The complete chloroplast of Chrysanthemum × morifolium 'Fubaiju'.

ABSTRACT: Chrysanthemum × morifolium 'Fubaiju,' which is native to Macheng, Hubei, China, has a long cultivation history almost dating back to the early 10th century Song dynasty, and is used as Chrysanthemum tea drink and Chinese traditional medicine. In this study, the complete chloroplast genome sequence of 'Fubaiju' was 151,109 bp, included a large single copy LSC (82,931 bp), a small single copy SSC (18,350 bp), and a pair of inverted repeat regions (24,941 bp). It contained 132 genes with 87 CDS, 8r RNA, and 37 tRNA. The phylogenetic analysis showed that the C. × morifolium 'Fubaiju' was clustered together with C. × morifolium 'Baekma'.

Complete chloroplast genome sequence of a Chinese traditional cultivar in Chrysanthemum, Chrysanthemum morifolium 'Anhuishiliuye'.

Project description:The complete plastid genome of Chrysanthemum morifolium 'Anhuishiliuye', a Chinese traditional cultivar, was determined and analyzed in this work. It had a circular-mapping molecular with the length of 151,059 bp.The LSC and SSC of 82,857 bp and 18,294 bp were separated by two IRs of 24,954 bp. The chloroplast genome of C. morifolium 'Anhuishiliuye' contains 125 genes, including 83 protein-coding genes, 34 ribosomal RNA genes and 8 transfer RNA genes. Phylogenetic analysis showed that C. morifolium 'Anhuishiliuye' clustered together with other Chrysanthemum species. The data provided would be useful for elucidation of phylogenetics and evolution in Chrysanthemum cultivars.

The complete chloroplast genome sequence of one economic cultivar 'Baekma' was 151,060 bp in length with a large single copy (LSC) region (82,862 bp), a small single copy (SSC) region (18,294 bp) and two inverted repeats (IRs) (24,952 bp). It contained 130 genes, including 85 protein-coding genes, 8 rRNAs and 37 tRNAs. The overall GC content was 37%. Phylogenetic analysis showed that C. morifolium 'Baekma' was grouped together with other Chrysanthemum species.

It is a circular chromosome and has a length of 151,060 bp. The LSC and SSC of 82,858 bp and 18,294 bp were separated by two IRs of 24,954 bp. The chloroplast genome of C. morifolium 'Orizaba' contains 125 genes, including 83 protein-coding genes, eight ribosomal RNA genes, and 34 transfer RNA genes. Phylogenetic analysis showed that C. morifolium 'Orizaba' clustered together with other Chrysanthemum varieties in the family Asteraceae. The plastome is useful for the elucidation of phylogenetics and evolution in the Asteraceae and Chrysanthemum varieties.

Identification of chrysanthemum (Chrysanthemum morifolium) self-incompatibility.

Project description:There has been a heated argument over self-incompatibilityof chrysanthemum (Chrysanthemum morifolium) among chrysanthemum breeders. In order to solve the argument, we investigated pistil receptivity, seed set, and compatible index of 24 chrysanthemum cultivars. It was found that the 24 cultivars averagely had 3.7-36.3 pollen grains germinating on stigmas at 24 hours after self-pollination through the fluorescence microscope using aniline blue staining method. However, only 10 of them produced self-pollinated seeds, and their seed sets and compatible indexes were 0.03-56.50% and 0.04-87.50, respectively. The cultivar "Q10-33-1" had the highest seed set (56.50%) and compatible index (87.50), but ten of its progeny had a wide range of separation in seed set (0-37.23%) and compatible index (0-68.65). The results indicated that most of chrysanthemum cultivars were self-incompatible, while a small proportion of cultivars were self-compatible. In addition, there is a comprehensive separation of self-incompatibility among progeny from the same self-pollinated self-compatible chrysanthemum cultivar. Therefore, it is better to emasculate inflorescences during chrysanthemum hybridization breeding when no information concerning its self-incompatibility characteristics is available. However, if it is self-incompatible and propagated by vegetative methods, it is unnecessary to carry out emasculation when it is used as a female plant during hybridization breeding.

MicroRNA Expression Profile during Aphid Feeding in Chrysanthemum (Chrysanthemum morifolium).

Project description:MicroRNAs (miRNAs) are important regulators of gene expression, affecting many biological processes. As yet, their roles in the response of chrysanthemum to aphid feeding have not been explored. Here, the identity and abundance of miRNAs induced by aphid infestation have been obtained using high-throughput Illumina sequencing platform. Three leaf small RNA libraries were generated, one from plants infested with the aphid Macrosiphoniella sanbourni (library A), one from plants with mock puncture treatment (library M), and the third from untreated control plants (library CK). A total of 7,944,797, 7,605,251 and 9,244,002 clean unique reads, ranging from 18 to 30 nucleotides (nt) in length, were obtained from library CK, A and M, respectively. As a result, 303 conserved miRNAs belonging to 276 miRNAs families and 234 potential novel miRNAs were detected in chrysanthemum leaf, out of which 80, 100 and 79 significantly differentially expressed miRNAs were identified in the comparison of CK-VS-A, CK-VS-M and M-VS-A, respectively. Several of the differentially abundant miRNAs (in particular miR159a, miR160a, miR393a) may be associated with the plant's response to aphid infestation.

