Copper (II) Ion-Modified Gold Nanoclusters as Peroxidase Mimetics for the Colorimetric Detection of Pyrophosphate.

ABSTRACT: Copper (II) ions have been shown to greatly improve the chemical stability and peroxidase-like activity of gold nanoclusters (AuNCs). Since the affinity between Cu2+ and pyrophosphate (PPi) is higher than that between Cu2+ and AuNCs, the catalytic activity of AuNCs-Cu2+ decreases with the introduction of PPi. Based on this principle, a new colorimetric detection method of PPi with high sensitivity and selectivity was developed by using AuNCs-Cu2+ as a probe. Under optimized conditions, the detection limit of PPi was 0.49 nM with a linear range of 0.51 to 30,000 nM. The sensitivity of the method was three orders of magnitude higher than that of a fluorescence method using AuNCs-Cu2+ as the probe. Finally, the AuNCs-Cu2+ system was successfully applied to directly determine the concentration of PPi in human urine samples.

Thermostable protein-stabilized gold nanoclusters as a peroxidase mimic.

Project description:Protein-stabilized gold nanoclusters (AuNCs) are fascinating nanostructures with exciting properties owing to their ultra-small sizes and functional shell. However, their applications under extreme conditions are still complicated, waiting for programmable solutions. Therefore, the design of a multi-functional protein stabilizer for specific purposes gains attention to improve the stability and functionality of AuNCs. Herein, we exploited the thermostability of genetically engineered KlenTaq DNA polymerase containing five cysteine residues (KTQ5C) to synthesize heat-stable AuNCs (AuNC@KTQ5C) and characterize optical, structural, and hydrodynamic properties. Besides their excellent photophysical properties, AuNC@KTQ5C also exhibit superior peroxidase-like (POD-like) catalytic activity following typical Michaelis-Menten kinetics together with a high affinity towards the POD substrate 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)-diammonium salt (ABTS). Moreover, FTIR and relative catalytic activity analysis of AuNC@KTQ5C reveal that KTQ5C is resistant to changes in protein secondary structure while the AuNCs conserve 70-80% of their catalytic performance after heat treatments up to more than 80 °C. Our findings show that stabilizing AuNCs with thermostable KTQ5C not only preserves the advantages of protein-stabilized AuNCs but can also promote the resistance of AuNCs against aggregation due to protein denaturation under extreme reaction temperatures, protecting their fluorescent emission or catalytic activity.

Silver Nanoclusters with Specific Ion Recognition Modulated by Ligand Passivation toward Fluorimetric and Colorimetric Copper Analysis and Biological Imaging.

Project description:Silver nanoclusters were synthesized and passivated by glutathione (GSH) ligand, with high aqueous stability and powerful red fluorescence and UV-vis yellow colour. Importantly, the specific recognition of the AgNCs was modulated from Hg(2+) ions to Cu(2+) ions upon the GSH passivation, of which the unique GSH-Cu(2+) chelating reaction could conduct the fluorescence quenching of AgNCs. Strong UV-vis absorbance of GSH-passivated AgNCs could also be realized depending on the Cu(2+) levels. Moreover, the Cu(2+)-induced loss of fluorescence and UV-vis absorbance of GSH-passivated AgNCs could be well restored by using stronger Cu(2+) chelating agent. A simultaneous and reversible fluorimetric and colorimetric sensing method was thereby developed for probing Cu(2+) ions in blood with high sensitivity and selectivity. Subsequently, the fluorescence-trackable imaging for live tissues and cells was demonstrated towards the analysis Cu(2+) ions using GSH-passivated AgNCs as the fluorescent probes. This study indicates that the use of functional ligands like GSH could not only modulate the specific ion recognition of AgNCs, but also endow them the high aqueous stability and powerful red fluorescence towards the wide applications for ion sensing and biological imaging in the complicated media like blood.

Protein-Localized Bright-Red Fluorescent Gold Nanoclusters as Cyanide-Selective Colorimetric and Fluorometric Nanoprobes.

