How production networks amplify economic growth.

ABSTRACT: Technological improvement is the most important cause of long-term economic growth. In standard growth models, technology is treated in the aggregate, but an economy can also be viewed as a network in which producers buy goods, convert them to new goods, and sell the production to households or other producers. We develop predictions for how this network amplifies the effects of technological improvements as they propagate along chains of production, showing that longer production chains for an industry bias it toward faster price reduction and that longer production chains for a country bias it toward faster growth. These predictions are in good agreement with data from the World Input Output Database and improve with the passage of time. The results show that production chains play a major role in shaping the long-term evolution of prices, output growth, and structural change.

Systemic acquired resistance networks amplify airborne defense cues.

Project description:Salicylic acid (SA)-mediated innate immune responses are activated in plants perceiving volatile monoterpenes. Here, we show that monoterpene-associated responses are propagated in feed-forward loops involving the systemic acquired resistance (SAR) signaling components pipecolic acid, glycerol-3-phosphate, and LEGUME LECTIN-LIKE PROTEIN1 (LLP1). In this cascade, LLP1 forms a key regulatory unit in both within-plant and between-plant propagation of immunity. The data integrate molecular components of SAR into systemic signaling networks that are separate from conventional, SA-associated innate immune mechanisms. These networks are central to plant-to-plant propagation of immunity, potentially raising SAR to the population level. In this process, monoterpenes act as microbe-inducible plant volatiles, which as part of plant-derived volatile blends have the potential to promote the generation of a wave of innate immune signaling within canopies or plant stands. Hence, plant-to-plant propagation of SAR holds significant potential to fortify future durable crop protection strategies following a single volatile trigger.

Global supply chains amplify economic costs of future extreme heat risk.

Project description:Evidence shows a continuing increase in the frequency and severity of global heatwaves1,2, raising concerns about the future impacts of climate change and the associated socioeconomic costs3,4. Here we develop a disaster footprint analytical framework by integrating climate, epidemiological and hybrid input-output and computable general equilibrium global trade models to estimate the midcentury socioeconomic impacts of heat stress. We consider health costs related to heat exposure, the value of heat-induced labour productivity loss and indirect losses due to economic disruptions cascading through supply chains. Here we show that the global annual incremental gross domestic product loss increases exponentially from 0.03 ± 0.01 (SSP 245)-0.05 ± 0.03 (SSP 585) percentage points during 2030-2040 to 0.05 ± 0.01-0.15 ± 0.04 percentage points during 2050-2060. By 2060, the expected global economic losses reach a total of 0.6-4.6% with losses attributed to health loss (37-45%), labour productivity loss (18-37%) and indirect loss (12-43%) under different shared socioeconomic pathways. Small- and medium-sized developing countries suffer disproportionately from higher health loss in South-Central Africa (2.1 to 4.0 times above global average) and labour productivity loss in West Africa and Southeast Asia (2.0-3.3 times above global average). The supply-chain disruption effects are much more widespread with strong hit to those manufacturing-heavy countries such as China and the USA, leading to soaring economic losses of 2.7 ± 0.7% and 1.8 ± 0.5%, respectively.

Systems-level interactions between insulin-EGF networks amplify mitogenic signaling.

Project description:Crosstalk mechanisms have not been studied as thoroughly as individual signaling pathways. We exploit experimental and computational approaches to reveal how a concordant interplay between the insulin and epidermal growth factor (EGF) signaling networks can potentiate mitogenic signaling. In HEK293 cells, insulin is a poor activator of the Ras/ERK (extracellular signal-regulated kinase) cascade, yet it enhances ERK activation by low EGF doses. We find that major crosstalk mechanisms that amplify ERK signaling are localized upstream of Ras and at the Ras/Raf level. Computational modeling unveils how critical network nodes, the adaptor proteins GAB1 and insulin receptor substrate (IRS), Src kinase, and phosphatase SHP2, convert insulin-induced increase in the phosphatidylinositol-3,4,5-triphosphate (PIP(3)) concentration into enhanced Ras/ERK activity. The model predicts and experiments confirm that insulin-induced amplification of mitogenic signaling is abolished by disrupting PIP(3)-mediated positive feedback via GAB1 and IRS. We demonstrate that GAB1 behaves as a non-linear amplifier of mitogenic responses and insulin endows EGF signaling with robustness to GAB1 suppression. Our results show the feasibility of using computational models to identify key target combinations and predict complex cellular responses to a mixture of external cues.

Platelets amplify inflammation in arthritis via collagen-dependent microparticle production.

