Molluscicides against the snail-intermediate host of Schistosoma: a review.

ABSTRACT: Schistosomiasis, a neglected tropical disease (NTD), is one of the most prevalent parasitoses in the World. Certain freshwater snail species are the intermediate host in the life cycle of schistosome species. Controlling snails employing molluscicides is an effective, quick, and convenient intervention strategy to prevent the spread of Schistosoma species in endemic regions. Advances have been made in developing both synthetic molluscicides and molluscicides derived from plants. However, at present, the development of molluscicides is not adapted to the actual demand for snails and schistosoma controlling. We undertake a systematic review of exploitation and application of synthetic molluscicides and molluscicides derived from plants to combat intermediate host snails. The detailed molluscicidal activity, structure-activity relationship, structural feature, and possible mechanism of some molluscicides are also highlighted, which may afford an important reference for the design of new, more effective molluscicides with low environmental impact and realize the aim of controlling schistosome at transmission stages.

Project description:BackgroundSchistosoma japonicum is a parasitic flatworm that causes human schistosomiasis, which is a significant cause of morbidity in China, the Philippines and Indonesia. Oncomelania hupensis (Gastropoda: Pomatiopsidae) is the unique intermediate host of S. japonicum. A complete genome sequence of O. hupensis will enable the fundamental understanding of snail biology as well as its co-evolution with the S. japonicum parasite. Assembling a high-quality reference genome of O. hupehensis will provide data for further research on the snail biology and controlling the spread of S. japonicum.MethodsThe draft genome was de novo assembly using the long-read sequencing technology (PacBio Sequel II) and corrected with Illumina sequencing data. Then, using Hi-C sequencing data, the genome was assembled at the chromosomal level. CAFE was used to do analysis of contraction and expansion of the gene family and CodeML module in PAML was used for positive selection analysis in protein coding sequences.ResultsA total length of 1.46 Gb high-quality O. hupensis genome with 17 unique full-length chromosomes (2n = 34) of the individual including a contig N50 of 1.35 Mb and a scaffold N50 of 75.08 Mb. Additionally, 95.03% of these contig sequences were anchored in 17 chromosomes. After scanning the assembled genome, a total of 30,604 protein-coding genes were predicted. Among them, 86.67% were functionally annotated. Further phylogenetic analysis revealed that O. hupensis was separated from a common ancestor of Pomacea canaliculata and Bellamya purificata approximately 170 million years ago. Comparing the genome of O. hupensis with its most recent common ancestor, it showed 266 significantly expanded and 58 significantly contracted gene families (P < 0.05). Functional enrichment of the expanded gene families indicated that they were mainly involved with intracellular, DNA-mediated transposition, DNA integration and transposase activity.ConclusionsIntegrated use of multiple sequencing technologies, we have successfully constructed the genome at the chromosomal-level of O. hupensis. These data will not only provide the compressive genomic information, but also benefit future work on population genetics of this snail as well as evolutional studies between S. japonicum and the snail host.

Project description:BackgroundNeotricula aperta is the snail-intermediate host of the parasitic blood-fluke Schistosoma mekongi which causes Mekong schistosomiasis in Cambodia and the Lao PDR. Despite numerous phylogenetic studies only one DNA-sequence based population-genetic study of N. aperta had been published, and the origin, structure and persistence of N. aperta were poorly understood. Consequently, a phylogenetic and population genetic study was performed, with addition of new data to pre-existing DNA-sequences for N. aperta from remote and inaccessible habitats, including one new taxon from Laos and 505 bp of additional DNA-sequence for all sampled taxa,.Principal findingsSpatial Principal Component Analysis revealed the presence of significant spatial-genetic clustering. Genetic-distance-based clustering indicated four populations with near perfect match to a priori defined ecogeographical regions. Spring-dwelling taxa were found to form an ecological isolate relative to other N. aperta. The poor dispersal capabilities suggested by spatial-genetic analyses were confirmed by Bayesian inference of migration rates. Population divergence time estimation implied a mid-Miocene colonisation of the present range, with immediate and rapid radiation in each ecogeographical region. Estimated effective population sizes were large (120-310 thousand).ConclusionsThe strong spatial-genetic structure confirmed the poor dispersal capabilities of N. aperta-suggesting human-mediated reintroduction of disease to controlled areas as the primary reason for control failure. The isolation of the spring-dwelling taxa and ecogeographical structure suggests adaptation of sub-populations to different habitats; the epidemiological significance of this needs investigation. The large effective population sizes indicate that the high population densities observed in surveyed habitats are also present in inaccessible areas; affording great potential for recrudescence driven by animal-reservoir transmission in remote streams. Mid-Miocene colonisation implies heterochronous evolution of these snails and associated schistosomes and suggests against coevolution of snail and parasite. Heterochronicity favours ecological factors as shapers of host-parasite specificity and greater potential for escape from schistosomiasis control through host-switching.

