The complete mitochondrial genome of Periacma orthiodes Meyrick, 1894 (Lepidoptera: Autostichidae).

ABSTRACT: The complete mitogenome of Periacma orthiodes Meyrick, 1894 was determined in this study. It was 15,306 bps long and strongly AT biased. It consisted of 13 PCGs, 22 tRNAs, 2 rRNAs and 1 non-coding control region (371 bps). Most PCGs used the typical ATN start codon, except for cox1. Four genes (cox1, cox2, nad4 and nad5) used truncated stop codons (a single T or TA) rather than the commonly used TAA or TAG. All tRNAs, excluding TrnS1, folded into the iconic cloverleaf structure. ML phylogenetic tree built on 13 PCGs from P. orthiodes and another 28 species in Gelechioidea demonstrated that the genus Periacma was a member of the family Autostichidae, which was consistent with the newest phylogenetic study.

Project description:The mitogenome of Ripeacma umbellata Wang, 2009 was reported in this study. It was 15,486 bps long and strongly AT biased, consisting of 13 protein-coding genes (PCGs), 22 transfer RNAs (tRNAs), 2 ribosomal RNAs (rRNAs), and 1 non-coding control region (351 bp). Most PCGs used the conventional ATN start codon, except for cox1 and cox2. Four genes used single T residue as stop codon rather than the routinely used TAA or TAG. All tRNAs, except for TrnS1, could fold into the cloverleaf secondary structure. Bayesian inference phylogenetic tree built on 13 PCGs from R. umbellata and another 21 species in Gelechioidea demonstrated that genus Ripeacma was a member in Autostichidae, which was consistent with the latest phylogenetic study.

Project description:The mitogenome of Meleonoma mirabilis was determined in this study. It was 15,268 bps long and strongly AT biased. It harbored 13 PCGs, 22 tRNAs, 2 rRNAs, and 1 non-coding control region (334 bps). BI phylogenetic analysis based on 13 concatenated PCGs from 24 moth species indicated that M. mirabilis was clustered in the family Autostichidae, which was consistent with the latest phylogenetic study.

Project description:The brown pansy, Junonia stygia (Aurivillius, 1894) (Lepidoptera: Nymphalidae), is a widespread West African forest butterfly. Genome skimming by Illumina sequencing allowed assembly of a complete 15,233 bp circular mitogenome from J. stygia consisting of 79.5% AT nucleotides. Mitochondrial gene order and composition is identical to other butterfly mitogenomes. Junonia stygia COX1 features an atypical CGA start codon, while ATP6, COX1, COX2, ND4, and ND4L exhibit incomplete stop codons. Phylogenetic reconstruction supports a monophyletic Subfamily Nymphalinae, Tribe Junoniini, and genus Junonia. The phylogenetic tree places Junonia iphita and J. stygia as basal mitogenome lineages sister to the remaining Junonia sequences.

Project description:Sixteen new species of the genus Meleonoma Meyrick, 1914 from Hainan Island, China are described: M. apicicurvata Wang, sp. nov., M. apicirectangula Wang, sp. nov., M. bicuspidata Wang, sp. nov., M. bidentata Wang, sp. nov., M. conica Wang, sp. nov., M. hainanensis Wang, sp. nov., M. latiunca Wang, sp. nov., M. linearis Wang, sp. nov., M. magnidentata Wang, sp. nov., M. ornithorrhyncha Wang, sp. nov., M. parilis Wang, sp. nov., M. pectinalis Wang, sp. nov., M. puncticulata Wang, sp. nov., M. quadritaeniata Wang, sp. nov., M. robustispina Wang, sp. nov. and M. rostellata Wang, sp. nov. Images of adult dorsal habitus and genitalia of the new species are provided. A map showing the collecting localities and photos of the habitat where the specimens were collected are provided, along with two maps showing the distribution of each species.

The complete mitochondrial genome of Rondotia menciana (Lepidoptera: Bombycidae).

Project description:The mulberry white caterpillar, Rondotia menciana Moore (Lepidoptera: Bombycidae) is a species with closest relationship with Bombyx mori and Bombyx mandarina, and the genetic information of R. menciana is important for understanding the diversity of the Bombycidae. In this study, the mitochondrial genome (mitogenome) of R. menciana was amplified by polymerase chain reaction and sequenced. The mitogenome of R. menciana was determined to be 15,301 bp, including 13 protein-coding genes (PCGs), 2 ribosomal RNA genes, 22 transfer RNA genes, and an AT-rich region. The A+T content (78.87%) was lower than that observed for other Bombycidae insects. All PCGs were initiated by ATN codons and terminated with the canonical stop codons, except for coxII, which was terminated by a single T. All the tRNA genes displayed a typical clover-leaf structure of mitochondrial tRNA. The length of AT-rich region (360 bp) of R. menciana mitogenome is shorter than that of other Bombycidae species. Phylogenetic analysis showed that the R. menciana was clustered on one branch with B. mori and B. mandarina from Bombycidae.

