Photochromic Polyamide 6 Based on Spiropyran Synthesized via Hydrolyzed Ring-Opening Polymerization.

ABSTRACT: We report photochromic polyamide 6 (PA6) which was synthesized by hydrolyzed ring-opening polymerization of ε-caprolactam with spiropyran (SP) embedded in the polymer chains. It indicated that crystallinity degree of the resulting copolymers was decreased since only PA6 segments can crystallize with increasing content of SP modifier. Meanwhile, toughness of photochromic PA6 was decreased. The photochromic property analysis indicated that the sample with more flexibility and more content of SP was more sensitive to UV light at the beginning of irradiation than other samples and its color after being irradiated for 1 min tended to reddish. Investigation revealed that the UV-vis absorbance of SP-PA6-3 had negligible decay after 10 cycles, which indicated SP-modified PA6 possessed excellent photoresponse reversibility and fatigue resistance.

Characterization of Aliphatic Polyesters Synthesized via Enzymatic Ring-Opening Polymerization in Ionic Liquids.

Project description:To evaluate the effects of ionic liquids (ILs) on the microstructural features of aliphatic polyesters for biomedical applications, a series of copolymers were synthesized by lipase ring opening polymerization of rac-lactide (rac-LA) and ε-caprolactone (CL). The chemical structures of resulting polymers were characterized by ¹H- and 13C-NMR and the average molecular weight (Mn) and dispersity index were characterized by gel permeation chromatography. The structure of the copolymers confirms the presence of linear polymer chains with end-functional hydroxyl groups allowing covalent coupling of the therapeutic agents. Chain microstructure of copolymers indicates the presence of both random and block copolymers depending on the synthesis conditions. Moreover, it was found that CL is the most active co-monomer during copolymerization which enhances the polymerizability of rac-LA and allows to obtain higher Mn of the copolymers. The results demonstrate that ILs could be promising solvents in synthesis of aliphatic esters for biomedical applications.

Ionizable Amino-Polyesters Synthesized via Ring Opening Polymerization of Tertiary Amino-Alcohols for Tissue Selective mRNA Delivery.

Project description:The utility of messenger RNA (mRNA) as a therapy is gaining a broad interest due to its potential for addressing a wide range of diseases, while effective delivery of mRNA molecules to various tissues still poses a challenge. This study reports on the design and characterization of new ionizable amino-polyesters (APEs), synthesized via ring opening polymerization (ROP) of lactones with tertiary amino-alcohols that enable tissue and cell type selective delivery of mRNA. With a diverse library of APEs formulated into lipid nanoparticles (LNP), structure-activity parameters crucial for efficient transfection are established and APE-LNPs are identified that can preferentially home to and elicit effective mRNA expression with low in vivo toxicity in lung endothelium, liver hepatocytes, and splenic antigen presenting cells, including APE-LNP demonstrating nearly tenfold more potent systemic mRNA delivery to the lungs than vivo-jetPEI. Adopting tertiary amino-alcohols to initiate ROP of lactones allows to control polymer molecular weight and obtain amino-polyesters with narrow molecular weight distribution, exhibiting batch-to-batch consistency. All of which highlight the potential for clinical translation of APEs for systemic mRNA delivery and demonstrate the importance of employing controlled polymerization in the design of new polymeric nanomaterials to improve in vivo nucleic acid delivery.

Electrochemical Ring-Opening and -Closing of a Spiropyran.

Project description:The bistability of molecular switches is an essential characteristic in their use as functional components in molecular-based devices and machines. For photoswitches, light-driven switching between two stable states proceeds via short-lived changes of the bond order in electronically excited states. Here, bistable switching of a ditertbutyl-substituted spiropyran photoswitch is instead demonstrated by oxidation and subsequent reduction in an overall four-state cycle. The spiropyran structure chosen has reduced sensitivity to the effect of secondary electrochemical processes such as H+ production and provides transient access to a decreased thermal Z-E isomerization barrier in the one electron oxidized state, akin to that achieved in the corresponding photochemical path. Thus, we show that the energy needed for switching spiropyrans to the merocyanine form on demand, typically delivered by a photon, can instead be provided electrochemically. This opens up further opportunities for the utilization of spiropyrans in electrically controlled applications and devices.