Project description:BackgroundThere is a need for current and more detailed information on the distribution of Biomphalaria pfeifferi snails in East Africa's river systems. B. pfeifferi is arguably the most important snail intermediate host in the transmission of schistosomiasis, a disease ranked second to malaria in terms of tropical diseases of public health importance.MethodsWe assessed the occurrence and geographical distribution of B. pfeifferi snails in Kenya, Uganda and Tanzania. Maximum entropy modelling was used to predict the potential distribution of B. pfeifferi snails and malacological surveys were conducted guided by MaxEnt predictions and information from previous studies. Malacological surveys were conducted at a total of 172 sites, including streams, rivers, dams, irrigation schemes and springs over a 3-y period from 2018 to 2020, with geospatial, ecological and physicochemical information recorded for each site.ResultsB. pfeifferi snails were found at 23 of the 172 sites and inhabited a variety of habitat types. Of the 23 sites where B. pfeifferi snails were found, 15 (65.2%) were streams, 3 rivers (13.04%), 2 dams (8.7%), 2 springs (8.7%) and 1 an irrigation scheme (4.35%). B. pfeifferi abundance showed a significant positive correlation with increasing water temperature and decreasing water depth. In Kenya, B. pfeifferi snails were found around the Lake Victoria basin, the Mwea irrigation scheme and in parts of the former Eastern Province of Kenya. In Uganda, B. pfeifferi snails were found in Jinja District, Ntoroko District and Soroti District. In Tanzania, B. pfeifferi snails were found in the Iringa, Tabora and Kigoma Regions. We observed moderate to high prevalence of Schistosoma mansoni infection, with S. mansoni-infected snails found at 11 of 23 sites and with an average prevalence of 24.9% at infected sites. In Kenya, S. mansoni-infected snails were found in the Lake Victoria basin (22.5% prevalence at infected sites) and the former Eastern Province (13.5% prevalence at infected sites). In Uganda, infected snails were found in Ntoroko District (100% infected) and Soroti District (20% infected). In Tanzania, infected snails were found in the Kigoma Region, with a prevalence of 10% at the infected site.ConclusionThis information on the distribution of B. pfeifferi snails and S. mansoni infection in East Africa's river systems can aid in developing better prevention and control strategies for human schistosomiasis. Regular surveys of the river systems for snail intermediate hosts followed by molecular detection of schistosome infection could form a basis for the development of a prompt and cost-effective surveillance system for schistosomiasis in the region.

Project description:Biomphalaria glabrata is the mollusc intermediate host for Schistosoma mansoni, a digenean flatworm parasite that causes human intestinal schistosomiasis. An estimated 200 million people in 74 countries suffer from schistosomiasis, in terms of morbidity this is the most severe tropical disease after malaria. Epigenetic information informs on the status of gene activity that is heritable, for which changes are reversible and that is not based on the DNA sequence. Epigenetic mechanisms generate variability that provides a source for potentially heritable phenotypic variation and therefore could be involved in the adaptation to environmental constraint. Phenotypic variations are particularly important in host-parasite interactions in which both selective pressure and rate of evolution are high. In this context, epigenetic changes are expected to be major drivers of phenotypic plasticity and co-adaptation between host and parasite. Consequently, with characterization of the genomes of invertebrates that are parasite vectors or intermediate hosts, it is also essential to understand how the epigenetic machinery functions to better decipher the interplay between host and parasite.The CpGo/e ratios were used as a proxy to investigate the occurrence of CpG methylation in B. glabrata coding regions. The presence of DNA methylation in B. glabrata was also confirmed by several experimental approaches: restriction enzymatic digestion with isoschizomers, bisulfite conversion based techniques and LC-MS/MS analysis.In this work, we report that DNA methylation, which is one of the carriers of epigenetic information, occurs in B. glabrata; approximately 2% of cytosine nucleotides are methylated. We describe the methylation machinery of B. glabrata. Methylation occurs predominantly at CpG sites, present at high ratios in coding regions of genes associated with housekeeping functions. We also demonstrate by bisulfite treatment that methylation occurs in multiple copies of Nimbus, a transposable element.This study details DNA methylation for the first time, one of the carriers of epigenetic information in B. glabrata. The general characteristics of DNA methylation that we observed in the B. glabrata genome conform to what epigenetic studies have reported from other invertebrate species.