The complete mitochondrial genome of Leucoptera malifoliella Costa (Lepidoptera: Lyonetiidae).

Project description:The mitochondrial genome (mitogenome) of Leucoptera malifoliella (=L. scitella) (Lepidoptera: Lyonetiidae) was sequenced. The size was 15,646 bp with gene content and order the same as those of other lepidopterans. The nucleotide composition of L. malifoliella mitogenome is highly A+T biased (82.57%), ranked just below Coreana raphaelis (82.66%) (Lepidoptera: Lycaenidae). All protein-coding genes (PCGs) start with the typical ATN codon except for the cox1 gene, which uses CGA as the initiation codon. Nine PCGs have the common stop codon TAA, four PCGs have the common stop codon T as incomplete stop codons, and nad4l and nad6 have TAG as the stop codon. Cloverleaf secondary structures were inferred for 22 tRNA genes, but trnS1(AGN) was found to lack the DHU stem. The secondary structure of rrnL and rrnS is generally similar to other lepidopterans but with some minor differences. The A+T-rich region includes the motif ATAGA, but the poly (T) stretch is replaced by a stem-loop structure, which may have a similar function to the poly (T) stretch. Finally, there are three long repeat (154 bp) sequences followed by one short repeat (56 bp) with four (TA)(n) intervals, and a 10-bp poly-A is present upstream of trnM. Phylogenetic analysis shows that the position of Yponomeutoidea, as represented by L. malifoliella, is the same as traditional classifications. Yponomeutoidea is the sister to the other lepidopteran superfamilies covered in the present study.

Project description:We describe the complete mitochondrial genome of Phiaris dolosana. It is 15,562 bp in length, and contains 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), 2 ribosomal RNA genes (rRNAs), and a 612?bp D-Loop. All PCGs start with ATN codon except for COI gene, which uses CGA as the initiation codon. Nine of 13 PCGs use a typical stop codon of TAA, the rest use incomplete stop codon of T or TA. Phylogenetic analysis of P. dolosana with other 17 leaf rollers is conducted with neighbor-joining method, the result is consistent with the conventional classification.

Complete mitochondrial genome of Cnidocampa flavescens (Lepidoptera: Limacodidae).

Project description:Cnidocampa flavescens, lives in Nepal, Bhutan, China, Far East of Russia, Korea, and Japan, belongs to the Lepidoptera family Limacodidae. In this study, we describe the genomic features of the mitogenome sequences of the insects. The mitogenome of C. flavescens is 15,406?bp long consisting a typical set of genes (13 protein-coding genes, 22 tRNA genes, and 2 rRNA genes) and one major 415?bp non-coding A?+?T-rich region. All PCGs of C. flavescens start with ATN codons and end with TAA codons. The gene arrangement of C. flavescens mitogenome is same to Monema flavescens while the intergenic spacers and overlaps are different. The 415?bp A?+?T-rich region contains a conserved ATAGA motif followed a poly-T stretch.

We described the complete mitogenome of M. intermedia, which is typical circular duplex molecules and 15,386 bp in length, containing the standard metazoan set of 13 protein-coding genes, 22 transfer RNA genes, 2 ribosomal RNA genes, and an A + T-rich region with macro-repeat sequences. All the inferred tRNA secondary structures show the common cloverleaf pattern, with the exception of trnS1(AGN) which lacks the DHU arm. The M. intermedia mitochondrial genome has the same gene order with other lepidopterans.

Complete mitochondrial genome of Casmara patrona (Lepidoptera: Oecophoridae).

Project description:The complete mitochondrial genome of Casmara patrona (Lepidoptera: Oecophoridae) was sequenced for a future phylogenetic study of Lepidoptera. The circle genome of the moth is 15,393 bp in length with a pronounced base bias of A + T (79.3%), containing 13 protein-coding genes, 22 transfer RNAs, two ribosomal RNAs, and a putative control region. The coxI gene had a CGA start codon as most lepidopteran species, other PCGs use the typical ATN codons. All PCGs end with the complete stop codon TAA. Phylogenetic analyses showed that the monophyly of Oecophoridae was highly supported based on the concatenated sequence of the 13 PCGs. In addition, Oecophoridae and Xyloryctidae had the closest relationship.
