Mechanisms underlying inactivation of SARS-CoV-2 by nano-sized electrostatic atomized water particles.

ABSTRACT: The coronavirus disease 2019 (COVID-19) pandemic caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is a serious global issue. To prevent viral transmission, it is important to disinfect contaminated environmental surfaces and aerosols. We previously demonstrated that nano-sized electrostatic atomized water particles (NEAWPs) inactivate SARS-CoV-2. Herein, we focused on the underlying mechanisms. Morphological observation by transmission electron microscopy revealed that compared with NEAWPs-untreated virus, the shapes of particles corresponding to the size of SARS-CoV-2 particles were distorted significantly when exposed to NEAWPs. The amounts of viral RNA and protein in NEAWPs-treated SARS-CoV-2 showed a significantly greater decline than those in viruses unexposed to NEAWPs. Furthermore, much less NEAWPs-treated SARS-CoV-2 than NEAWPs-untreated virus bound to host cells. These results strongly suggest that NEAWPs damage the viral envelope, as well as viral protein and RNA, thereby impairing the ability of the virus to bind to host cells. Reactive oxygen species in NEAWPs may be involved in the inactivating effects on SARS-CoV-2.

Project description:The postharvest quality of muskmelon can be affected by fruit rot caused by the fungus Fusarium incarnatum, resulting in loss of quality. The utilization of electrostatic atomized water particles (EAWPs) in agriculture applications has been shown to induce disease resistance in plants. Therefore, in this study, we determined the effect of electrostatic atomized water particles (EAWPs) on the disease resistance of muskmelon fruits against postharvest fruit rot caused by F. incarnatum. EAWPs were applied to muskmelon fruits for 0, 30, 60, and 90 min. EAWP-treated muskmelon fruits were inoculated with F. incarnatum, and disease progress was measured. Quantitative reverse-transcription polymerase chain reaction (qRT-PCR) of the chitinase (CmCHI) and β-1,3-glucanase (CmGLU) genes of Cucumis melo (muskmelon) was performed for EAWP-treated and -untreated muskmelon fruits. The activities of cell-wall-degrading enzymes (CWDEs), chitinase, and β-1,3-glucanase were also assayed in EAWP-treated and -untreated muskmelon fruits. The results showed that disease progress was limited by EAWP treatment for 30 min prior to pathogen inoculation. Muskmelon fruits treated with EAWPs for 30 min showed an upregulation of CWDE genes, CmCHI and CmGLU, as observed by qRT-PCR, leading to high chitinase and β-1,3-glucanase activities, as observed through enzyme assays. The results of SEM microscopy revealed that the effect of the crude enzymes of EAWP-treated muskmelon caused morphological changes in F. incarnatum mycelia. Furthermore, treatment with EAWPs preserved postharvest quality in muskmelon, including with regard to texture stiffness and total chlorophyll contents, compared to untreated muskmelon. These results demonstrate that the pretreatment of muskmelon with EAWPs suppresses the development of F. incarnatum in the early stage of infection by regulating gene expression of CWDEs and elevating the activities of CWDEs, while also maintaining postharvest muskmelon quality.

Project description:In the later stage of natural gas reservoir exploration, the wellbore pressure is reduced and the liquid accumulation is serious, in order to solve the problem of liquid accumulation and low production in low-pressure and low-yield gas wells, the supersonic atomization drainage gas recovery technology is used to improve the recovery rate. By studying the influence of working condition parameters of downhole nozzle atomization drainage gas recovery on atomization effect and liquid carrying rate, a new physical model of atomization nozzle is established, the back propagation (BP) neural network atomization model and BP neural network atomization model optimized by genetic algorithm (GA) is established, and the Matlab is used to train the 45 groups of data sets before the experiment. After the model training, the normalized atomization parameters are trained for sensitivity analysis. The relationship between the strength and weakness of the factors affecting Sotel's average droplet particle size (SMD) is as follows: gas flow (Qg) > liquid inlet diameter (d) > liquid phase flow (Ql). The last 15 sets of data sets outside the training samples were tested by BP model and BP neural model optimized by genetic algorithm (GA-BP), and the size of SMD was predicted. The experimental results show that the determination coefficient R2 of the established GA-BP network model to the experimental parameters is 0.979 and the goodness of fit is high; the mean square error (MSE), mean absolute error (MAE) and mean absolute percentage error (MAPE) of the predicted value of GA-BP atomization model and the experimental value are 4.471, 1.811 and 0.031 respectively, the error is small, the prediction accuracy is high, and the establishment of the model is accurate. The GA-BP model can efficiently predict SMD under different operating conditions, at present, the new supersonic atomizing nozzle has been successfully applied to the Xushen gas field block of Daqing Oilfield, which can improve the recovery rate of natural gas by 4.5-8.6%, alleviate the problem of effusion near the end of oil exploration, and has certain guiding significance for solving the problem of wellbore effusion and improving production efficiency.

Project description:BackgroundNanoparticle (NP) toxicity testing comes with many challenges. Characterization of the test substance is of crucial importance and in the case of NPs, agglomeration/aggregation state in physiological media needs to be considered. In this study, we have addressed the effect of agglomerated versus single particle suspensions of nano- and submicron sized gold on the inflammatory response in the lung. Rats were exposed to a single dose of 1.6 mg/kg body weight (bw) of spherical gold particles with geometric diameters of 50 nm or 250 nm diluted either by ultrapure water or by adding phosphate buffered saline (PBS). A single dose of 1.6 mg/kg bw DQ12 quartz was used as a positive control for pulmonary inflammation. Extensive characterization of the particle suspensions has been performed by determining the zetapotential, pH, gold concentration and particle size distribution. Primary particle size and particle purity has been verified using transmission electron microscopy (TEM) techniques. Pulmonary inflammation (total cell number, differential cell count and pro-inflammatory cytokines), cell damage (total protein and albumin) and cytotoxicity (alkaline phosphatase and lactate dehydrogenase) were determined in bronchoalveolar lavage fluid (BALF) and acute systemic effects in blood (total cell number, differential cell counts, fibrinogen and C-reactive protein) 3 and 24 hours post exposure. Uptake of gold particles in alveolar macrophages has been determined by TEM.ResultsParticles diluted in ultrapure water are well dispersed, while agglomerates are formed when diluting in PBS. The particle size of the 50 nm particles was confirmed, while the 250 nm particles appear to be 200 nm using tracking analysis and 210 nm using TEM. No major differences in pulmonary and systemic toxicity markers were observed after instillation of agglomerated versus single gold particles of different sizes. Both agglomerated as well as single nanoparticles were taken up by macrophages.ConclusionPrimary particle size, gold concentration and particle purity are important features to check, since these characteristics may deviate from the manufacturer's description. Suspensions of well dispersed 50 nm and 250 nm particles as well as their agglomerates produced very mild pulmonary inflammation at the same mass based dose. We conclude that single 50 nm gold particles do not pose a greater acute hazard than their agglomerates or slightly larger gold particles when using pulmonary inflammation as a marker for toxicity.
