Ecological effect assessment by species sensitivity distribution for 38 pesticides with various modes of action.

ABSTRACT: Species sensitivity distributions (SSDs) of 38 pesticides with various modes of action were analyzed as a higher-tier ecological effect assessment based on collected acute toxicity data. Then the 5% hazardous concentrations (HC5) based on each SSD were calculated as the predicted no-effect concentrations for aquatic ecosystems. The differences between HC5 and registration criteria were small (within ten-fold) for 35 of the 38 pesticides. However, there were more than ten-fold differences for a fungicide and two herbicides. These results suggest that the current effect assessment scheme could underestimate the effect of such pesticides. This could be caused by differences in sensitivity of specific properties of the mode of action.

Project description:In this study, the species sensitivity distributions (SSDs) of 68 pesticides commonly used in Japanese paddy fields were analyzed based on collected acute toxicity data as a higher-tier ecological effect assessment. Then, based on each SSD, the 5% hazardous concentration (HC5) values were calculated as the predicted no-effect concentrations for aquatic ecosystems. The differences between HC5 and registration criteria were small (within 10-fold) for 50 of the 68 pesticides. However, there were more than 10-fold differences for 9 insecticides and 9 herbicides of specific modes of action. These results suggest that the current effect assessment scheme could underestimate the effect of such pesticides. This is caused by the mode of action specific properties of species sensitivity difference.

Sensitivity differences among seven algal species to 12 herbicides with various modes of action.

Project description:Seven algal species were used to conduct toxicity assays with 12 herbicides to determine differences in species sensitivity. A fluorescence microplate toxicity assay was used as an efficient and economical high-throughput assay. The obtained toxicity data were standardized based on the species sensitivity distribution concept. The most-sensitive individual species differed among herbicides: Desmodesmus subspicatus was most sensitive to chloronitrofen and pendimethalin; Achnanthidium minutissimum was most sensitive to chlorpropham; Nitzschia palea was most sensitive to diquat, glyphosate, and dichlobenil; Navicula pelliculosa was most sensitive to trifluralin; and Pseudanabaena foetida was most sensitive to glufosinate, asulam, and 2,4-D. Surprisingly, Raphidocelis (formerly Pseudokirchneriella) subcapitata, a standard green alga, was not the most sensitive to any of the herbicides. The results clearly showed that a single algal species cannot represent the algal assemblage in terms of sensitivity. Therefore, multispecies algal toxicity data sets are essential for assessing the ecological effect of herbicides.

Distribution of toxicity values across different species and modes of action of pesticides from PESTIMEP and PPDB databases.

Project description:The continuous use of compounds contained in commodities such as processed food, medicines, and pesticides, demands safety measures, in particular, for those in direct contact with humans and the environment. Safety measures have evolved and regulations are now in place around the globe. In the case of pesticides, attempts have been made to use toxicological data to inform of potentially harmful compounds either across species, on different routes of exposure, or entirely new chemicals. The generation of models, based on statistical and molecular modeling studies, allows for such predictions. However, the use of these models is framed by the available data, the experimental errors, the complexity of the measurement, and the available computational algorithms, among other factors. In this work, we present the methodologies used for extrapolation across different species and routes of administration and show the appropriateness of developing predictive models of pesticides based on their type and mode of action. The analyses include comparisons based on structural characteristics and physicochemical properties. Whenever possible, the scope and limitations of the methodologies are discussed. We expect that this work will serve as a useful introductory guide of the tools employed in the toxicity assessment of agrochemical compounds.

Use of the Species Sensitivity Distribution Approach to Derive Ecological Threshold of Toxicological Concern (eco-TTC) for Pesticides.

Project description:The species sensitivity distribution (SSD) calculates the hazardous concentration at which 5% of species (HC5) will be potentially affected. For many compounds, HC5 values are unavailable impeding the derivation of SSD curves. Through a detailed bibliographic survey, we selected HC5 values (from acute toxicity tests) for freshwater aquatic species and 129 pesticides. The statistical distribution and variability of the HC5 values within the chemical classes were evaluated. Insecticides are the most toxic compounds in the aquatic communities (HC5 = 1.4 × 10-3 µmol L-1), followed by herbicides (HC5 = 3.3 × 10-2 µmol L-1) and fungicides (HC5 = 7.8 µmol L-1). Subsequently, the specificity of the mode of action (MoA) of pesticides on freshwater aquatic communities was investigated by calculating the ratio between the estimated baseline toxicity for aquatic communities and the HC5 experimental values gathered from the literature. Moreover, we proposed and validated a scheme to derive the ecological thresholds of toxicological concern (eco-TTC) of pesticides for which data on their effects on aquatic communities are not available. We proposed eco-TTCs for different classes of insecticides, herbicides, and fungicides with a specific MoA, and three eco-TTCs for those chemicals with unavailable MoA. We consider the proposed approach and eco-TTC values useful for risk management purposes.

Comprehensive Ecological Risk Assessment of Heavy Metals Based on Species Sensitivity Distribution in Aquatic of Coastal Areas in Hong Kong.

