Airborne bacterial community associated with fine particulate matter (PM_2.5) under different air quality indices in Temuco city, southern Chile.

ABSTRACT: Temuco (Chile) is one of the most polluted cities in Chile and Latin America. Although the fine fraction of particulate matter (PM_2.5) has been extensively studied and monitored due to its negative impact on public health, its microbiological components remain unknown. We explored, the airborne bacterial community in PM_2.5 under good, moderate, alert, pre-emergency and emergency indices of air quality (AQIs) established by the Chilean government. Bacterial community relationship with environmental factors (PM_2.5, PM₁₀, carbon monoxide, among others), was also evaluated. Significant differences in PM_2.5 bacterial community composition associated with AQIs were revealed, using 16S rRNA target sequences of denaturing gradient gel electrophoresis (DGGE) bands. Bacterial communities in PM_2.5 were mainly clustered (80%) into emergency and pre-emergency samples. The dominant phylum was Proteobacteria and most abundant genus was Novosphingobium, traditionally related to opportunistic respiratory diseases. The main factors associated with community structure were PM_2.5, PM₁₀ and carbon monoxide concentrations. This study exposed that bacterial community composition in Temuco varies according to AQIs, with the occurrence of potential opportunistic bacteria on heavily polluted days.

Project description:BackgroundThe Metropolitan Area of São Paulo has a unique composition of atmospheric pollutants, and positive correlations between exposure and the risk of diseases and mortality have been observed. Here we assessed the effects of ambient fine particulate matter (PM2.5) on genotoxic and global DNA methylation and hydroxymethylation changes, as well as the activities of antioxidant enzymes, in tissues of AJ mice exposed whole body to ambient air enriched in PM2.5, which was concentrated in a chamber near an avenue of intense traffic in São Paulo City, Brazil.ResultsMice exposed to concentrated ambient PM2.5 (1 h daily, 3 months) were compared to in situ ambient air exposed mice as the study control. The concentrated PM2.5 exposed group presented increased levels of the oxidized nucleoside 8-oxo-7,8-dihydro-2'-deoxyguanosine in lung and kidney DNA and increased levels of the etheno adducts 1,N6-etheno-2'-deoxyadenosine and 1,N2-etheno-2'-deoxyguanosine in kidney and liver DNA, respectively. Apart from the genotoxic effects, the exposure to PM2.5 led to decreased levels of the epigenetic mark 5-hydroxymethylcytosine (5-hmC) in lung and liver DNA. Changes in lung, liver, and erythrocyte antioxidant enzyme activities were also observed. Decreased glutathione reductase and increased superoxide dismutase (SOD) activities were observed in the lungs, while the liver presented increased glutathione S-transferase and decreased SOD activities. An increase in SOD activity was also observed in erythrocytes. These changes are consistent with the induction of local and systemic oxidative stress.ConclusionsMice exposed daily to PM2.5 at a concentration that mimics 24-h exposure to the mean concentration found in ambient air presented, after 3 months, increased levels of DNA lesions related to the occurrence of oxidative stress in the lungs, liver, and kidney, in parallel to decreased global levels of 5-hmC in lung and liver DNA. Genetic and epigenetic alterations induced by pollutants may affect the genes committed to cell cycle control, apoptosis, and cell differentiation, increasing the chance of cancer development, which merits further investigation.

Project description:Fine particulate matter is considered to be the most significant ambient air pollutant in terms of potential health impacts. Therefore, it is important that regulators are able to accurately assess the exposure of populations to PM10 and PM2.5 across municipal areas. We report on the practicalities of using a laser light scattering portable particulate monitor (Turnkey Instruments DustMate), in combination with a GPS, to map PM10 and PM2.5 concentrations on city-wide scales in Newcastle upon Tyne/Gateshead (UK), during a series of walking surveys. A heated inlet is necessary to remove moisture droplets from the sampled air prior to analysis by the instrument, though this also results in the loss of volatile particulate components, particularly from the PM2.5 fraction. A co-location calibration study was carried out with a reference urban background Tapered Element Oscillating Micro-Balance/Filter Dynamics Measuring System (TEOM-FDMS) system in Newcastle that is part of the UK's Automatic Urban and Rural Network (AURN) of air quality monitoring stations. For PM10, orthogonal regression of the DustMate against TEOM-FDMS data gave a slope and intercept of 1.02 ± 0.06 and -3.7 ± 1.2, respectively (R2 = 0.73), whereas for PM2.5, the respective values were 0.78 ± 0.06 and -0.63 ± 0.55 (R2 = 0.79). These parameters are comparable to literature calibration studies using this technology. There was good agreement between simultaneous samples taken using two DustMate instruments: for PM10, a slope and intercept of 1.05 ± 0.03 and 0.36 ± 0.5, respectively (R2 = 0.73), were obtained, whereas for the PM2.5, the respective values were 0.79 ± 0.01 and 0.19 ± 0.06 (R2 = 0.86). Correction factors based on the slope and intercepts obtained from the calibration exercise were applied to raw data collected from the DustMate. An annually-normalised correction procedure was then used to account for different background particulate concentrations on different sampling days. These corrected PM10 and PM2.5 concentrations and corresponding GPS coordinates were displayed on a base map using Google Fusion Tables and Google Earth Professional. Almost all areas surveyed in Newcastle/Gateshead were well below the EU Air Quality Standards for PM10 and PM2.5.
