Project description:HCN1 hyperpolarization-activated cation channels act as an inhibitory constraint of both spatial learning and synaptic integration and long-term plasticity in the distal dendrites of hippocampal CA1 pyramidal neurons. However, as HCN1 channels provide an excitatory current, the mechanism of their inhibitory action remains unclear. Here we report that HCN1 channels also constrain CA1 distal dendritic Ca2+ spikes, which have been implicated in the induction of LTP at distal excitatory synapses. Our experimental and computational results indicate that HCN1 channels provide both an active shunt conductance that decreases the temporal integration of distal EPSPs and a tonic depolarizing current that increases resting inactivation of T-type and N-type voltage-gated Ca2+ channels, which contribute to the Ca2+ spikes. This dual mechanism may provide a general means by which HCN channels regulate dendritic excitability.

Project description:Bivalent ligands targeting putative melanocortin receptor dimers have been developed and characterized in vitro; however, studies of their functional in vivo effects have been limited. The current report compares the effects of homobivalent ligand CJL-1-87, Ac-His-DPhe-Arg-Trp-PEDG20-His-DPhe-Arg-Trp-NH2, to monovalent ligand CJL-1-14, Ac-His-DPhe-Arg-Trp-NH2, on energy homeostasis in mice after central intracerebroventricular (ICV) administration into the lateral ventricle of the brain. Bivalent ligand CJL-1-87 had noteworthy advantages as an antiobesity probe over CJL-1-14 in a fasting-refeeding in vivo paradigm. Treatment with CJL-1-87 significantly decreased food intake compared to CJL-1-14 or saline (50% less intake 2-8 h after treatment). Furthermore, CJL-1-87 treatment decreased the respiratory exchange ratio (RER) without changing the energy expenditure indicating that fats were being burned as the primary fuel source. Additionally, CJL-1-87 treatment significantly lowered body fat mass percentage 6 h after administration (p < 0.05) without changing the lean mass percentage. The bivalent ligand significantly decreased insulin, C-peptide, leptin, GIP, and resistin plasma levels compared to levels after CJL-1-14 or saline treatments. Alternatively, ghrelin plasma levels were significantly increased. Serum stability of CJL-1-87 and CJL-1-14 (T1/2 = 6.0 and 16.8 h, respectively) was sufficient to permit physiological effects. The differences in binding affinity of CJL-1-14 compared to CJL-1-87 are speculated as a possible mechanism for the bivalent ligand's unique effects. We also provide in vitro evidence for the formation of a MC3R-MC4R heterodimer complex, for the first time to our knowledge, that may be an unexploited neuronal molecular target. Regardless of the exact mechanism, the advantageous ability of CJL-1-87 compared to CJL-1-14 to increase in vitro binding affinity, increase the duration of action in spite of decreased serum stability, decrease in vivo food intake, decrease mice's body fat percent, and differentially affect mouse hormone levels demonstrates the distinct characteristics achieved from the current melanocortin agonist bivalent design strategy.

Project description:Soil acidification often occurs when the concentration of ammonium (NH4 +) in soil rises, such as that observed in farmland. Both soil acidification and excess NH4 + have serious adverse effects on crop growth and food production. However, we still do not know which of these two inhibitors has a greater impact on the growth of crops, and the degree of their inhibitory effect on crop growth have not been accurately evaluated. 31 wheat cultivars originating in various areas of China were planted under 5 mM sole NH4 + (ammonium nitrogen, AN) or nitrate nitrogen in combined with two pH levels resembling acidified conditions (5.0 and 6.5). The results showed that the shoots and roots biomass were severely reduced by AN in both and these reduction effects were strengthened by a low medium pH. The concentration of free NH4 + and amino acids, the glutamine synthetase activity were significantly higher, but the total soluble sugar content was reduced under NH4 + conditions, and the glutamine synthetase activity was reduced by a low medium pH. Cultivar variance was responsible for the largest proportion of the total variance in plant dry weight, leaf area, nodal root number, total root length and root volume; the nitrogen (N) form explains most of the variation in N and C metabolism; the effects of pH were the greatest for plant height and root average diameter. So, soil acidification and excess NH4 + would cause different degrees of inhibition effects on different plant tissues. The findings are expected to be useful for applying effective strategies for reducing NH4 + stress in the field.

