Project description:Molecular machines and switches composed of flexible pseudorotaxanes respond to external stimuli, transducing incident energy into mechanical motions. This study presents thermo- and photoresponsive dynamic pseudorotaxane crystals composed of axle molecules containing ferrocene or ruthenocene groups threaded through dibenzo[24]crown-8 ether rings. The ruthenocene-containing pseudorotaxane exhibits a crystal-to-crystal thermal phase transition at 86?°C, which is much lower than that of the ferrocene-containing pseudorotaxane (128?°C). Single-crystal X-ray crystallography at various temperatures reveals the details of the structural changes, and shows that the bulky ruthenocene provides distortion in the pseudorotaxane structure to facilitate twisting of the axle molecule. A mixed ferrocene and ruthenocene pseudorotaxane crystal is applied to photomechanical conversion under 405?nm laser irradiation at 85?°C and provides a lifting force 6,400-times the weight of the crystal itself upon phase transition.

Project description:The first stable base-free terminal uranium phosphinidene metallocene is presented; and its structure and reactivity have been studied in detail and compared to that of the corresponding thorium derivative. Salt metathesis reaction of the methyl iodide uranium metallocene Cp'''2 U(I)Me (2, Cp'''=η5 -1,2,4-(Me3 C)3 C5 H2 ) with Mes*PHK (Mes*=2,4,6-(Me3 C)3 C6 H2 ) in THF yields the base-free terminal uranium phosphinidene metallocene, Cp'''2 U=PMes* (3). In addition, density functional theory (DFT) studies suggest substantial 5f orbital contributions to the bonding within the uranium phosphinidene [U]=PAr moiety, which results in a more covalent bonding between the [Cp'''2 U]2+ and [Mes*P]2- fragments than that for the related thorium derivative. This difference in bonding besides steric reasons causes different reactivity patterns for both molecules. Therefore, the uranium derivative 3 may act as a Cp'''2 U(II) synthon releasing the phosphinidene moiety (Mes*P:) when treated with alkynes or a variety of hetero-unsaturated molecules such as imines, thiazoles, ketazines, bipy, organic azides, diazene derivatives, ketones, and carbodiimides.

Project description:In order to explore how specific atom-to-atom replacements change the electrostatic potentials on 1,3,4-chalcogenadiazole derivatives, and to deliberately alter the balance between intermolecular interactions, four target molecules were synthesized and characterized. DFT calculations indicated that the atom-to-atom substitution of Br with I, and S with Se enhanced the σ-hole potentials, thus increasing the structure directing ability of halogen bonds and chalcogen bonds as compared to intermolecular hydrogen bonding. The delicate balance between these intermolecular forces was further underlined by the formation of two polymorphs of 5-(4-iodophenyl)-1,3,4-thiadiazol-2-amine; Form I displayed all three interactions while Form II only showed hydrogen and chalcogen bonding. The results emphasize that the deliberate alterations of the electrostatic potential on polarizable atoms can cause specific and deliberate changes to the main synthons and subsequent assemblies in the structures of this family of compounds.

Project description:The diuranium(III) compound [UN''2]2(μ-η(6):η(6)-C6H6) (N''=N(SiMe3)2) has been studied using variable, high-pressure single-crystal X-ray crystallography, and density functional theory . In this compound, the low-coordinate metal cations are coupled through π- and δ-symmetric arene overlap and show close metal-CH contacts with the flexible methyl CH groups of the sterically encumbered amido ligands. The metal-metal separation decreases with increasing pressure, but the most significant structural changes are to the close contacts between ligand CH bonds and the U centers. Although the interatomic distances are suggestive of agostic-type interactions between the U and ligand peripheral CH groups, QTAIM (quantum theory of atoms-in-molecules) computational analysis suggests that there is no such interaction at ambient pressure. However, QTAIM and NBO analyses indicate that the interaction becomes agostic at 3.2 GPa.

Project description:In this article, three organotin complexes formulated as [(Me)2Sn(H2L1)] (1), [(Ph)2Sn(H2L1)]·MeOH (2) and [(Me)2Sn(HL2)(OAc)]4(Me)2O (3) (H4L1 = bis(2-hydroxybenzaldehyde) thiocarbohydrazone and H2L2 = bis(2-acetylpyrazine) thiocarbonohydrazone) have been synthesized and structurally characterized. Growth inhibition assays indicated that both the proligands and the three complexes are capable of showing anticancer activity against the human hepatocellular carcinoma HepG2 cells with H2L2 and complex 3 showing much higher cytotoxic potential. Subsequent toxicity studies on normal QSG7701cells showed that complex 3 has the highest tumor cell selectivity, and its IC50 value on QSG7701 cells is 8.48 fold higher than that in HepG2 cells. In acute toxicity experiments, complex 3 produces a dose-dependent effect in NIH mice with a LD50 value of 17.2 mg kg-1.

