Project description:The applied pyrolysis temperature was found to strongly affect composition, structure, and oxidation behavior of pure and iron oxide nanoparticle (NP)-loaded carbon materials originating from hydrothermal carbonization (HTC) of cellulose. A strong loss of functional groups during pyrolysis at temperatures beyond 300 °C of the HTC-derived hydrochars was observed, resulting in an increase of the carbon content up to 95 wt% for the carbon materials pyrolyzed at 800 °C and an increase of the specific surface area with a maximum of 520 m2 g-1 at a pyrolysis temperature of 600 °C. Devolatilization mainly took place in the range from 300 to 500 °C, releasing light pyrolysis gases such as CO, CO2, H2O and larger oxygen-containing molecules up to C11. The presence of iron oxide NPs lowered the specific surface areas by about 200 m2 g-1 and resulted in the formation of mesopores. For the iron oxide-containing composites pyrolyzed up to 500 °C, the oxidation temperature was decreased by about 100 °C, indicating tight contact between the iron oxide NPs and the carbon matrix. For higher pyrolysis temperatures, this catalytic effect of iron oxide on carbon oxidation vanished due to carbothermal reduction to iron and iron carbide, which, however, catalyzed the graphitization of the carbon matrix. Thus, the well-controlled two-step synthesis based on a biomass-derived precursor yielded stably embedded iron NPs in a corrosion-resistant graphitic carbon matrix.

Project description:Effects of physical and chemical states of iron-based catalysts on the formation of carbon-encapsulated iron nanoparticles (CEINs) synthesized thermally from kraft lignin were investigated. Experimental results indicated that if solution-based iron nitrate (FeN) was used as an iron source for the catalyst, CEINs observed were ?-Fe and ?-Fe-based cores encapsulated by few layers graphitic-carbon (mostly 1-5 layers) and the majority of these CEINs were embedded in amorphous carbon matrix. The formation of graphitic-carbon shells is believed based on the dissolution and precipitation mechanism of amorphous carbon acting as the carbon source. If solid-based iron nanoparticles (FePs) were used as the catalyst, CEINs observed were ?-Fe, ?-Fe, and Fe?C-based cores encapsulated with tangled graphitic-carbon nanoribbons and carbon tubules and the majority of these CEINs were found along the edge of amorphous carbon matrix. The growth of tangled graphitic-carbon nanoribbons and carbon tubules is based on a chemical vapor decomposition process, i.e., the carbonaceous gases from kraft lignin decomposition served as the carbon source.

Project description:Trackable spheres of similar size to those typically used for sustained protein delivery are prepared by incorporating superparamagnetic iron oxide (SPIO) nanoparticles into the core of poly(lactide-co-glycolide) microspheres. The visibility of injections in static and temporally in dynamic tissue systems is demonstrated. This method improves upon other, less sensitive imaging modalities in their ability to track injectable delivery systems. The results obtained confirm the localization of microspheres to the injected target area and highlight the novelty of tracking delivery vehicles for other applications.

Project description:In this research, we report the size-controlled synthesis and surface-functionalization of magnetite with the natural antioxidant gallic acid (GA) as a ligand, using in situ and post-synthesis methods. GA functionalization provided narrow size distribution, with an average particle size of 5 and 8 nm for in situ synthesis of gallic acid functionalized magnetite IONP@GA1 and IONP@GA2, respectively, which are ultra-small particles as compared to unfunctionalized magnetite (IONP) and post functionalized magnetite IONP@GA3 with average size of 10 and 11 nm respectively. All the IONPs@GA samples were found hydrophilic with stable aggregation state. Prior to commencement of experimental lab work, PASS software was used to predict the biological activities of GA and it is found that experimental antioxidant activity using 2,2-diphenyl-1-picrylhydrazyl (DPPH) assay and antimicrobial studies using well diffusion method are in good agreement with the simulated results. Furthermore, the half maximal inhibitory concentration (IC50) values of DPPH antioxidant assay revealed a 2-4 fold decrease as compared to unfunctionalized IONP. In addition to antioxidant activity, all the three IONP@GA proved outstanding antimicrobial activity while testing on different bacterial and fungal strains. The results collectively indicate the successful fabrication of novel antioxidant, antimicrobial IONP@GA composite, which are magnetically separable, efficient, and low cost, with potential applications in polymers, cosmetics, and biomedical and food industries.

Project description:Carbon-encapsulated iron nanoparticles (CEINs) are emerging as promising biomedical tools due to their unique physicochemical properties. In this study, the cytotoxic effect of CEINs (the mean diameter distribution ranges 46-56 nm) has been explored by MTT, LDH leakage, Calcein-AM/propidium iodide (PI) and Annexin V-FITC/PI assays in human melanoma (HTB-140), mouse melanoma (B16-F10) cells, and human dermal fibroblasts (HDFs). The results demonstrated that CEINs produce mitochondrial and cell membrane cytotoxicities in a dose (0.0001-100 ?g/ml)-dependent manner. Moreover, the studies elucidated some differences in cytotoxic effects between CEINs used as raw and purified materials composing of the carbon surface with acidic groups. Experiments showed that HTB-140 cells are more sensitive to prone early apoptotic events due to raw CEINs as compared to B16-F10 or HDF cells, respectively. Taken together, these results suggest that the amount of CEINs administered to cells and the composition of CEINs containing different amounts of iron as well as the carbon surface modification type is critical determinant of cytotoxic responses in both normal and cancer (melanoma) cells.

