ABSTRACT: Bacteria of the genus Bacillus have been investigated due to the ability that many species have of accumulating polyhydroxyalkanoates (PHA) via a wide variety of raw materials as their carbon source. Herein, we report the draft whole-genome sequence of the putative PHA-accumulating strain Bacillus paramycoides LB_RP2, isolated from an Amazonian river.

Production, Characterization and Application of a Novel Chitosanase from Marine Bacterium <i>Bacillus paramycoides</i> BP-N07.

Project description:Chitooligosaccharides (COS), a high-value chitosan derivative, have many applications in food, pharmaceuticals, cosmetics and agriculture owing to their unique biological activities. Chitosanase, which catalyzes the hydrolysis of chitosan, can cleave β-1,4 linkages to produce COS. In this study, a chitosanase-producing Bacillus paramycoides BP-N07 was isolated from marine mud samples. The chitosanase enzyme (BpCSN) activity was 2648.66 ± 20.45 U/mL at 52 h and was able to effectively degrade chitosan. The molecular weight of purified BpCSN was approximately 37 kDa. The yield and enzyme activity of BpCSN were 0.41 mg/mL and 8133.17 ± 47.83 U/mg, respectively. The optimum temperature and pH of BpCSN were 50 °C and 6.0, respectively. The results of the high-performance liquid chromatography (HPLC) and thin-layer chromatography (TLC) of chitosan treated with BpCSN for 3 h showed that it is an endo-chitosanase, and the main degradation products were chitobiose, chitotriose and chitotetraose. BpCSN was used for the preparation of oligosaccharides: 1.0 mg enzyme converted 10.0 g chitosan with 2% acetic acid into oligosaccharides in 3 h at 50 °C. In summary, this paper reports that BpCSN has wide adaptability to temperature and pH and high activity for hydrolyzing chitosan substrates. Thus, BpCSN is a chitosan decomposer that can be used for producing chitooligosaccharides industrially.

Thauera aminoaromatica MZ1T Identified as a Polyhydroxyalkanoate-Producing Bacterium within a Mixed Microbial Consortium.

Project description:Polyhydroxyalkanoates (PHAs) form a highly promising class of bioplastics for the transition from fossil fuel-based plastics to bio-renewable and biodegradable plastics. Mixed microbial consortia (MMC) are known to be able to produce PHAs from organic waste streams. Knowledge of key-microbes and their characteristics in PHA-producing consortia is necessary for further process optimization and direction towards synthesis of specific types of PHAs. In this study, a PHA-producing mixed microbial consortium (MMC) from an industrial pilot plant was characterized and further enriched on acetate in a laboratory-scale selector with a working volume of 5 L, and 16S-rDNA microbiological population analysis of both the industrial pilot plant and the 5 L selector revealed that the most dominant species within the population is Thauera aminoaromatica MZ1T, a Gram-negative beta-proteobacterium belonging to the order of the Rhodocyclales. The relative abundance of this Thauera species increased from 24 to 40% after two months of enrichment in the selector-system, indicating a competitive advantage, possibly due to the storage of a reserve material such as PHA. First experiments with T. aminoaromatica MZ1T showed multiple intracellular granules when grown in pure culture on a growth medium with a C:N ratio of 10:1 and acetate as a carbon source. Nuclear magnetic resonance (NMR) analyses upon extraction of PHA from the pure culture confirmed polyhydroxybutyrate production by T. aminoaromatica MZ1T.

Genome analysis of biosurfactant producing bacterium, Bacillus tequilensis.

Project description:Bioremediation is crucial for recuperating polluted water and soil. By expanding the surface area of substrates, biosurfactants play a vital role in bioremediation. Biosurfactant-producing microbes release certain biosurfactant compounds, which are promoted for oil spill remediation. In the present investigation, a biosurfactant-producing bacterium Bacillus tequilensis was isolated from Chilika Lake, Odisha, India (latitude and longitude: 19.8450 N 85.4788 E). Whole-Genome Sequencing (WGS) of Bacillus tequilensis was carried out using Illumina NextSeq 500. The size of the whole genome of Bacillus tequilensis was 4.47 MB consisting of 4,478,749 base pairs forming a circular chromosome with 528 scaffolds, 4492 protein-encoding genes (ORFs), 81 tRNA genes, and 114 ribosomal RNA transcription units. The total raw reads were 4209415, and the processed reads were 4058238 with 4492 genes. The whole genome obtained from the present investigation was used for genome annotation, variant calling, variant annotation, and comparative genome analysis with other existing Bacillus species. In this study, a pathway was constructed which describes the biosurfactant metabolism of Bacillus tequilensis. The study identified that genes such as SrfAD, SrfAC, SrfAA and SrfAB are involved in biosurfactant synthesis. The sequence of the genes SrfAD, SrfAC, SrfAA, SrfAB was deposited in GenBank database with accession MUG02427.1, MUG02428.1, MUG02429.1, MUG03515.1 respectively. The whole genome sequence was submitted to GenBank with an accession RMVO00000000 and the raw fastq reads were submitted to SRA, NCBI repository with an accession: SRX5023292.

Project description:Cupriavidus necator KK10 has been investigated in azaarene and diesel fuel biodegradation studies and is capable of polyhydroxyalkanoate (PHA) production. Its complete genome sequence revealed two closed circular sequences, the chromosome (6.68 Mb) and plasmid (1.67 Mb). The KK10 genome carries functional genes potentially responsible for azaarene biodegradation and polyhydroxyalkanoate biosynthesis.

Draft Genome Sequence of the Tacrolimus-Producing Bacterium Streptomyces tsukubaensis F601.

Project description:Streptomyces tsukubaensis strain F601 was found to be a producer of the immunosuppressive drug tacrolimus. The draft genome sequence of this strain was approximately 8.52 Mbp. Genes involved in the biosynthesis of tacrolimus were identified in the genome. This draft genome sequence will provide insights into the genetic basis of tacrolimus biosynthesis and regulation.

Draft genome sequence of Paenibacillus strain LK1, a phytohormone producing bacterium.

Project description:A high-quality draft genome sequence of a multi-antibiotic resistant strain, Paenibacillus strain LK1 isolated from the rhizosphere of Auricularia auricular is described. Strain LK1 produced several types of phytohormones, which included Indole-3-acetic acid (IAA) and Zeatin. Phylogenetic analysis of the 16S rRNA gene of strain LK1 showed a close relationship to P. taichungensis and P. pabuli (99% gene sequence similarity). The genome size of strain LK1 was estimated to be 7.16 Mb that consisted of 82 contigs contained in 49 scaffolds. The G + C content of the genome was 45.63% and it encoded 6499 genes, 115 pseudo genes, 8 rRNAs, 33 tRNAs, and 4 ncRNAs, based on the Prokaryotic Genome Annotation Pipeline (PGAP). Further analysis of the genome of strain LK1 identified 16 genes encoding monooxygenase and six genes encoding cytochrome P450, which may be potentially involved in zeatin biosynthesis. The draft whole genome sequence and annotation of strain LK1 has been deposited at DDBJ/EMBL/GenBank under the accession number (PEII00000000).
