Visible light-induced oxidative N-dealkylation of alkylamines by a luminescent osmium(vi) nitrido complex.

ABSTRACT: N-Dealkylation of amines by metal oxo intermediates (M[double bond, length as m-dash]O) is related to drug detoxification and DNA repair in biological systems. In this study, we report the first example of N-dealkylation of various alkylamines by a luminescent osmium(vi) nitrido complex induced by visible light.

A breast cancer stem cell-selective, mammospheres-potent osmium(VI) nitrido complex.

Project description:The effect of a newly developed osmium(VI) nitrido complex, 1, on breast cancer stem cells (CSCs) is reported. The complex displays selective toxicity for HMLER breast cancer cells enriched with CD44-positive, CSC-like cells over the same cells having reduced CSC character. Remarkably, 1 also reduces the proportion of CSCs within a heterogeneous breast cancer cell population and irreversibly inhibits the formation of free-floating mammospheres to an extent similar to that of salinomycin, a natural product that targets CSCs. Detailed mechanistic studies reveal that in breast cancer cells 1 induces DNA damage and endoplasmic reticulum stress, the latter being responsible for the CSC selectivity. The anti-CSC properties of 1 provide a strong impetus for the development of new metal-based compounds to target CSCs and to treat chemotherapy-resistant and relapsed tumors.

Bidentate ligands on osmium(VI) nitrido complexes control intracellular targeting and cell death pathways.

Project description:The cellular response evoked by antiproliferating osmium(VI) nitrido compounds of general formula OsN(N^N)Cl3 (N^N = 2,2'-bipyridine 1, 1,10-phenanthroline 2, 3,4,7,8-tetramethyl-1,10-phenanthroline 3, or 4,7-diphenyl-1,10-phenanthroline 4) can be tuned by subtle ligand modifications. Complex 2 induces DNA damage, resulting in activation of the p53 pathway, cell cycle arrest at the G2/M phase, and caspase-dependent apoptotic cell death. In contrast, 4 evokes endoplasmic reticulum (ER) stress leading to the upregulation of proteins of the unfolded protein response pathway, increase in ER size, and p53-independent apoptotic cell death. To the best of our knowledge, 4 is the first osmium compound to induce ER stress in cancer cells.

High-Spin Iron(VI), Low-Spin Ruthenium(VI), and Magnetically Bistable Osmium(VI) in Molecular Group 8 Nitrido Trifluorides NMF3.

Project description:Pseudo-tetrahedral nitrido trifluorides N≡MF3 (M=Fe, Ru, Os) and square pyramidal nitrido tetrafluorides N≡MF4 (M=Ru, Os) were formed by free-metal-atom reactions with NF3 and subsequently isolated in solid neon at 5 K. Their IR spectra were recorded and analyzed aided by quantum-chemical calculations. For a d2 electron configuration of the N≡MF3 compounds in C3v symmetry, Hund's rule predict a high-spin 3 A2 ground state with two parallel spin electrons and two degenerate metal d(δ)-orbitals. The corresponding high-spin 3 A2 ground state was, however, only found for N≡FeF3 , the first experimentally verified neutral nitrido FeVI species. The valence-isoelectronic N≡RuF3 and N≡OsF3 adopt different angular distorted singlet structures. For N≡RuF3 , the triplet 3 A2 state is only 5 kJ mol-1 higher in energy than the singlet 1 A' ground state, and the magnetically bistable molecular N≡OsF3 with two distorted near degenerate 1 A' and 3 A" electronic states were experimentally detected at 5 K in solid neon.

Electrophilic Activation of Osmium-Nitrido Corroles: The OsN Triple Bond as a π-Acceptor Metallaligand in a Heterobimetallic OsVIN-PtII Complex.

Project description:Presented herein is a first investigation of the chemical reactivity of osmium-nitrido corroles, which are known for their unusual thermal, chemical, and photochemical stability. Elemental chlorine perchlorinates the β-positions of the triarylcorrole but leaves the OsN unit untouched. The OsN unit is also unaffected by a variety of other electrophilic and nucleophilic reagents. Upon photolysis, however, the anion of Zeise's salt associates with the nitrido ligand to generate an OsVI≡N-PtII complex. The very short OsN-Pt linkage [1.895(9)-1.917(8) Å] and the downfield 195Pt NMR resonance (-2702 ppm) suggest that the OsN corrole acts as a π-accepting ligand toward the Pt(II) center. This finding represents a rare example of the successful photochemical activation of a metal-ligand multiple bond that is too kinetically inert to exhibit any appreciable reactivity under thermal conditions.

Modular synthesis of α-branched secondary alkylamines via visible-light-mediated carbonyl alkylative amination.

