Facile Synthesis of Pd-Ir Nanocubes for Biosensing.

ABSTRACT: Displaying extremely high peroxidase-like activity and uniform cubic structure enclosed by (100) facets, Pd-Ir nanocubes are an attractive nanomaterial for bioanalysis. However, there exists a great challenge to deposit atomic layers of Ir on the surface of Pd nanocubes due to the relatively low energy barrier of homogeneous nucleation of Ir atoms compared to heterogeneous nucleation. Here, a simple and surfactant-free approach is presented to synthesize Pd-Ir nanocubes with atomic Ir shell thickness in an aqueous solution at room temperature. Biomolecules such as antibodies and nucleic acids have free access to the surface of Pd-Ir nanocubes. Applications of Pd-Ir nanocubes in immunoassays and aptamer-based biosensors are realized, exploiting the excellent peroxidase activity and fluorescence quenching ability of Pd-Ir nanocubes. This work makes a significant step forward towards the practical utility of Pd-Ir nanocubes in bioanalysis.

One-pot high-yield synthesis of Pd nanocubes for Pd-Ir nanocube-based immunoassay of nucleocapsid protein from SARS-CoV-2.

Project description:Pd-Ir nanocubes are promising peroxidase-mimicking nanozymes for immunoassays, enabled by their excellent stability, relatively high catalytic activity, and reproducible performance. A key step involved in the preparation of Pd-Ir nanocubes is the synthesis of Pd nanocubes. However, the traditional method to synthesize Pd nanocubes requires sophisticated and expensive equipment to precisely control the reaction temperature and highly skilled technicians to achieve satisfactory and reproducible product yields. Herein, we report a simple, cost-effective, high-yield (> 99%) and one-pot strategy to synthesize Pd nanocubes with sizes of 7, 18, and 51 nm for the preparation of Pd-Ir nanocubes. The resulting 18 nm Pd-Ir nanocubes display three orders of magnitude higher peroxidase activity compared to horseradish peroxidase, leading to a significantly increased detection sensitivity when applied in the immunoassay of nucleocapsid protein from SARS-CoV-2. Due to the simplicity in both material synthesis and assaying procedures and the excellent detection sensitivity, our method should allow for the generalized application of Pd-Ir nanocube-based immunoassays for the diagnosis of human diseases.

Facile Synthesis of Pd Nanocubes with Assistant of Iodide and Investigation of Their Electrocatalytic Performances Towards Formic Acid Oxidation.

Project description:This article presents a facile, one-pot method using the aqueous phase for the synthesis of high-quality Pd nanocubes. In this study, Pd chloride was used as the precursor, sodium iodide as capping agent, and poly(vinylpyrrolidone) as surfactant and reducing agent. The effects of different halogens on the morphology of Pd nanocrystals were investigated. The results showed that, in this synthesis system, the selection and proper amount of sodium iodide was essential to the preparation of high-quality Pd nanocubes. When iodide was replaced by other halogens (such as bromide and chloride), Pd nanocrystals with cubic morphology could not be obtained. In addition, we have found that NaBH₄ can be used to efficiently remove inorganic covers, such as iodide, from the surface of Pd nanoparticles as synthesized. The Pd nanoparticles obtained were employed as electro-catalysts for formic acid oxidation, and they exhibited excellent catalytic activity and good stability towards this reaction.

Fluorescent PEI@Pd nanoclusters: facile synthesis and application.

Project description:Metal nanoclusters (NCs) have recently emerged as a novel class of luminescent nanomaterials and held significant potential in analytical chemistry. In this work, novel polyethyleneimine stabilized palladium nanoclusters (PEI-Pd NCs) were synthesized by chemical reduction at 60 °C for 6 h, and used as a fluorescent nanosensor for the detection of oxytetracycline (OTC). The spectral characteristics, surface structure and morphology of the Pd NCs were studied. The selectivity and stability of the nanosensor were also investigated. The experimental results showed that the Pd NCs had good biocompatibility, stability and photobleaching resistance in aqueous solution. The fluorescence quenching effect showed a good linear relationship with the degree of fluorescence quenching of Pd NCs and OTC in the range of 25-440 nM, with a correlation coefficient of 0.99. The limit of detection (LOD) of the proposed nanosensor for OTC was calculated to be 22 nM. The mechanism of determination was thought to be an inner filter effect (IFE) between OTC and Pd NCs. Based on this, we have established a new nanosensing analysis method for detecting OTC.

