The complete chloroplast genome of <i>Stephania tetrandra</i> (Menispermaceae).

Project description:The complete chloroplast genome of Stephania tetrandra was sequenced and assembled for the first time. The chloroplast genome is 159,974 bp in length, containing a large single-copy (LSC) region of 90,539 bp and a small single-copy region (SSC) of 20,735 bp, separated by a pair of inverted repeats (IRs) of 24,350 bp. The genome contains 113 unique genes, including 79 protein-coding genes (PCGs), 30 tRNA genes, and four rRNA genes. Among them, 15 genes have one intron each and three genes contain two introns. The overall GC content is 37.8%, while the corresponding values of LSC, SSC, and IR regions are 35.8, 32.4, and 43.7%, respectively. Phylogenetic analysis showed that S. tetrandra is more closely related to the clade of two species within Stephania, providing new insight into the evolution of Menispermaceae.

Project description:Stephania kwangsiensis (Menispermaceae) is a rare and critically endangered species endemic to China. Here, we first report and characterize its complete chloroplast genome sequence based on Illumina paired-end sequencing data. The complete plastid genome was 156,624 bp, which contained inverted repeats (IR) of 24,348 bp separated by a large single-copy (LSC) and a small single copy (SSC) of 87,759 bp and 20,169 bp, respectively. The cpDNA contains 132 genes, comprising 85 protein-coding genes, 37 tRNA genes, eight rRNA genes, and two processed pseudogenes. The overall GC content of the plastome is 38.4%. The phylogenetic analysis of 18 selected chloroplast genomes demonstrated that S. kwangsiensis is related to the congeneric S. japonica.

Project description:Stephania epigaea H. S. Lo, 1978 is a medicinal plant commonly used in southwest China. This study characterized the first complete chloroplast (cp) genome sequence of this species. The complete cp was 157,738 bp in length, containing a large single-copy region (LSC) of 88,460 bp, a small single-copy region (SSC) of 19,778 bp, and a pair of inverted repeat regions (IRs) of 24,750 bp. It encoded 130 genes, including 85 protein-coding genes, 37 tRNA genes, and 8 rRNA genes. The GC content of the complete genome was 36.7%. Phylogenetic analysis of complete cp sequences revealed that S. epigaea was clustered with S. japonica from the Menispermaceae family.

Alkaloid variations within the genus <i>Stephania</i> (Menispermaceae) in China.

Project description:The genus Stephania, which is rich in alkaloids, has been used as a traditional medicine or folklore herb against numerous ailments in China. However, the understanding of the variation within the genus Stephania is obscure, which limits the optimal utilization of the genus. An evaluation of the variation within the genus Stephania would help screen the ideal Stephania genotypes for drug utilization. In the present study, alkaloids in the tubers of four commonly cultivated Stephania species in China, i.e., the genotype Stephania kwangsiensis Lo. (SK-guangxi) from Guangxi Province and three Stephania yunnanensis H.S. Lo. genotypes (SY-xueteng, SY-hongteng, and SY-lvteng) sourced from Yunnan Province, were investigated, and the genus variations were compared. The results revealed significant variations in the abundance of alkaloids in tubers within the genus Stephania. The Stephania genotypes SY-xueteng and SY-hongteng showed a relatively high abundance of total alkaloids compared with the Stephania genotypes SK-guangxi and SY-lvteng. Specifically, the Stephania genotype SY-xueteng had a relatively high abundance of palmatine in tubers, and the Stephania genotype SY-hongteng exhibited a high abundance of stephanine in tubers. Our study provides foundations for further utilization of ideal Stephania genotypes by clarifying the variations in the alkaloid contents within the genus in China.