Project description:Understanding the structural dynamics of lead-halide perovskites is essential for their advanced use as photovoltaics. Here, the structural dynamics of the CH3NH3 cation and PbBr6 octahedra in the perovskite CH3NH3PbBr3 were studied via nuclear magnetic resonance (NMR) to determine the mechanism of the transition from the tetragonal to cubic phase. The chemical shifts were obtained by 1H, 13C, and 207Pb magic angle spinning NMR and 14N static NMR. The chemical shifts of the 1H nuclei in CH3 and NH3 remained constant with increasing temperature, whereas those of the 13C and 207Pb nuclei varied near the phase transition temperature (TC?=?236 K), indicating that the structural environments of 13C and 207Pb change near TC. The spin-lattice relaxation time T1? values for 1H, 13C, and 207Pb nuclei increased with increasing temperature and did not exhibit an abrupt change near TC. In addition, the two lines in the 14N NMR spectra superposed into one line near TC, indicating the occurrence of a phase transition to a cubic phase with higher symmetry than tetragonal. Consequently, the main factor causing the phase transition from the tetragonal to cubic phase near TC is a change in the surroundings of the 207Pb nuclei in the PbBr6 octahedra and of the C-N groups in the CH3NH3 cations.

At the plasma membrane, sensory TRPs act as neuronal detectors of physical and chemical environmental signals, and receptor-operated (metabotropic) TRPs decode extracellular neuroendocrine cues to control body homeostasis. In intracellular membranes, such as those in lysosomes, organellar TRPs respond to compartment-derived signals to control membrane trafficking, signal transduction, and organelle function. Complementing mouse and human genetics and high-resolution structural approaches, physiological studies employing natural agonists and synthetic inhibitors have become critical in resolving the in vivo functions of metabotropic, sensory, and organellar TRPs.

Project description:BackgroundIn northwest of China, mini Chinese cabbage (Brassica pekinensis) is highly valued by consumers, and is widely cultivated during winter in solar-greenhouses where low light (LL) fluence (between 85 and 150 μmol m-2 s-1 in day) is a major abiotic stress factor limiting plant growth and crop productivity. The mechanisms with which various NH4+: NO3- ratios affected growth and photosynthesis of mini Chinese cabbage under normal (200 μmol m-2 s-1) and low (100 μmol m-2 s-1) light conditions was investigated. The four solutions with different ratios of NH4+: NO3- applied were 0:100, 10:90, 15:85 and 25:75 with the set up in a glasshouse in hydroponic culture. The most appropriate NH4+: NO3- ratio that improved the tolerance of mini Chinese cabbage seedlings to LL was found in our current study.ResultsUnder low light, the application of NH4+: NO3- (10:90) significantly stimulated growth compared to only NO3- by increasing leaf area, canopy spread, biomass accumulation, and net photosynthetic rate. The increase in net photosynthetic rate was associated with an increase in: 1) maximum and effective quantum yield of PSII; 2) activities of Calvin cycle enzymes; and 3) levels of mRNA relative expression of several genes involved in Calvin cycle. In addition, glucose, fructose, sucrose, starch and total carbohydrate, which are the products of CO2 assimilation, accumulated most in the cabbage leaves that were supplied with NH4+: NO3- (10:90) under LL condition. Low light reduced the carbohydrate: nitrogen (C: N) ratio while the application of NH4+: NO3- (10:90) alleviated the negative effect of LL on C: N ratio mainly by increasing total carbohydrate contents.ConclusionsThe application of NH4+:NO3- (10:90) increased rbcL, rbcS, FBA, FBPase and TK expression and/or activities, enhanced photosynthesis, carbohydrate accumulation and improved the tolerance of mini Chinese cabbage seedlings to LL. The results of this study would provide theoretical basis and technical guidance for mini Chinese cabbage production. In practical production, the ratio of NH4+:NO3- should be adjusted with respect to light fluence for successful growing of mini Chinese cabbage.

Project description:Aqueous ammonium ion batteries are regarded as eco-friendly and sustainable energy storage systems. And applicable host for NH4+ in aqueous solution is always in the process of development. On the basis of density functional theory calculations, the excellent performance of NH4+ insertion in Prussian blue analogues (PBAs) is proposed, especially for copper hexacyanoferrate (CuHCF). In this work, we prove the outstanding cycling and rate performance of CuHCF via electrochemical analyses, delivering no capacity fading during ultra-long cycles of 3000 times and high capacity retention of 93.6% at 50 C. One of main contributions to superior performance from highly reversible redox reaction and structural change is verified during the ammoniation/de-ammoniation progresses. More importantly, we propose the NH4+ diffusion mechanism in CuHCF based on continuous formation and fracture of hydrogen bonds from a joint theoretical and experimental study, which is another essential reason for rapid charge transfer and superior NH4+ storage. Lastly, a full cell by coupling CuHCF cathode and polyaniline anode is constructed to explore the practical application of CuHCF. In brief, the outstanding aqueous NH4+ storage in cubic PBAs creates a blueprint for fast and sustainable energy storage.

