The complete mitochondrial genome of the white seabream Diplodus sargus (Perciformes: Sparidae) from the Tyrrhenian sea.

ABSTRACT: The white seabream Diplodus sargus (Linnaeus, 1758) is a species of interest for commercial fisheries throughout its range of distribution and it is also reared using aquaculture techniques. Herein, we present the first complete sequence and annotation of the mitochondrial genome of this species. The D. sargus mitogenome is 16,515 base pairs in length and contains 13 protein-coding genes, 2 rRNA, 22 tRNA, and 2 non-coding regions (D-loop and L-origin). The overall nucleotide composition is: 27.3% A, 28.9% C, 26.8% T, and 17.0% G. Maximum likelihood analyses placed D. sargus as a sister species of Diplodus puntazzo. This study provides valuable information for further studying identification methods and evolutionary relationships of Sparidae species.

Project description:Survival during the settlement window is a limiting variable for recruitment. The survival is believed to be strongly determined by biological interactions and sea conditions, however it has been poorly investigated. We examined the settlement patterns related to relevant biotic and abiotic factors (i.e. Density-dependence, wind stress, wave height and coastal current velocity) potentially determining post-settler survival rates of a coastal necto-benthic fish of wide distribution in the Mediterranean and eastern Atlantic, the white seabream (Diplodus sargus). An observational study of the demography of juveniles of this species was carried out at six coves in Menorca Island (Balearic Islands, western Mediterranean). Three of the coves were located in the northern and wind exposed coast, at the Northeast (NE) side; while the other three were found along the southern and sheltered coast, at the Southwest (SW) side of the island. The settlement period extended from early May to late June and maximum juvenile densities at the sampling sites varied between 5 and 11 ind. m-1 with maximum values observed in late May simultaneously occurring in the two coasts. Our analysis of juvenile survival, based on the interpretation of the observed patters using an individual based model (IBM), revealed two stages in the size-mortality relationships. An initial density-dependent stage was observed for juveniles up to 20 mm TL, followed by a density independent stage when other factors dominated the survival at sizes > 20 mm TL. No significant environmental effects were observed for the small size class (<20mm TL). Different significant environmental effects affecting NE and SW coves were observed for the medium (20-30mm TL) and large (>30mm TL) size class. In the NE, the wind stress consistently affected the density of fish of 20-30 mm and >30 mm TL with a dome-shape effect with higher densities at intermediate values of wind stress and negative effect at the extremes. The best models applied in the SW coves showed a significant non-linear negative effect on fish density that was also consistent for both groups 20-30 mm and >30 mm TL. Higher densities were observed at low values of wave height in the two groups. Because of these variations, the number of juveniles present at the end of the period was unrelated to their initial density and average survival varied among locations. In consequence, recruitment was (1) primarily limited by denso-dependient procedures at settlement stage, and (2) by sea conditions at post-settlement, where extreme wave conditions depleted juveniles. Accordingly, regional hydrodynamic conditions during the settlement season produced significant impacts on the juvenile densities depending on their size and with contrasted effects in respectto cove orientation. The similar strength in larval supply between coves, in addition to the similar mean phenology for settlers in the north and south of the Island, suggests that all fish may come from the same parental reproductive pool. These factors should be taken into account when assessing relationships between settlers, recruits and adults of white seabream.

Project description:BACKGROUND:Sparid fishes of the genus Diplodus show a complex life history. Juveniles have adaptations well suited to life in the water column. When fishes recruit into the adult population, individuals develop a radically differentiated shape that reflects their adaptation to the new benthic environment typical of the adult. A comparative analysis of ontogenetic trajectories was performed to assess the presence of divergence in the developmental pattern. By using a geometric morphometric approach, we investigated the pattern of shape variation across ontogenetic stages that span from early settlement to the adult stage in four species of the genus Diplodus. Landmarks were collected on the whole body of fishes to quantify the phenotypic variation along two well defined life stages, i.e. juvenile and adult. A comparative analysis of ontogenetic trajectories was performed to assess the presence of divergence in the developmental pattern. Subsequently, we investigated the patterns of integration and modularity as proxy for the alteration of the developmental processes. This have allowed to give an insight in morphological developmental patterns across ecologically and ontogenetically differentiated life stages and to investigate the process leading to the adult shape. RESULT:Our results suggest that the origin of morphological novelties in Diplodus spp. arise from shifts of the ontogenetic trajectories during development. During the settlement phase, the juveniles' morphological shapes converge towards similar regions of the morphospace. When the four species approach the transition between settlement and recruitment, we observe the lowest level of inter- and intra-specific disparity. After this transition we detect an abrupt shift of ontogenetic trajectories, i.e. the path taken by species during development, that led to highly divergent adult phenotypes. DISCUSSION:We suggest that the evolution of new ecomorphologies, better suited to exploit different niches (pelagic vs. benthonic) and reduce inter-specific competition in Diplodus spp., are related to the shift in the ontogenetic trajectory that in turn is associated to changes in modularity and integration pattern.

Project description:Recent literature suggests that anthropogenic stressors can disrupt ecologically relevant behaviours in fish, such as the ability to escape from predators. Disruption of these behaviours at critical life history transitions, such as the transition from the pelagic environment to the juvenile/adult habitat, may have even greater repercussions. The literature suggests that an increase in temperature can affect fish escape response, as well as metabolism; however, few studies have focused on the acute sensitivity responses and the potential for acclimation through developmental plasticity. Here, we aimed at evaluating the acute and long-term effects of exposure to warming conditions on the escape response and routine metabolic rate (RMR) of early life stages of the white seabream, Diplodus sargus. Additionally, as food availability may modulate the response to warming, we further tested the effects of long-term exposure to high temperature and food shortage, as individual and interacting drivers, on escape response and RMR. Temperature treatments were adjusted to ambient temperature (19°C) and a high temperature (22°C). Feeding treatments were established as high ration and low ration (50% of high ration). Escape response and RMR were measured after the high temperature was reached (acute exposure) and after 4 weeks (prolonged exposure). Acute warming had a significant effect on escape response and generated an upward trend in RMR. In the long term, however, there seems to be an acclimation of the escape response and RMR. Food shortage, interacting with high temperature, led to an increase in latency response and a significant reduction in RMR. The current study provides relevant experimental data on fishes' behavioural and physiological responses to the combined effects of multiple stressors. This knowledge can be incorporated in recruitment models, thereby contributing to fine-tuning of models required for fisheries management and species conservation.