Project description:The development of methods for the assembly of secondary α-alkyl amines remains a central challenge to chemical synthesis because of their critical importance in modulating the physical properties of biologically active molecules. Despite decades of intensive research, chemists still rely on selective N-alkylation and carbonyl reductive amination to make most amine products. Here we report the further evolution of a carbonyl alkylative amination process that, for the first time, brings together primary amines, aldehydes and alkyl iodides in a visible-light-mediated multicomponent coupling reaction for the synthesis of a wide range of α-branched secondary alkylamines. In addition to exploring the tolerance and limitations in each reaction component, we also report preliminary applications to the telescoped synthesis of α-branched N-heterocycles and an N-alkylation protocol that is selective for primary over cyclic secondary amines. Our data support a mechanism involving addition of an alkyl radical to an uncharged alkyl imine which, to the best of our knowledge, has not previously been described. We believe that this method will enable practitioners of synthetic chemistry in academic and industrial settings to approach the synthesis of these important molecules in a manner that is streamlined compared to established approaches.

Visible-light mediated carbonyl trifluoromethylative amination as a practical method for the synthesis of β-trifluoromethyl tertiary alkylamines.

Project description:We report the development of an operationally straigtforward, visible-light-mediated multicomponent strategy for the construction of β-trifluoromethylated tertiary alkylamines from feedstock aldehydes, secondary amines and a convenient source of trifluoromethyl iodide. The new process does not require a photocatalyst, is metal-free, displays a broad functional group tolerance and offers rapid, one-pot access to trifluoromethylated drug-like compounds that will be of interest in medicinal chemistry.

[2+2] cycloadditions by oxidative visible light photocatalysis.

Project description:Photochemical reactions are remarkable for their ability to easily assemble cyclobutanes and other strained ring systems that are difficult to construct using other conventional synthetic methods. We have previously shown that Ru(bpy)(3)(2+) is an efficient photocatalyst that promotes the [2+2] cycloadditions of electron-deficient olefins with visible light. Here, we show that Ru(bpy)(3)(2+) is also an effective photocatalyst for the [2+2] cycloaddition of electron-rich olefins. This transformation is enabled by the versatile photoelectrochemical properties of Ru(bpy)(3)(2+), which enables either one-electron reduction or one-electron oxidation of interesting organic substrates under appropriate conditions.

Visible Light-Promoted Oxidative Cross-Coupling of Alcohols to Esters.

Project description:Ester is one of the most significant functional groups in organic chemistry and is enclosed in several valued molecules. Usually, esters are prepared through the acid-catalyzed esterification reaction of carboxylic acids with alcohols, transesterification of esters with alcohols, or via activation of carboxylic acids followed by the addition of alcohols. However, these procedures typically imply the excess use of reactants and harsh reaction conditions. Visible light-mediated photoreactions have been disclosed to display a safe, sustainable, and accessible alternative to traditional methods, and to lead new reactivity modes in organic procedures. In this context, we propose a transition metal-based and organic-based photocatalyst-free synthesis of esters from alcohols induced by visible light. The methodology can be carried out using sunlight or artificial visible light as a solar simulator or a blue LED source.

Green Synthesis of 8-Hydroxyquinoline Barium as Visible-Light-Excited Luminescent Material Using Mechanochemical Activation Method.

Project description:Using high-energy UV-light to excite 8-hydroxyquinoline barium (BaQ2) is a short slab for this emerging area of organic luminescent materials. However, using visible light to excite BaQ2 has not been reported. To solve this problem, this study proposes the mechanochemical activation method to synthesize luminescent material of visible-light-excited BaQ2. This research applies infrared spectroscopy, X-ray diffraction (XRD), X-ray photoelectron spectroscopy, scanning electron microscopy, energy dispersive spectroscopy, and fluorescence spectrometry to analyze the structure and luminescence properties. XRD shows that BaQ2 has a high crystallinity, small crystalline size, and high purity. According to the Scherrer equation, the mean particle size is 56 nm. The results of fluorescence spectrometry show that the excitation spectrum of the product is red-shifted, and the maximum excitation wavelength is 408 nm. According to these results, the product has a high fluorescence and can be excited under visible light. This research explains the high efficiency of the mechanochemical-activation method by thermodynamic and dynamic principles. This research also exemplifies luminescence theory of BaQ2 and at the microlevel explains the theory of visible-light-excited theory and the principle of luminescent intensity enhancement from the point of crystallography.

Green Synthesis of Luminescent Gold-Zinc Oxide Nanocomposites: Cell Imaging and Visible Light-Induced Dye Degradation.

Project description:Green synthesis of gold-zinc oxide (Au-ZnO) nanocomposite was successfully attempted under organic solvent-free conditions at room temperature. Prolonged stirring of the reaction mixture introduced crystallinity in the ZnO phase of Au-ZnO nanocomposites. Luminescence properties were observed in these crystalline Au-ZnO nanocomposites due to in situ embedding of gold nanoparticles (AuNP) of 5-6 nm diameter on the surface. This efficient strategy involved the reduction of Au(III) by Zn(0) powder in aqueous medium, where sodium citrate (NaCt) was the stabilizing agent. Reaction time and variation of reagent concentrations were investigated to control the Au:Zn ratio within the nanocomposites. The reaction with the least amount of NaCt for a long duration resulted in Au-ZnO/Zn(OH)2 nanocomposite. X-ray photoelectron spectroscopy (XPS) confirmed the formation of Zn(OH)2 and ZnO in the same nanocomposite. These nanocomposites were reconnoitered as bioimaging materials in human cells and applied for visible light-induced photodegradation of rhodamine-B dye.