Ring-Opening Polymerization of 1,3-Benzoxazines via Borane Catalyst.

Project description:Tris(pentafluorophenyl)borane was used as Lewis acid catalyst to lower the ring opening polymerization temperature (ROP) of 1,3-benzoxazines. Dynamic scanning calorimeter studies revealed that on-set ROP temperatures were decreased as much as 98 °C for model benzoxazine compounds. Catalytic polymerization was traced by both FTIR and ¹H NMR, and revealed that tris(pentafluorophenyl)borane acted rapidly and fast curing achieved. Moreover, thermal properties of resulting polybenzoxazines were investigated by thermogravimetric analysis (TGA) and found out that the catalyst has high impact on char yield and even 3 mol % catalyst augmented char yields up to 13%.

Two-Dimensional Controlled Syntheses of Polypeptide Molecular Brushes via <i>N</i>-Carboxyanhydride Ring-Opening Polymerization and Ring-Opening Metathesis Polymerization.

Project description:Well-defined molecular brushes bearing polypeptides as side chains were prepared by a "grafting through" synthetic strategy with two-dimensional control over the brush molecular architectures. By integrating N-carboxyanhydride ring-opening polymerizations (NCA ROPs) and ring-opening metathesis polymerizations (ROMPs), desirable segment lengths of polypeptide side chains and polynorbornene brush backbones were independently constructed in controlled manners. The N2 flow accelerated NCA ROP was utilized to prepare polypeptide macromonomers with different lengths initiated from a norbornene-based primary amine, and those macromonomers were then polymerized via ROMP. It was found that a mixture of dichloromethane and an ionic liquid were required as the solvent system to allow for construction of molecular brush polymers having densely-grafted peptide chains emanating from a polynorbornene backbone, poly(norbornene-graft-poly(β-benzyl-l-aspartate)) (P(NB-g-PBLA)). Highly efficient postpolymerization modification was achieved by aminolysis of PBLA side chains for facile installment of functional moieties onto the molecular brushes.

Rapid, Highly Sustainable Ring-Opening Polymerization via Resonant Acoustic Mixing.

Project description:Reported herein is the first combination of resonant acoustic mixing (RAM) and controlled ring-opening polymerization (ROP) to deliver fully sustainable, end-functionalized, biodegradable polymers via a manufacturing route with a much-reduced environmental impact. This includes the successful use of agriculturally sourced functionalized initiators (terpene alcohols) in ROP synthesis of cyclic esters to generate an array of novel, biodegradable polyesters applicable to numerous biomedical applications, such as drug delivery. Furthermore, RAM was utilized as a novel mixing technique, resulting in a synthetic process that was conducted: (a) with minimal use of toxic, flammable, costly, and environmentally detrimental solvents, (b) in the absence of organometallic catalysts, and (c) with significantly shorter ROP reaction times and temperatures. Consequent comparison with conventional magnetic stirring or sonication-based mixing methods showed that RAM allowed the more facile, kilogram-scale synthesis of polyesters via reactions conducted at room temperature rather than 150 °C and without the need for a metal catalyst. As a proof of concept, the polymers were used to encapsulate bovine serum albumin as a model protein, and its release was measured using an automated, high-throughput protein assay. This study demonstrated that the headgroup chemistry appears to affect the release rate of protein from the polymers.

Programming Photodegradability into Vinylic Polymers via Radical Ring-Opening Polymerization.

