Discovery of a new species of Adder's tongue fern from India with comparative analysis of morphological and molecular attributes.

ABSTRACT: Eusporangiate fern genus Ophioglossum L. is commonly known as Adder's tongue fern as its fertile frond gives the appearance of snake tongue. A new species in this fern genus, O. trilokinathii belonging to Ophioglossaceae family has been discovered from the plateau region of Rajasthan State of northwestern India. The new species can be distinguished from other taxa of this genus by its smaller habit, subglobose-tuberous rhizome, basipetal emergence of young roots, aggregation of old decaying roots on rhizome apex, fertile stalk as well as spike short and thick, trophophylls in rosette, ovate or orbicular and a unique sporoderm sculpture pattern under SEM having broad reticulations with thick and raised muri enclosing large hexagonal or irregular areas on the distal and proximal faces of the spores hitherto unreported in any of the presently known taxa of Ophioglossum. In addition, comparative study of stomatal structure, foliar anatomy and nucleotide sequence data of its three chloroplast DNA markers (trnL-F, rbcL and psbA-trnH) was carried out. In view of all the attributes including habitat, ecology, morphology, foliar anatomy, stomatal features, palynology and molecular phylogenetic data, the present study suggests that the Ophioglossum specimen collected from plateau region of Rajasthan represents a hitherto undescribed species thereby warranting its establishment as O. trilokinathii sp. nov. A detailed comparative account of the new taxon with its allied species has also been provided.

Morphological description and molecular characterisation of a new species of <i>Anilocra</i> Leach, 1818 (Crustacea: Isopoda: Cymothoidae) from India.

Project description:A fish parasitic isopod recently reported from India as Anilocra leptosoma Bleeker, 1857, was re-examined and morphologically compared to five closely related species: A. capensis Leach, 1818, A. clupei Williams and Bunkley-Williams, 1986, A. leptosoma Bleeker, 1857, A. paulsikkeli Welicky and Smit, 2019 and A. pilchardi Bariche and Trilles, 2006. This species was sequenced and compared to other known Anilocra species based on mitochondrial cytochrome c oxidase subunit I (COI) gene fragments. Both morphological and molecular data corroborate that the Anilocra species parasitising the clupeid fish Tenualosa toli (Valenciennes, 1847) from India should be recognised as a new species, and we describe Anilocra grandmaae n. sp. based on the holotype and paratype females. The key characters of A. grandmaae n. sp. include the body being less than 4.0 times as long as wide; antennula article 3 anterodistal margin expanded, 1.2-1.4 times as wide as long; pleonite 1 concealed by pereonite 7 and lateral margin posteriorly produced; pereopods 1-4 with three nodules on dactylus; endopod of pleopods 3-5 with a proximomedial lobe and folding; and pleotelson ovate, with lateral margins converging smoothly to a caudomedial point.

Adiantumshastense, a new species of maidenhair fern from California.

Project description:A new species of Adiantum is described from California. This species is endemic to northern California and is currently known only from Shasta County. We describe its discovery after first being collected over a century ago and distinguish it from Adiantumjordanii and Adiantumcapillus-veneris. It is evergreen and is sometimes, but not always, associated with limestone. The range of Adiantumshastense Huiet & A.R.Sm., sp. nov., is similar to several other Shasta County endemics that occur in the mesic forests of the Eastern Klamath Range, close to Shasta Lake, on limestone and metasedimentary substrates.

Discovery of two new species of Crotalaria (Leguminosae, Crotalarieae) from Western Ghats, India.