Project description:BackgroundAmmonium (NH4+), a key nitrogen form, becomes toxic when it accumulates to high levels. Ammonium transporters (AMTs) are the key transporters responsible for NH4+ uptake. AMT activity is under allosteric feedback control, mediated by phosphorylation of a threonine in the cytosolic C-terminus (CCT). However, the kinases responsible for the NH4+-triggered phosphorylation remain unknown.ResultsIn this study, a functional screen identified protein kinase CBL-Interacting Protein Kinase15 (CIPK15) as a negative regulator of AMT1;1 activity. CIPK15 was able to interact with several AMT1 paralogs at the plasma membrane. Analysis of AmTryoshka, an NH4+ transporter activity sensor for AMT1;3 in yeast, and a two-electrode-voltage-clamp (TEVC) of AMT1;1 in Xenopus oocytes showed that CIPK15 inhibits AMT activity. CIPK15 transcript levels increased when seedlings were exposed to elevated NH4+ levels. Notably, cipk15 knockout mutants showed higher 15NH4+ uptake and accumulated higher amounts of NH4+ compared to the wild-type. Consistently, cipk15 was hypersensitive to both NH4+ and methylammonium but not nitrate (NO3-).ConclusionTaken together, our data indicate that feedback inhibition of AMT1 activity is mediated by the protein kinase CIPK15 via phosphorylation of residues in the CCT to reduce NH4+-accumulation.

Project description:Compared to the commonly applied metallic ion charge carriers (e.g., Li+ and Na+ ), batteries using nonmetallic charge carriers (e.g., H+ and NH4 + ) generally have much faster kinetics and high-rate capability thanks to the small hydrated ionic sizes and nondiffusion control topochemistry. However, the hosts for nonmetallic charge carriers are still limited. In this work, it is suggested that mixed ionic-electronic conductors can serve as a promising host for NH4 + storage. Using hexagonal tungsten oxide (h-WO3 ) as an example, it is shown that the existence of ionic conductive tunnels greatly promotes the high-rate NH4 + storage. Specifically, a much higher capacity of 82 mAh g-1 at 1 A g-1 is achieved on h-WO3 , in sharp contrast to 14 mAh g-1 of monoclinic tungsten oxide (m-WO3 ). In addition, unlike layered materials, the insertion and desertion of NH4 + ions are confined within the tunnels of the h-WO3 , which minimizes the damage to the crystal structure. This leads to outstanding stability of up to 200 000 cycles with 68% capacity retention at a high current of 20 A g-1 .

Project description:BackgroundNitrogen-13 has a 10-min half-life which places time constraints on the complexity of viable synthetic methods for its incorporation into PET imaging agents. In exploring ways to overcome this limitation, we have used the Ugi reaction to develop a rapid one-pot method for radiolabelling peptidic molecules using [13N]NH3 as a synthetic precursor.MethodsCarrier-added [13N]NH3 (50 μL) was added to a solution of carboxylic acid, aldehyde, and isocyanide in 2,2,2-TFE (200 μL). The mixture was heated in a microwave synthesiser at 120 °C for 10 min. Reactions were analysed by radio-HPLC and radio-LCMS. Isolation of the target 13N-labelled peptidic Ugi compound was achieved via semi-preparative radio-HPLC as demonstrated for Ugi1.ResultsRadio-HPLC analysis of each reaction confirmed the formation of radioactive products co-eluting with their respective reference standards with radiochemical yields of the crude products ranging from 11% to 23%. Two cyclic γ-lactam structures were also achieved via intra-molecular reactions. Additional radioactive by-products observed in the radio-chromatogram were identified as 13N-labelled di-imines formed from the reaction of [13N]NH3 with two isocyanide molecules. The desired 13N-labelled Ugi product was isolated using semi-preparative HPLC.ConclusionWe have developed a one-pot method that opens up new routes to radiolabel complex, peptidic molecules with 13N using aqueous [13N]NH3 as a synthetic precursor.