Project description:Incorporation of photolabile moieties into the polymer backbone holds promise to remotely-control polymer degradation. However, suitable synthetic avenues are limited, especially for radical polymerizations. Here we report a strategy to program photodegradability into vinylic polymers by exploiting the wavelength selectivity of photocycloadditions for radical ring-opening polymerization (rROP). Irradiation of coumarin terminated allylic sulfides with UVA light initiated intramolecular [2+2] photocycloaddition producing cyclic macromonomers. Subsequent RAFT-mediated rROP with methyl acrylate yielded copolymers that inherited the photoreactivity of the cyclic parent monomer. Irradiation with UVB initiated efficient photocycloreversion of the coumarin dimers, causing polymer degradation within minutes under UVB light or days under sunlight exposure. Our synthetic strategy may pave the way to insert photolabile linkages into vinylic polymers, tuning degradation for specific wavelengths.

Water-soluble polyphosphonate-based bottlebrush copolymers <i>via</i> aqueous ring-opening metathesis polymerization.

Project description:Ring-opening metathesis polymerization (ROMP) is a versatile method for synthesizing complex macromolecules from various functional monomers. In this work, we report the synthesis of water-soluble and degradable bottlebrush polymers, based on polyphosphoesters (PPEs) via ROMP. First, PPE-macromonomers were synthesized via organocatalytic anionic ring-opening polymerization of 2-ethyl-2-oxo-1,3,2-dioxaphospholane using N-(hydroxyethyl)-cis-5-norbornene-exo-2,3-dicarboximide as the initiator and 1,8-diazabicyclo[5.4.0]undec-7-ene (DBU) as the catalyst. The resulting norbornene-based macromonomers had degrees of polymerization (DPn) ranging from 25 to 243 and narrow molar mass dispersity (Đ ≤ 1.10). Subsequently, these macromonomers were used in ROMP with the Grubbs 3rd-generation bispyridyl complex (Ru-G3) to produce a library of well-defined bottlebrush polymers. The ROMP was carried out either in dioxane or in aqueous conditions, resulting in well-defined and water-soluble bottlebrush PPEs. Furthermore, a two-step protocol was employed to synthesize double hydrophilic diblock bottlebrush copolymers via ROMP in water at neutral pH-values. This general protocol enabled the direct combination of PPEs with ROMP to synthesize well-defined bottlebrush polymers and block copolymers in water. Degradation of the PPE side chains was proven resulting in low molar mass degradation products only. The biocompatible and biodegradable nature of PPEs makes this pathway promising for designing novel biomedical drug carriers or viscosity modifiers, as well as many other potential applications.

Carbodiimide Ring-Opening Metathesis Polymerization.

Project description:Controlled incorporation of nitrogen into macromolecular skeletons is a long-standing challenge whose resolution would enable the preparation of soft materials with the scalability of man-made plastics and functionality of Nature's proteins. Nylons and polyurethanes notwithstanding, nitrogen-rich polymer backbones remain scarce, and their synthesis typically lacks precision. Here we report a strategy that begins to address this limitation founded on a mechanistic discovery: ring-opening metathesis polymerization (ROMP) of carbodiimides followed by carbodiimide derivatization. An iridium guanidinate complex was found to initiate and catalyze ROMP of N-aryl and N-alkyl cyclic carbodiimides. Nucleophilic addition to the resulting polycarbodiimides enabled the preparation of polyureas, polythioureas, and polyguanidinates with varied architectures. This work advances the foundations of metathesis chemistry and opens the door to systematic investigations of structure-folding-property relationships in nitrogen-rich macromolecules.

Reversible photodynamic chloride-selective sensor based on photochromic spiropyran.

Project description:We report here for the first time on a reversible photodynamic bulk optode sensor based on the photoswitching of a spiropyran derivative (Sp). The photoswitching of Sp induces a large basicity increase in the polymeric phase, which triggers the extraction of Cl(-) and H(+). Cl(-) is stabilized by a lipophilic chloride-selective ionophore inside the membrane, while H(+) binds with the open form of Sp and induces a spectral change, hence providing the sensor signal. The system was studied with spectroscopic and electrochemical methods.