Project description:The quest for alternative drugs with respect to the well-known cis-platin and its derivatives, which are still used in more than 50% of the treatment regimens for patients suffering from cancer, is highly needed. In this context, organometallic compounds, which are defined as metal complexes containing at least one direct covalent metal-carbon bond, have recently been found to be promising anticancer drug candidates. A series of new metallocene complexes with scandium, yttrium, and neodymium have been prepared and characterized. Some of these compounds show a very interesting anti-proliferative activity in triple negative breast cancer cell line (MDA.MB231) and the non-hormone sensitive prostate cancer cell line (DU145). Moreover, the interaction of some of them with biological membranes, evaluated using liposomes as bio-membrane mimetic model systems, seems to be relevant. The biological activity of these compounds, particularly those based on yttrium, already effective at low concentrations on both cancer cell lines, should be taken into account with regard to new therapeutic approaches in anticancer therapy.

Project description:Thermal or photochemical metal-centered cycloaddition reactions of azidocobaltocenium hexafluoridophosphate or azidoferrocene with (cyclooctadiene)(cyclopentadienyl)cobalt(I) afforded the first metallocenyl-substituted tetrazene cyclopentadienyl cobalt complexes together with azocobaltocenium or azoferrocene as side products. The trimetallic CpCo compounds are highly conjugated, colored, and redox-active metallo-aromatic compounds, as shown by their spectroscopic, structural, and electrochemical properties. The CpCo-tetrazenido complex with two terminally appended cobaltocene units catalyzes electrochemical proton reduction from acetic acid at a mild overpotential (0.35 V). Replacing cobaltocene with ferrocene moieties rendered the complex inactive toward catalysis.

Project description:Two dimeric supramolecular structures {MgPc(H2O)·4-methylmorpholine}2-(1) and {ZnPc(H2O)·4-methylmorpholine}2-(2) formed from two molecules of magnesium and zinc aquaphthalocyanines in 4-methylmorpholine solution were revealed by single-crystal X-ray diffraction. Coordinated H2O molecules to the metal center of respective Mg or Zn phthalocyanines oriented face-to-face to form two hydrogen bonds, one of them connecting via O-H···N-azamethine atom to form a dimeric structure in the crystals with ring-to-ring distances of 3.412(3) Å in 1 and 3.403(5) Å in 2, whereas the second hydrogen bonds link the solvent 4-methylpholine molecules to form supramolecular dimeric {MgPc(H2O)·4-methylmorpholine}2-(1) and {ZnPc(H2O)·4-methylmorpholine}2-(2) structures. The interaction of the metal center of MgPc or ZnPc with O atom of the water molecule results its displacement of ∼0.5 Å from the N4-isonodole plane of the phthalocyaninate(2-) macrocycle that adopts a saucer-shape form. To better understand the interactions leading to the existence of the dimeric structures in the crystals, theoretical calculations of dimer stabilization energy composed of two monomers (MgPcH2O and ZnPcH2O), as well as three-dimensional molecular electrostatic potentials, have been performed using the density functional theoretical method. The calculated absorption spectra of both supramolecular monomers and dimers were compared with the experimental ones in 4-methylmorpholine solution. The water axial ligation of MgPc and ZnPc and the formation of the monomeric and dimeric supramolecular structures only slightly change the energy gap between highest occupied molecular orbital and lowest unoccupied molecular orbital levels that is compared with those of the parent MgPc and ZnPc pigments.

Project description:The TetR family of transcription regulators are diverse proteins capable of sensing and responding to various structurally dissimilar antimicrobial agents. Upon detecting these agents, the regulators allow transcription of an appropriate array of resistance markers to counteract the deleterious compounds. Campylobacter jejuni CmeR is a pleiotropic regulator of multiple proteins, including the membrane-bound multidrug efflux transporter CmeABC. CmeR represses the expression of CmeABC and is induced by bile acids, which are substrates of the CmeABC tripartite pump. The multiligand-binding pocket of CmeR has been shown to be very extensive and consists of several positively charged and multiple aromatic amino acids. Here we describe the crystal structures of CmeR in complexes with the bile acids, taurocholate and cholate. Taurocholate and cholate are structurally related, differing by only the anionic charged group. However, these two ligands bind distinctly in the binding tunnel. Taurocholate spans the novel bile acid binding site adjacent to and without overlapping with the previously determined glycerol-binding site. The anionic aminoethanesulfonate group of taurocholate is neutralized by a charge-dipole interaction. Unlike taurocholate, cholate binds in an anti-parallel orientation but occupies the same bile acid-binding site. Its anionic pentanoate moiety makes a water-mediated hydrogen bond with a cationic residue to neutralize the formal negative charge. These structures underscore the promiscuity of the multifaceted binding pocket of CmeR. The capacity of CmeR to recognize bile acids was confirmed using isothermal titration calorimetry and fluorescence polarization. The results revealed that the regulator binds these acids with dissociation constants in the micromolar region.

Project description:The crystal structures of Mycobacterium smegmatis RecA (RecA(Ms)) and its complexes with ADP, ATPgammaS, and dATP show that RecA(Ms) has an expanded binding site like that in Mycobacterium tuberculosis RecA, although there are small differences between the proteins in their modes of nucleotide binding. Nucleotide binding is invariably accompanied by the movement of Gln 196, which appears to provide the trigger for transmitting the effect of nucleotide binding to the DNA-binding loops. These observations provide a framework for exploring the known properties of the RecA proteins.