Facile Controlled Synthesis of Pd-ZnO Nanostructures for Nitrite Detection.

Project description:The electrocatalytic characteristics of nanostructures are significantly affected by surface structure. The strict regulation of structural characteristics is highly beneficial for the creation of novel nanocatalysts with enhanced electrocatalytic performance. This work reports a nitrite electrochemical sensor based on novel flower-like Pd-ZnO nanostructures. The Pd-ZnO nanocatalysts were synthesized through a simple hydrothermal method, and their morphology and structure were characterized via field-emission scanning electron microscopy (FE-SEM), X-ray photoelectron spectroscopy (XPS), and X-ray diffraction (XRD). Their electrocatalytical performance in the nitrite oxidation reaction was studied via cyclic voltammetry (CV) and the amperometric technique. Compared to pure ZnO and Pd nanoparticles, the Pd-ZnO nanostructures exhibited enhanced electrochemical performance in the nitrite oxidation reaction. In order to investigate the relationships between the structures of Pd-ZnO nanocatalysts and the corresponding electrocatalytic performances, different surface morphologies of Pd-ZnO nanocatalysts were fabricated by altering the solution pH. It was found that the flower-like Pd-ZnO nanostructures possessed larger effective surface areas and faster electron transfer rates, resulting in the highest electrocatalytic performance in the nitrite oxidation reaction. The designed nitrite sensor based on flower-like Pd-ZnO displayed a wide concentration linear range of 1 μM-2350 μM, a low detection limit of 0.2 μM (S/N of 3), and high sensitivity of 151.9 μA mM-1 cm-2. Furthermore, the proposed sensor exhibited perfect selectivity, excellent reproducibility, and long-time stability, as well as good performance in real sample detection.

Facile synthesis of gold nanocages with silver nanocubes templates dual metal effects enabled SERS imaging and catalytic reduction.

Project description:Silver (Ag) nanomaterials featuring a cubic shape particularly represent supreme class of advance nanomaterials. This work explored a new precursor and its effect on morphological features of silver (Ag) nanocubes (NCs) serving as sacrificial templates for facile synthesis of gold NCs. The AgNCs were initially prepared utilizing sodium thiosulphate (Na2S2O3) as relatively stable S2- producing species along with a soft etchant source KCl. The effects of different potassium halides were evaluated to grasp control over seed mediated growth of Ag nanocubes. Taking the advantages of dual metallic properties, Ag@4MBA@AuNCs nanostructure was synthesized using 4-mercaptobenzoic acid (4MBA) as a Raman reporter molecule. This nanostructure showed 1010-times enhancement in surface enhanced Raman scattering (SERS) signal, leading to a highly sensitive imaging probe for the detection of even three breast cancer cells (MCF-7 cells) in vitro. Subsequently, the oxidative nanopeeling well accompanied by incorporation of Au/Ag alloy nanoparticles on AuNCs corona assembly was achieved, which facilitated the catalytic reduction of toxic nitrophenol to eco-friendly aminophenol. Such sophisticated and engineered nanoassemblies possess broad applications in bioanalysis.

Facile aqueous-phase synthesis of Ag-Cu-Pt-Pd quadrometallic nanoparticles.

Project description:Ag-Cu-Pt-Pd quadrometallic nanoparticles which small Pt and Pd nanoparticles were attached on the surface of AgCu Janus nanoparticles were firstly synthesized by sequential reduction of Pt and Pd precursor in the presence of Janus AgCu bimetallic nanoparticles as seeds in an aqueous solution. Even though there was a small amount of Cu2O on the surface, the synthesized nanoparticles were mainly composed of four independent metallic part, not alloy parts. By theoretical calculation and growth mechanism study, we found that different reducing rate between Ag+ and Cu2+ and sequential reduction of Pt and Pd precursors would be key roles for the formation of the quadrometallic nanoparticles.

Facile synthesis of Ag nanocubes of 30 to 70 nm in edge length with CF(3)COOAg as a precursor.