Project description:Herein, we describe a bright-red-emitting ovalbumin-protected gold nanoclusters (OVA-AuNCs) that were prepared and applied as a luminescent probe for a simple, rapid, and highly sensitive determination of cyanide ions (CN- ions) based on an emission quenching and colorimetric method. Initially, an intense red-emissive fluorescence of the OVA-AuNCs successfully disappeared upon the addition of CN- ions. The resultant emission-quenching process involved CN- ions etching the OVA-AuNC surface, which produced AuCN2 - complexes in the presence of ambient oxygen. Under optimized experimental conditions, the relative emission intensity is inversely relative to CN- ion concentrations ranging from 5.00 × 10-7 to 75.00 × 10-7 mol/L with a linear correlation coefficient of 0.9932. Furthermore, OVA-AuNC-based optical detection systems on both colorimetric and fluorometric assays were tested, which expose highly sensitive and specific determination of CN- ions, and it is easily visualized by the naked eye (day light and UV light). Because of the distinct Elsner reaction between Au atoms of OVA-AuNCs and CN- ions, the recent nanoprobe offered ultrasensitivity and good selectivity with the lowest limit of detection value of 68.00 × 10-9 mol/L. In addition, this fluorescence "turn-off" CN- ion detection method was executed in real water samples. The demonstrated route of OVA-AuNC preparation is extremely easy and quick, making the proposed selective and sensitive CN- ion sensing assay based on the fluorescence response of the OVA-AuNCs for numerous practical applications.

Tandem copper and gold nanoclusters for two-color ratiometric explosives detection.

Project description:We report a sensory platform for the determination of common explosive species (e.g., TNT, PETN, RDX) based on the differential response from two different luminescent metal nanoclusters. In particular, whereas the red emission from bovine serum albumin-protected gold nanoclusters was strongly quenched by nitro-, nitrate-, and nitroamine-containing explosive organic molecules, blue-emitting glutathione-capped copper nanoclusters proved inert to quenching by these same analytes, instead showing evidence for aggregation-induced emission enhancement (AIEE). As a result, this discrete gold/copper nanocluster pairing provides a dual-probe, ratiometric (red-to-blue) system signaling the presence of TNT and other common explosives. This strategy opens up new potential for nanocluster-based analyte signaling, with implications to fluorescence resonance energy transfer (FRET) strategies as well.

A Quinoxaline-Naphthaldehyde Conjugate for Colorimetric Determination of Copper Ion.

Project description:This work facilitates detection of bivalent copper ion by a simple Schiff base probe QNH based on a quinoxaline-naphthaldehyde framework. The detailed study in absorption spectroscopy and theoretical aspects and crystal study of the probe and probe-copper complex has been discussed. The detection limit of the probe in the presence of Cu2+ is 0.45 µM in HEPES-buffer/acetonitrile (3/7, v/v) medium for absorption study. The reversibility of the probe-copper complex has been investigated by EDTA. The selective visual detection of copper has been established also in gel form.

Niacin Alternatives for Dyslipidemia: Fool's Gold or Gold Mine? Part II: Novel Niacin Mimetics.

Project description:Two cardiovascular outcome trials established niacin 3 g daily prevents hard cardiac events. However, as detailed in part I of this series, an extended-release (ER) alternative at only 2 g nightly demonstrated no comparable benefits in two outcome trials, implying the alternative is not equivalent to the established cardioprotective regimen. Since statins leave a significant treatment gap, this presents a major opportunity for developers. Importantly, the established regimen is cardioprotective, so the pathway is likely beneficial. Moreover, though effective, the established cardioprotective regimen is cumbersome, limiting clinical use. At the same time, the ER alternative has been thoroughly discredited as a viable substitute for the established cardioprotective regimen. Therefore, by exploiting the pathway and skillfully avoiding the problems with the established cardioprotective regimen and the ER alternative, developers could validate cardioprotective variations facing little meaningful competition from their predecessors. Thus, shrewd developers could effectively tap into a gold mine at the grave of the ER alternative. The GPR109A receptor was discovered a decade ago, leading to a large body of evidence commending the niacin pathway to a lower cardiovascular risk beyond statins. While mediating niacin's most prominent adverse effects, GPR109A also seems to mediate anti-lipolytic, anti-inflammatory, and anti-atherogenic effects of niacin. Several developers are investing heavily in novel strategies to exploit niacin's therapeutic pathways. These include selective GPR109A receptor agonists, niacin prodrugs, and a niacin metabolite, with encouraging early phase human data. In part II of this review, we summarize the accumulated results of these early phase studies of emerging niacin mimetics.

One-Pot Synthesis of Thiol-Modified Liquid Crystals Conjugated Fluorescent Gold Nanoclusters.

