individually incorporated as an antifouling agent to form a newly fabricated marine paint formulation. The newly prepared formulations were applied on unprimed steel panels. A comparative study with a commercial antifouling paint (Sipes Transocean Coatings Optima) was carried out. After 108 days of the coated steel panels' immersion in the Eastern Harbour seawater of Alexandria-Egypt, the prepared paints using B. tenella and C. foliascens extracts demonstrated an excellent antifouling performance toward fouling organisms by inhibiting their settlement and controlling their adhesion onto the immersed panels. In contrast, heavy fouling with barnacles was observed on the surface of the coated panel with the commercial paint. The physicochemical parameters of the seawater surrounding the immersed coated panels were estimated to investigate the influence of the fabricated paint formulations. Interestingly, no effects of the immersed coated panels on the physicochemical characteristics of the surrounding seawater were observed. Based on the obtained results and a comparison with commercially available antifouling products, the marine extract based TiO2 nanoparticle preparations of B. tenella and C. foliascens are promising candidates for eco-friendly antifouling agents.