Project description:This paper describes a new protocol to synthesize Ag nanocubes of 30 to 70 nm in edge length with the use of CF(3)COOAg as a precursor to elemental silver. By adding a trace amount of NaSH and HCl to the polyol synthesis, Ag nanocubes were obtained with good quality, high reproducibility, and on a scale up to 0.19 g per batch for the 70 nm Ag nanocubes. The Ag nanocubes were found to grow in size at a controllable pace over the course of synthesis. The linear relationship between the edge length of the Ag nanocubes and the position of localized surface plasmon resonance (LSPR) peak provides a simple method for finely tuning and controlling the size of the Ag nanocubes by monitoring the UV/Vis spectra of the reaction at different times.

Facile Coating Strategy to Functionalize Inorganic Nanoparticles for Biosensing.

Project description:The use of inorganic nanoparticles (NPs) for biosensing requires that they exhibit high colloidal stability under various physiological conditions. Here, we report on a general approach to render hydrophobic NPs into hydrophilic ones that are ready for bioconjugation. The method uses peglyated polymers conjugated with multiple dopamines, which results in multidentate coordination. As proof-of-concept, we applied the coating to stabilize ferrite and lanthanide NPs synthesized by thermal decomposition. Both polymer-coated NPs showed excellent water solubility and were stable at high salt concentrations under physiological conditions. We used these NPs as molecular-sensing agents to detect exosomes and bacterial nucleic acids.

Facile synthesis of Pd@graphene nanocomposites with enhanced catalytic activity towards Suzuki coupling reaction.

Project description:A facile and chemical specific method to synthesize highly reduced graphene oxide (HRG) and Pd (HRG@Pd) nanocomposite is presented. The HRG surfaces are tailored with amine groups using 1-aminopyrene (1-AP) as functionalizing molecules. The aromatic rings of 1-AP sit on the basal planes of HRG through π-π interactions, leaving amino groups outwards (similar like self-assembled monolayer on 2D substrates). The amino groups provide the chemically specific binding sites to the Pd nucleation which subsequently grow into nanoparticles. HRG@Pd nanocomposite demonstrated both uniform distribution of Pd nanoparticles on HRG surface as well as excellent physical stability and dispersibility. The surface functionalization was confirmed using, ultraviolet-visible (UV-Vis), Fourier transform infra-red and Raman spectroscopy. The size and distribution of Pd nanoparticles on the HRG and crystallinity were confirmed using high-resolution transmission electron microscopy and powder X-ray diffraction and X-ray photoelectron spectroscopy. The catalytic efficiency of highly reduced graphene oxide-pyrene-palladium nanocomposite (HRG-Py-Pd) is tested towards the Suzuki coupling reactions of various aryl halides. The kinetics of the catalytic reaction (Suzuki coupling) using HRG-Py-Pd nanocomposite was monitored using gas chromatography (GC).

Facile synthesis of N-rich carbon quantum dots from porphyrins as efficient probes for bioimaging and biosensing in living cells.

Project description:N-rich metal-free and metal-doped carbon quantum dots (CQDs) have been prepared through one-step hydrothermal method using tetraphenylporphyrin or its transition metal (Pd or Pt) complex as precursor. The structures and morphology of the as-prepared nanoparticles were analyzed by X-ray diffraction, high-resolution transmission electron microscopy, X-ray photoelectron spectroscopy, and Fourier transform infrared spectra. Three kinds of nanocomposites show similar structures except for the presence of metal ions in Pd-CQDs and Pt-CQDs indicated by X-ray photoelectron spectroscopy. All of them display bright blue emission upon exposure to ultraviolet irradiation. The CQDs exhibit typical excitation-dependent emission behavior, with the emission quantum yield of 10.1%, 17.8%, and 15.2% for CQDs, Pd-CQDs, and Pt-CQDs, respectively. Moreover, the CQDs, Pd-CQDs, and Pt-CQDs could serve as fluorescent probes for the specific and sensitive detection of Fe3+ ions in aqueous solution. The low cytotoxicity of CQDs is demonstrated by MTT assay against HeLa cells. Therefore, the CQDs can be used as efficient probes for cellular multicolor imaging and fluorescence sensors for the detection of Fe3+ ions due to their low toxicity, excellent biocompatibility, and low detection limits. This work provides a new route to synthesize highly luminescent N-rich metal-free or metal-doped CQDs for multifunctional applications.