Project description:Two new species of Fabaceae-Papilionoideae are described and illustrated. Crotalaria suffruticosa from Karul Ghat region of Maharashtra is morphologically close to C. albida and C. epunctata. C. multibracteata from Panhala region of Maharashtra resembles C. vestita. C. suffruticosa differs from C. albida and C. epunctata in its habit, leaf, inflorescence, callosity, keel type, stigma, style morphology and number of seeds/pod. To test if the new species differ from their morphologically most similar species, we measured various traits and performed a Principal Component Analysis (PCA). This analysis shows that the new species differs from similar species in gross morphology for several diagnostic traits and showed correlations between the variables or distance among groups and estimated the contribution of each character. Phylogenetic analyses were also conducted based on nuclear (ITS) and plastid (matK) markers. The analyses revealed nucleotide differences between the new species and their close allies attributing to their distinctiveness. A map and key including all species of Crotalaria from Maharashtra state are provided. Conservation status of the two new species have also been assessed.

Comparative Morphological, Ultrastructural, and Molecular Studies of Four Cicadinae Species Using Exuvial Legs.

Project description:Previous studies have suggested that exuviae can be used for the identification of cicada species, but the precise characteristics that differ among species have not been determined. Thus, we performed the first comparative analyses of the leg morphology, ultrastructure, and mitochondrial DNA sequences of exuviae of four dominant cicada species in Korea, Hyalessa maculaticollis (Motschulsky, 1866), Meimuna opalifera (Walker, 1850), Platypleura kaempferi (Fabricius, 1794) and Cryptotympana atrata (Fabricius, 1775), the source of Cicadidae Periostracum, a well-known traditional medicine. A morphological analysis revealed that the profemur length, femoral tooth angle, and distance between the intermediate and last tooth of the femoral comb are useful characteristics for identification. We also evaluated the usefulness of the size, degree of reflex, and number of spines on the mid-legs and hind legs as diagnostic features. An ultrastructural study showed that Meimuna opalifera has a unique surface pattern on the legs. The sequences obtained using exuviae were identical to previously obtained sequences for adult tissues. Moreover, in a phylogenetic analysis using CO1 sequences, each species formed a monophyletic cluster with high bootstrap support. Accordingly, multiple methodological approaches using exuviae might provide highly reliable identification tools. The integrative data provide useful characteristics for the exuviae-based identification of closely related species and for further taxonomic and systematic studies of Cicadinae.

A comparative assessment of morphological and molecular diversity among Sechium edule (Jacq.) Sw. accessions in India.

Project description:Genetic variation of 36 Sechium edule accessions collected across 12 states in India was assessed using morphological traits and DAMD markers. Eighteen fruit morphological traits (both qualitative and quantitative) were evaluated to confirm the variations in the present collection. Quantitative traits showed major variations with respect to fruit weight (7.85-498.33 g/fruit), fruit length (5.8-15 cm/fruit), fruit diameter (6-28 cm/fruit) and length of the spine (0-5 cm). Qualitative traits were also diverse in fruit colour, shape, spine density, reticulation, flexibility of spine and furrow depth. The first six principle components showed 82.88% variation in the principal component analysis. The principal component analysis revealed that fruit weight, fruit width, fruit diameter, fruit shape, length of spine, spine density and furrow depth had a significant contribution to the total variation. The DNA analysis performed using DAMD primers were used for deducing the diversity at DNA level. The collection produced 102 bands out of which 97 were polymorphic and the percentage polymorphism ranged between 66.66 and 100 per primer. Discrete pattern of clustering was obtained using UPGMA method of complete linkage percent disagreement revealing high diversity among the collected accessions. Thus, the present study indicates that molecular and morphological marker map would improve our knowledge of S. edule and would facilitate efforts to breed improved S. edule cultivars.

Genetic and morphological identification of a recurrent Dicksonia tree fern hybrid in New Zealand.