Gene expression profiles responses to aphid feeding in chrysanthemum (Chrysanthemum morifolium).

Project description:BackgroundChrysanthemum is an important ornamental plant all over the world. It is easily attacked by aphid, Macrosiphoniella sanbourni. The molecular mechanisms of plant defense responses to aphid are only partially understood. Here, we investigate the gene expression changes in response to aphid feeding in chrysanthemum leaf by RNA-Seq technology.ResultsThree libraries were generated from pooled leaf tissues of Chrysanthemum morifolium 'nannongxunzhang' that were collected at different time points with (Y) or without (CK) aphid infestations and mock puncture treatment (Z), and sequenced using an Illumina HiSeqTM 2000 platform. A total of 7,363,292, 7,215,860 and 7,319,841 clean reads were obtained in library CK, Y and Z, respectively. The proportion of clean reads was >97.29% in each library. Approximately 76.35% of the clean reads were mapped to a reference gene database including all known chrysanthemum unigene sequences. 1,157, 527 and 340 differentially expressed genes (DEGs) were identified in the comparison of CK-VS-Y, CK-VS-Z and Z-VS-Y, respectively. These DEGs were involved in phytohormone signaling, cell wall biosynthesis, photosynthesis, reactive oxygen species (ROS) pathway and transcription factor regulatory networks, and so on.ConclusionsChanges in gene expression induced by aphid feeding are shown to be multifaceted. There are various forms of crosstalk between different pathways those genes belonging to, which would allow plants to fine-tune its defense responses.

Chrysanthemum (Chrysanthemum morifolium) CmHRE2-like negatively regulates the resistance of chrysanthemum to the aphid (Macrosiphoniella sanborni).

Project description:BackgroundThe growth and ornamental value of chrysanthemums are frequently hindered by aphid attacks. The ethylene-responsive factor (ERF) gene family is pivotal in responding to biotic stress, including insect stress. However, to date, little is known regarding the involvement of ERF transcription factors (TFs) in the response of chrysanthemum to aphids.ResultsIn the present study, CmHRE2-like from chrysanthemum (Chrysanthemum morifolium), a transcription activator that localizes mainly to the nucleus, was cloned. Expression is induced by aphid infestation. Overexpression of CmHRE2-like in chrysanthemum mediated its susceptibility to aphids, whereas CmHRE2-like-SRDX dominant repressor transgenic plants enhanced the resistance of chrysanthemum to aphids, suggesting that CmHRE2-like contributes to the susceptibility of chrysanthemum to aphids. The flavonoids in CmHRE2-like-overexpression plants were decreased by 29% and 28% in two different lines, whereas they were increased by 42% and 29% in CmHRE2-like-SRDX dominant repressor transgenic plants. The expression of Chrysanthemum-chalcone-synthase gene(CmCHS), chalcone isomerase gene (CmCHI), and flavonoid 3'-hydroxylase gene(CmF3'H) was downregulated in CmHRE2-like overexpression plants and upregulated in CmHRE2-like-SRDX dominant repressor transgenic plants, suggesting that CmHRE2-like regulates the resistance of chrysanthemum to aphids partially through the regulation of flavonoid biosynthesis.ConclusionCmHRE2-like was a key gene regulating the vulnerability of chrysanthemum to aphids. This study offers fresh perspectives on the molecular mechanisms of chrysanthemum-aphid interactions and may bear practical significance for developing new strategies to manage aphid infestation in chrysanthemums.

Genetic mapping of quantitative trait loci underlying flowering time in chrysanthemum (Chrysanthemum morifolium).

Project description:Flowering time is an important trait in chrysanthemum, but its genetic basis remains poorly understood. An intra-specific mapping population bred from the cross between the autumn-flowering cultivar 'Yuhualuoying' and the summer-flowering 'Aoyunhanxiao' was used to determine the number and relative effect of QTL segregating for five measures of flowering time. From flowering time data recorded over two consecutive seasons, 35 additive QTL were detected, each explaining between 5.8% and 22.7% of the overall phenotypic variance. Of these, 13 were detected in both years. Nine genomic regions harboring QTL for at least two of the five traits were identified. Ten pairs of loci epistatically determined the flowering time, but their contribution to the overall phenotypic variance was less than for the additive QTL. The results suggest that flowering time in chrysanthemum is principally governed by main effect QTL but that epistasis also contributes to the genetic architecture of the trait, and the major QTL identified herein are useful in our ongoing efforts to streamline the improvement of chrysanthemum via the use of molecular methodology.

It was predicted to contain 131 genes, including 87 protein-coding genes, eight rRNAs and 46 tRNAs. Phylogenetic analysis of chloroplast genomes clustered C. boreale with other Chrysanthemum and Asteraceae species.
