Pyrrolysyl-tRNA synthetase (PylRS) is frequently used for site-specific incorporation of noncanonical amino acids (ncAAs) into proteins. Recently, the active site of <i>Methanomethylophilus alvus</i> PylRS (<i>Ma</i>PylRS) has been rationally engineered to expand its substrate compatibility, enabling the incorporation of difficult ncAAs.