Project description:Hybridization is common in many ferns and has been a significant factor in fern evolution and speciation. However, hybrids are rare between the approximately 30 species of Dicksonia tree ferns world-wide, and none are well documented. In this study we examine the relationship of a newly-discovered Dicksonia tree fern from Whirinaki, New Zealand, which does not fit the current taxonomy of the three species currently recognized in New Zealand. Our microsatellite genotyping and ddRAD-seq data indicate these plants are F1 hybrids that have formed multiple times between D. fibrosa and D. lanata subsp. lanata. The Whirinaki plants have intermediate morphology between D. fibrosa and D. lanata subsp. lanata and their malformed spores are consistent with a hybrid origin. The Whirinaki plants-Dicksonia fibrosa × D. lanata subsp. lanata-are an example of hybridization between distantly related fern lineages, with the two parent species estimated to have diverged 55-25 mya. Our chloroplast sequencing indicates asymmetric chloroplast inheritance in the Whirinaki morphology with D. lanata subsp. lanata always contributing the chloroplast genome.

Corroborating molecular species discovery: Four new pine-feeding species of Chionaspis (Hemiptera, Diaspididae).

Project description:The genus Chionaspis (Hemiptera, Diaspididae) includes two North American species of armored scale insects feeding on Pinaceae: Chionaspis heterophyllae Cooley, and Chionaspis pinifoliae (Fitch). Despite the economic impact of conifer-feeding Chionaspis on horticulture, the species diversity in this group has only recently been systematically investigated using samples from across the group's geographic and host range. This paper provides morphological recognition characters for four new species that were recently hypothesized to exist on the basis of molecular evidence. The new species, here described, are Chionaspis brachycephalon Vea sp. n., Chionaspis caudata Vea sp. n., Chionaspis sonorae Vea sp. n. and Chionaspis torreyanae Vea sp. n. One of the new species, Chionaspis caudata Vea, has a gland spine at the apex of the pygidium, between the median lobes, unlike any other species of Chionaspis. An identification key to the species of Chionaspis feeding on pine in North America is provided.

Three new species of EntolomasubgenusPouzarella from China based on morphological and molecular data.

Project description:In the present paper, three additional species of EntolomasubgenusPouzarella viz. E.erectoides, E.griseocarpum and E.rubropilosum are described from China. E.rubropilosum is a typical species in section Pouzarella; E.griseocarpum and E.erectoides are members of sect. Dysthales. The taxa are further confirmed by ITS, RPB2, LSU and mtSSU analyses and phylogenetic relationships with other Entolomasubgen.Pouzarella species are also discussed. ITS sequence analysis showed that the sizes of the entire ITS region and ITS1 are remarkably divergent, while the ITS2 is conserved in length within Entolomasubgen.Pouzarella. Molecular analyses, based on the combined dataset, demonstrated that species diversity of subgen.Pouzarella in China is much higher than previously thought, in the present study twenty phylogenetic species from China are taken into consideration. On the other hand, morphological and molecular analyses suggested that classification of Entolomasubgen.Pouzarella probably has to be fundamentally re-adjusted based on additional data.

Two New Species of Liuixalus (Rhacophoridae, Anura): Evidence from Morphological and Molecular Analyses.

Project description:Due to small body sizes, superficial similarities in morphologies, and obscure activity behaviors, the phylogeny and taxonomy of species in the genus Liuixalus were very troublesome. Some species might comprise a complex of cryptic species. To investigate the species of group, we constructed the matrilineal genealogy of the genus using 16s rRNA mitochondrial DNA sequences. Analyses recovered six well supported matrilines that involved L. romeri, L. ocellatus, L. hainanus, L. calcarius, Liuixalus shiwandashan sp. nov. and Liuixalus jinxiuensis sp. nov., though the historical relationships among them remained unresolved. Currently, Liuixalus included 4 species, distributed eastwards from northern Vietnam to Hong Kong, China. Based on genealogical and morphological distinctiveness, we described Liuixalus jinxiuensis sp. nov. from the type locality Mt. Dayao, Jinxiu, China and Liuixalus shiwandashan sp. nov. from the type locality Mt. Shiwanda, China. A combination of morphological measurements, genetic, bioacoustic and osteological analysis was served to diagnose the new taxa.
