ABSTRACT: Xanthomonas Oryzae pv. oryzae (Xoo) causes bacterial blight and Rhizoctonia solani (R. solani) causes sheath blight in rice accounting for >75% of crop losses. Therefore, there is an urgent need to develop strategies for the mitigation of these pathogen infections. In this study, we report the antimicrobial efficacy of Cholic Acid-Glycine Conjugates (CAGCs) against Xoo and R. solani. We show that CAGC C6 is a broad-spectrum antimicrobial and is also able to degrade biofilms. The application of C6 did not hamper plant growth and showed minimal effect on the plant cell membranes. Exogenous application of C6 on pre-infection or post-infection of Xoo on rice susceptible genotype Taichung native (TN1) can mitigate the bacterial load and improve resistance through upregulation of plant defense genes. We further demonstrate that C6 can induce plant defense responses when seeds were primed with C6 CAGC. Therefore, this study demonstrates the potential of CAGCs as effective antimicrobials for crop protection that can be further explored for field applications.

Project description:Soil salinity is one of the important abiotic stress factors that affect rice productivity and quality. Research with several dicotyledonous plants indicated that the detrimental effects associated with salinity stress can (partly) be overcome by the external application of antioxidative substances. For instance, sodium selenate (Na2SeO4) significantly improved the growth and productivity of several crops under various abiotic stress conditions. At present there is no report describing the impact of Na2SeO4 on salinity stressed cereals such as rice. Rice cultivation is threatened by increasing salinity stress, and in future this problem will further be aggravated by global warming and sea level rise, impacting coastal areas. The current study reports on the effect of Na2SeO4 in alleviating salinity stress in rice plants. The optimal concentration of Na2SeO4 and the most efficient mode of selenium application were investigated. Selenium, sodium, and potassium contents in leaves were determined. Antioxidant enzyme activities as well as proline, hydrogen peroxide (H2O2), and malondialdehyde (MDA) concentrations were analyzed. In addition, the transcript levels for OsNHX1, an important Na+/H+ antiporter, were quantified. Treatment of 2-week-old rice plants under 150 mM NaCl stress with 6 mg l-1 Na2SeO4 improved the total biomass. A significantly higher biomass was observed for the plants that received Na2SeO4 by a combination of seed priming and foliar spray compared to the individual treatments. The Na2SeO4 application enhanced the activity of antioxidant enzymes (SOD, APX, CAT, and GSH-Px), increased the proline content, and reduced H2O2 and MDA concentrations in plants under NaCl stress. These biochemical changes were accompanied by increased transcript levels for OsNHX1 resulting in a higher K+/Na+ ratio in the rice plants under NaCl stress. The results suggest that Na2SeO4 treatment alleviates the adverse effect of salinity on rice plant growth through enhancing the antioxidant defense system and increase of OsNHX1 transcript levels.

Project description:Susceptibility of plants to abiotic stresses, including extreme temperatures, salinity and drought, poses an increasing threat to crop productivity worldwide. Here the drought-induced response of maize was modulated by applications of methyl jasmonate (MeJA) and salicylic acid (SA) to seeds prior to sowing and to leaves prior to stress treatment. Pot experiments were conducted to ascertain the effects of exogenous applications of these hormones on maize growth, physiology and biochemistry under drought stress and well-watered (control) conditions. Maize plants were subjected to single as well as combined pre-treatments of MeJA and SA. Drought stress severely affected maize morphology and reduced relative water content, above and below-ground biomass, rates of photosynthesis, and protein content. The prolonged water deficit also led to increased relative membrane permeability and oxidative stress induced by the production of malondialdehyde (from lipid peroxidation), lipoxygenase activity (LOX) and the production of H2O2. The single applications of MeJA and SA were not found to be effective in maize for drought tolerance while the combined pre-treatments with exogenous MeJA+SA mitigated the adverse effects of drought-induced oxidative stress, as reflected in lower levels of lipid peroxidation, LOX activity and H2O2. The same pre-treatment also maintained adequate water status of the plants under drought stress by increasing osmolytes including proline, total carbohydrate content and total soluble sugars. Furthermore, exogenous applications of MeJA+SA approximately doubled the activities of the antioxidant enzymes catalase, peroxidase and superoxide dismutase. Pre-treatment with MeJA alone gave the highest increase in drought-induced production of endogenous abscisic acid (ABA). Pre-treatment with MeJA+SA partially prevented drought-induced oxidative stress by modulating levels of osmolytes and endogenous ABA, as well as the activities of antioxidant enzymes. Taken together, the results show that seed and foliar pre-treatments with exogenous MeJA and/or SA can have positive effects on the responses of maize seedlings to drought.