Project description:Gold nanoclusters (AuNCs) and liquid crystals (LCs) have shown great potential in nanobiotechnology applications due to their unique optical and structural properties. Herein, the hardcore of the 4-cyano biphenyl group for commonly used LCs of 4-cyano-4'-pentylbiphenyl (5CB) was utilized to synthesize 4'-(2-mercaptoethyl)-(1,1'-biphenyl)-4-carbonitrile (TAT-12) based on Suzuki coupling and Appel reaction. The structural and optical properties of thiol-modified TAT-12 LCs were demonstrated by nuclear magnetic resonance (NMR) spectroscopy, ultraviolet-visible (UV-vis) spectroscopy and differential scanning calorimetry (DSC). By one-pot synthesis, thiol-modified TAT-12 LCs were used as the ligands to prepare fluorescent gold nanoclusters (AuNCs@TAT-12) according to the Au-S bond between AuNCs and TAT-12. The spectra of UV-vis absorption and X-ray photoelectron spectroscopy (XPS) of AuNCs@TAT-12 indicated that the core of gold of AuNCs@TAT-12 exhibited high gold oxidation states. The fluorescence of AuNCs@TAT-12 was observed with a maximum intensity at ~352 nm coming from TAT-12 on AuNCs@TAT-12 and the fluorescence quantum yield of AuNCs@TAT-12 was calculated to be 10.1%. Furthermore, the fluorescence with a maximum intensity at ~448 nm was attributed to a ligand-metal charge transfer between the ligands of TAT-12 LCs and the core of AuNCs. The image of transmission electron microscopy (TEM) further demonstrated an approximately spherical shape of AuNCs@TAT-12 with an average size of 2.3 nm. A combination of UV-vis absorption spectra, XPS spectra, fluorescence spectra and TEM image, fluorescent AuNCs@TAT-12 were successfully synthesized via one-pot synthesis. Our work provides a practical approach to the synthesis of LCs conjugated AuNCs for future applications in nanobiotechnology.

Tryptophanhydroxamic Acid-Stabilized Ultrasmall Gold Nanoclusters: Tuning the Selectivity for Metal Ion Sensing.

Project description:Sub-nanometer-sized gold nanoclusters (Au NCs) were prepared via the spontaneous reduction of [AuCl4]-- ions with a hydroxamate derivative of L-tryptophan (Trp) natural amino acid (TrpHA). The prepared TrpHA-Au NCs possess intense blue emission (λem = 470 nm; λex = 380 nm) with a 2.13% absolute quantum yield and 1.47 ns average lifetime. The Trp-stabilized noble metal NCs are excellent metal ion sensors for Fe3+, but in this work, we highlighted that the incorporation of the hydroxamate functional group with an excellent metal ion binding capability can tune the selectivity and sensitivity of these NCs, which is a promising way to design novel strategies for the detection of other metal ions as well. Moreover, their simultaneous identification can also be realized. By decreasing the sensitivity of our nano-sensor for Fe3+ (limit of detection (LOD) ~11 µM), it was clearly demonstrated that the selectivity for Cu2+-ions can be significantly increased (LOD = 3.16 µM) in an acidic (pH = 3-4) condition. The surface-bounded TrpHA molecules can coordinate the Cu2+ confirmed by thermodynamic data, which strongly generates the linking of the NCs via the Cu2+ ions in acidic pH, and a parallel fluorescence quenching occurs. In the case of Fe3+, the degree of quenching strongly depends on the metal ion concentration, and it only occurs when the NCs are not able to bind more Fe3+ (~10 µM) on the surface, causing the NCs' aggregation.

Trace detection of tetrabromobisphenol A by SERS with DMAP-modified magnetic gold nanoclusters.

Project description:Magnetic gold nanoclusters (MGNCs) functionalized with 4-dimethylaminopyridine (DMAP) enables the trace detection of tetrabromobisphenol A (TBBPA), an environmental pollutant, using surface-enhanced Raman scattering (SERS) spectroscopy. The synthesis, cleansing, and functionalization of MGNCs are conducted in aqueous solutions; SERS samples are prepared by magnetic precipitation in the presence of trace analyte. The limit of detection (LOD) for TBBPA is greatly increased by the use of DMAP as a reporter molecule: DMAP-modified MGNCs can detect TBBPA at 10 pM in water, whereas the LOD for TBBPA by unfunctionalized Au is 1 nM. The reproducibility of picomolar TBBPA detection with DMAP-modified MGNCs is confirmed by two-dimensional correlation analysis. The high SERS sensitivity for TBBPA can be attributed to its capacity to modulate the Raman spectrum of adsorbed DMAP. This indirect mode of detection can also be applied toward the detection of other hydrophobic analytes, each identifiable by its characteristic SERS identity.