Project description:A metal-free carbon catalyst is a kind of oxygen reduction catalyst with great prospects. It is an important material with potential to replace the traditional Pt catalyst. In this paper, a kind of irregular and ultra-thin carbon nanosheet (K180M-300-900) with high catalytic activity was synthesized by hydrothermal calcination using okra as a biomass and NH4Cl as an N source. The prepared nitrogen-doped metal-free catalyst with high pyridine-N and graphitic-N provides an extremely large number of active sites and has certain lattice defects. Ultra-thin carbon nanosheets promote sufficient contact between the catalyst and electrolyte, promote the diffusion of oxygen, and result in a faster transfer rate of electrons. The initial potential and half-slope potential of K180M-300-900 are 0.99 V and 0.82 V, respectively, which are comparable to those of 20% Pt/C. In addition, the stability and methanol tolerance of this catalyst (K180M-300-900) are better than 20% Pt/C, so it has great development potential and application value. This result provides a new method to prepare metal-free carbon materials that will take the place of traditional Pt catalysts.

Project description:Sub-50 nm iron-nitrogen-doped hollow carbon sphere-encapsulated iron carbide nanoparticles (Fe3C-Fe,N/C) are synthesized by using a triblock copolymer of poly(styrene-b-2-vinylpyridine-b-ethylene oxide) as a soft template. Their typical features, including a large surface area (879.5 m2 g-1), small hollow size (≈16 nm), and nitrogen-doped mesoporous carbon shell, and encapsulated Fe3C nanoparticles generate a highly active oxygen reduction reaction (ORR) performance. Fe3C-Fe,N/C hollow spheres exhibit an ORR performance comparable to that of commercially available 20 wt% Pt/C in alkaline electrolyte, with a similar half-wave potential, an electron transfer number close to 4, and lower H2O2 yield of less than 5%. It also shows noticeable ORR catalytic activity under acidic conditions, with a high half-wave potential of 0.714 V, which is only 59 mV lower than that of 20 wt% Pt/C. Moreover, Fe3C-Fe,N/C has remarkable long-term durability and tolerance to methanol poisoning, exceeding Pt/C regardless of the electrolyte.

Project description:The widespread use of fuel cells is currently limited by the lack of efficient and cost-effective catalysts for the oxygen reduction reaction. Iron-based non-precious metal catalysts exhibit promising activity and stability, as an alternative to state-of-the-art platinum catalysts. However, the identity of the active species in non-precious metal catalysts remains elusive, impeding the development of new catalysts. Here we demonstrate the reversible deactivation and reactivation of an iron-based non-precious metal oxygen reduction catalyst achieved using high-temperature gas-phase chlorine and hydrogen treatments. In addition, we observe a decrease in catalyst heterogeneity following treatment with chlorine and hydrogen, using Mössbauer and X-ray absorption spectroscopy. Our study reveals that protected sites adjacent to iron nanoparticles are responsible for the observed activity and stability of the catalyst. These findings may allow for the design and synthesis of enhanced non-precious metal oxygen reduction catalysts with a higher density of active sites.

Project description:A magneto-responsive energy/drug carrier that enhances deep tumor penetration with a porous nano-composite is constructed by using a tumor-targeted lactoferrin (Lf) bio-gate as a cap on mesoporous iron oxide nanoparticles (MIONs). With a large payload of a gas-generated molecule, perfluorohexane (PFH), and a hydrophobic anti-cancer drug, paclitaxel (PTX), Lf-MIONs can simultaneously perform bursting gas generation and on-demand drug release upon high-frequency magnetic field (MF) exposure. Biocompatible PFH was chosen and encapsulated in MIONs due to its favorable phase transition temperature (56 °C) and its hydrophobicity. After a short-duration MF treatment induces heat generation, the local pressure increase via the gasifying of the PFH embedded in MION can substantially rupture the three-dimensional tumor spheroids in vitro as well as enhance drug and carrier penetration. As the MF treatment duration increases, Lf-MIONs entering the tumor spheroids provide an intense heat and burst-like drug release, leading to superior drug delivery and deep tumor thermo-chemo-therapy. With their high efficiency for targeting tumors, Lf-MIONs/PTX-PFH suppressed subcutaneous tumors in 16 days after a single MF exposure. This work presents the first study of using MF-induced PFH gasification as a deep tumor-penetrating agent for drug delivery.

Project description:Nanostructured materials that have low tissue toxicity, multi-modal imaging capability and high photothermal conversion efficiency have great potential to enable image-guided near infrared (NIR) photothermal therapy (PTT). Here, we report a bifunctional nanoparticle (BFNP, ?16?nm) comprised of a magnetic Fe3O4 core (?9.1?nm) covered by a fluorescent carbon shell (?3.4?nm) and prepared via a one-pot solvothermal synthesis method using ferrocene as the sole source. The BFNP exhibits excitation wavelength-tunable, upconverted and near-infrared (NIR) fluorescence property due to the presence of the carbon shell, and superparamagnetic behavior resulted from the Fe3O4 core. BFNPs demonstrate dual-modal imaging capacity both in vitro and in vivo with fluorescent imaging excited under a varying wavelength from 405?nm to 820?nm and with T2-weighted magnetic resonance imaging (r2?=?264.76?mM-1?s-1). More significantly, BFNPs absorb and convert NIR light to heat enabling photothermal therapy as demonstrated mice bearing C6 glioblastoma. These BFNPs show promise as an advanced nanoplatform to provide imaging guided photothermal therapy.