Project description:In recent decades, the ecological environment of some coastal areas in China has been seriously affected by terrestrial pollutants, and there is an urgent need for ecological risk assessment of China's coastal environment. The assessment of heavy metal pollution in Hong Kong waters was carried out using different environmental and ecological indicators. The heavy metal contents (Cu, Pb, Zn, Cd, As, Cr, and Hg) in the near coast of Hong Kong were analyzed for two different seasons of the year 2018 (April-spring and September-autumn). We assessed the distribution and enrichment of heavy metals in the near coast of Hong Kong, and the potential biohazardous effects were assessed using the species sensitivity distribution method. The results showed that only Pb, Zn, and Hg in seawater exceeded the Class I standard. Pb, Zn, Cd, and As in organisms exceeded the standard, and no heavy metals exceeded the standard in sediments. The species sensitivity distribution method indicated that the biohazardous factor of heavy metals of the Hong Kong coast is higher in spring than in autumn, and the potential hazard ratio has the characteristics of high northwest and low southeast, which leads to its msPAF also having these characteristics. From the correlational analyses among heavy metals, we found that the pH change in seawater was related to the concentration of heavy metals, the concentration of heavy metals in seawater was proportional to the salinity of seawater, Pb and Cu were likely to have the same source, and Zn and Cd may not have the same emission sources as the other heavy metals. Overall, heavy metal contamination of seawater, sediments, and organisms near the Hong Kong coast was within acceptable limits, but the problem of heavy metal dispersion should be prevented.

Temporal distribution and ecological risk assessment for pesticides in water from the north-central coastal zone of Sinaloa, Mexico.

Project description:Water contamination with pesticides is one of the major pollution problems in northwestern Mexico, and this is due to the extensive use of pesticides in agriculture. In this research, water samples of ten sampling sites (fishing grounds, beaches, and both) were analyzed in the search for 28 pesticides (organochlorines, organophosphates, pyrethroids, carbamates, among other chemical classes), supplemented with a calculation of the resulting potential environmental risk. Pesticides were separated from the matrix by liquid-liquid extraction and quantified by gas chromatography coupled to electron micro-capture (organohalogenated) and pulsed flame photometric detectors (organophosphates). In addition, the ecotoxicological risk of pesticides in algae, invertebrates, and fish was assessed, based on seawater pesticide concentrations using the Risk Quotient (RQ) and Toxic Units (TU) approach. The results showed 18 pesticides identified in the analyzed samples, where cypermethrin and chlorpyrifos were identified with the maximum concentrations of 1.223 and 0.994 μg L-1, respectively. In addition, these two pesticides have been associated with acute toxic effects on algae, invertebrates, and fish. It is important to pay particular attention to the search for long-term alternatives to the use of chlorpyrifos and cypermethrin due to their high detection rates and the risks associated with their toxic properties. However, the adoption of alternative measures to synthetic pesticide control should be a priority, moving towards sustainable practices such as the use of biopesticides, crop rotation and polycultures.

The interplay of various sources of noise on reliability of species distribution models hinges on ecological specialisation.

Project description:Digitized species occurrence data provide an unprecedented source of information for ecologists and conservationists. Species distribution model (SDM) has become a popular method to utilise these data for understanding the spatial and temporal distribution of species, and for modelling biodiversity patterns. Our objective is to study the impact of noise in species occurrence data (namely sample size and positional accuracy) on the performance and reliability of SDM, considering the multiplicative impact of SDM algorithms, species specialisation, and grid resolution. We created a set of four 'virtual' species characterized by different specialisation levels. For each of these species, we built the suitable habitat models using five algorithms at two grid resolutions, with varying sample sizes and different levels of positional accuracy. We assessed the performance and reliability of the SDM according to classic model evaluation metrics (Area Under the Curve and True Skill Statistic) and model agreement metrics (Overall Concordance Correlation Coefficient and geographic niche overlap) respectively. Our study revealed that species specialisation had by far the most dominant impact on the SDM. In contrast to previous studies, we found that for widespread species, low sample size and low positional accuracy were acceptable, and useful distribution ranges could be predicted with as few as 10 species occurrences. Range predictions for narrow-ranged species, however, were sensitive to sample size and positional accuracy, such that useful distribution ranges required at least 20 species occurrences. Against expectations, the MAXENT algorithm poorly predicted the distribution of specialist species at low sample size.

Ecological Risk Assessment of Nano-enabled Pesticides: A Perspective on Problem Formulation.

Project description:Plant protection products containing nanomaterials that alter the functionality or risk profile of active ingredients (nano-enabled pesticides) promise many benefits over conventional pesticide products. These benefits may include improved formulation characteristics, easier application, better targeting of pest species, increased efficacy, lower application rates, and enhanced environmental safety. After many years of research and development, nano-enabled pesticides are starting to make their way into the market. The introduction of this technology raises a number of issues for regulators, including how does the ecological risk assessment of nano-enabled pesticide products differ from that of conventional plant protection products? In this paper, a group drawn from regulatory agencies, academia, research, and the agrochemicals industry offers a perspective on relevant considerations pertaining to the problem formulation phase of the ecological risk assessment of nano-enabled pesticides.

Various modes of action of dietary phytochemicals, sulforaphane and phenethyl isothiocyanate, on pathogenic bacteria.

Project description:Isothiocyanates (ITCs) derived from cruciferous plants reveal antibacterial activity, although detailed mechanism is not fully elucidated. Recently it has been reported that ITCs induce the stringent response in Escherichia coli strains. The aim of this work was to determine whether two isothiocyanates, sulforaphane (SFN) and phenethyl isothiocyanate (PEITC), similarly as in E. coli induce stringent response in Bacillus subtilis, model Gram(+) bacterium, and test their potency against a panel of clinical isolates belonging to Gram(+) or Gram(-) groups. Minimal inhibitory concentrations were determined as well as effect of ITCs on membranes integrity, synthesis of DNA, RNA and stringent response alarmones was assessed. SFN and PEITC are effective against B. subtilis and bacterial isolates, namely E. coli, K. pneumonia, S. aureus, S. epidermidis and E. faecalis. Interestingly, in B. subtilis and E. faecalis the inhibition of growth and nucleic acids synthesis is independent of ppGpp accumulation. In bacteria, which do not induce the stringent response in the presence of ITCs, membrane integrity disruption is observed. Thus, ITCs are effective against different pathogenic bacteria and act by at least two mechanisms depending on bacteria species.