Project description:In addition to their pivotal role in thrombosis and wound repair, platelets participate in inflammatory responses. We investigated the role of platelets in the autoimmune disease rheumatoid arthritis. We identified platelet microparticles--submicrometer vesicles elaborated by activated platelets--in joint fluid from patients with rheumatoid arthritis and other forms of inflammatory arthritis, but not in joint fluid from patients with osteoarthritis. Platelet microparticles were proinflammatory, eliciting cytokine responses from synovial fibroblasts via interleukin-1. Consistent with these findings, depletion of platelets attenuated murine inflammatory arthritis. Using both pharmacologic and genetic approaches, we identified the collagen receptor glycoprotein VI as a key trigger for platelet microparticle generation in arthritis pathophysiology. Thus, these findings demonstrate a previously unappreciated role for platelets and their activation-induced microparticles in inflammatory joint diseases.

Secretory MPP3 reinforce myeloid differentiation trajectory and amplify myeloid cell production.

Project description:Hematopoietic stem cells (HSC) and downstream lineage-biased multipotent progenitors (MPP) tailor blood production and control myelopoiesis on demand. Recent lineage tracing analyses revealed MPPs to be major functional contributors to steady-state hematopoiesis. However, we still lack a precise resolution of myeloid differentiation trajectories and cellular heterogeneity in the MPP compartment. Here, we found that myeloid-biased MPP3 are functionally and molecularly heterogeneous, with a distinct subset of myeloid-primed secretory cells with high endoplasmic reticulum (ER) volume and FcγR expression. We show that FcγR+/ERhigh MPP3 are a transitional population serving as a reservoir for rapid production of granulocyte/macrophage progenitors (GMP), which directly amplify myelopoiesis through inflammation-triggered secretion of cytokines in the local bone marrow (BM) microenvironment. Our results identify a novel regulatory function for a secretory MPP3 subset that controls myeloid differentiation through lineage-priming and cytokine production and acts as a self-reinforcing amplification compartment in inflammatory stress and disease conditions.

Past world economic production constrains current energy demands: Persistent scaling with implications for economic growth and climate change mitigation.

Project description:Climate change has become intertwined with the global economy. Here, we describe the contribution of inertia to future trends. Drawing from thermodynamic principles, and using 38 years of available statistics between 1980 to 2017, we find a constant scaling between current rates of world primary energy consumption [Formula: see text] and the historical time integral W of past world inflation-adjusted economic production Y, or [Formula: see text]. In each year, over a period during which both [Formula: see text] and W more than doubled, the ratio of the two remained nearly unchanged, that is [Formula: see text] Gigawatts per trillion 2010 US dollars. What this near constant implies is that current growth trends in energy consumption, population, and standard of living, perhaps counterintuitively, are determined by past innovations that have improved the economic production efficiency, or enabled use of less energy to transform raw materials into the makeup of civilization. Current observed growth rates agree well with predictions derived from available historical data. Future efforts to stabilize carbon dioxide emissions are likely also to be constrained by the contributions of past innovation to growth. Assuming no further efficiency gains, options look limited to rapid decarbonization of energy consumption through sustained implementation of at least one Gigawatt of renewable or nuclear power capacity per day. Alternatively, with continued reliance on fossil fuels, civilization could shift to a steady-state economy, one that devotes economic production exclusively to maintining ongoing metabolic needs rather than to material expansion. Even if such actions could be achieved immediately, energy consumption would continue at its current level, and atmospheric carbon dioxide concentrations would only begin to balance natural sinks at concentrations exceeding 500 ppmv.

Performance analysis of relay subset selection for amplify-and-forward cognitive relay networks.

Project description:Cooperative communication is regarded as a key technology in wireless networks, including cognitive radio networks (CRNs), which increases the diversity order of the signal to combat the unfavorable effects of the fading channels, by allowing distributed terminals to collaborate through sophisticated signal processing. Underlay CRNs have strict interference constraints towards the secondary users (SUs) active in the frequency band of the primary users (PUs), which limits their transmit power and their coverage area. Relay selection offers a potential solution to the challenges faced by underlay networks, by selecting either single best relay or a subset of potential relay set under different design requirements and assumptions. The best relay selection schemes proposed in the literature for amplify-and-forward (AF) based underlay cognitive relay networks have been very well studied in terms of outage probability (OP) and bit error rate (BER), which is deficient in multiple relay selection schemes. The novelty of this work is to study the outage behavior of multiple relay selection in the underlay CRN and derive the closed-form expressions for the OP and BER through cumulative distribution function (CDF) of the SNR received at the destination. The effectiveness of relay subset selection is shown through simulation results.
