Project description:In this study we investigate the transcriptomic response of Escherichia coli to CdTe-2.4 and benign CdSe-2.4 quantum dots, each with and without illumination to elucidate the phototherapeutic effect of CdTe-2.4. Our analysis sought to separate the transcriptomic responses of E. coli to the presence of superoxide and the presence of cadmium chalcogenide nanoparticles. We found eight genes to be consistently differentially expressed as a response to superoxide generation, and these genes demonstrate a consistent association with the DNA damage response and deactivation of iron-sulfur clusters, characteristic of a superoxide response. We found eighteen genes associated the presence of cadmium-based quantum dots, in isolation from the superoxide effect. In further analysis of these genes, we performed both amino acid supplementation and gene knockout experiments, identifying the importance of leucyl-tRNA downregulation as a cadmium-based QD response, as well as reinforcing the relationship between CdTe-2.4 stress and iron-sulfur clusters through the gene tusA. This study demonstrates the transcriptomic response of E. coli to CdTe-2.4 and CdSe-2.4 quantum dots and parses the different effects of superoxide versus material effects on the bacteria. Our findings may provide useful information towards the development of quantum dot-based antibacterial therapy in the future.

Project description:Hybrid nanoarchitectures of AgInS2 and TiO2 photocatalysts were prepared by using a modified sol-gel method. The experimental results reveal that these nanocomposites display enhanced visible light absorption and effective charge carrier separation compared to their pristine parent samples (AgInS2 or TiO2). 0.5 wt % AgInS2 loading was found to be the optimum concentration for photocatalytic applications. More than 95% of doxycycline degradation was achieved within 180 min of solar light illumination. Similarly, the dopant concentrations at lower values (<2 wt %) exhibited 300 times higher H2 generation rate under visible light irradiation compared to AgInS2 and TiO2. The microbial strains (Escherichia coli and Staphylococcus aureus) exhibited a 99.999% reduction within half an hour of simulated solar light illumination. The computational investigation was employed to understand the structural, electronic, and the dielectric properties of AgInS2 and TiO2 composites. The improved photocatalytic results are explained as a result of the decreased rate of exciton recombination. The current investigation opens up new insights into the use of novel ternary heterostructure nanocomposites for improved visible light activity.

Project description:Next-generation sequencing (NGS) technology has revolutionized sequence-based research. In recent years, high-throughput sequencing has become the method of choice in studying the toxicity of chemical agents through observing and measuring changes in transcript levels. Engineered nanomaterial (ENM)-toxicity has become a major field of research and has adopted microarray and newer RNA-Seq methods. Recently, nanotechnology has become a promising tool in the diagnosis and treatment of several diseases in humans. However, due to their high stability, they are likely capable of remaining in the body and environment for long periods of time. Their mechanisms of toxicity and long-lasting effects on our health is still poorly understood. This review explores the effects of three ENMs including carbon nanotubes (CNTs), quantum dots (QDs), and Ag nanoparticles (AgNPs) by cross examining publications on transcriptomic changes induced by these nanomaterials.

Project description:The chemical species (ligands) at the surface of colloidal inorganic semiconductor nanocrystals (QDs) mediate their interactions with the surroundings. The solvation of the QDs reflects a subtle interplay between ligand-solvent and ligand-ligand interactions, which eventually compete with the coordination of the ligands at the QD surface. The QD surface coordination and solvation are indeed fundamental to preserve their optoelectronic properties and to foster the effective application of QD-based inks and nanocomposites. Here we investigate such ligand interactions by exploiting diffusion ordered NMR spectroscopy (DOSY), which is suggested as an essential complement to spectral line width analysis. To this end, we use colloidal metal chalcogenide (CdS, CdSe, and PbS) QDs with (metal-)oleate ligands at their surface in several solvents exhibiting different viscosities and polarities. We demonstrate that the ligand shell is dynamically bound to the metal chalcogenide QDs, and is thus in equilibrium between the QD surface and the surrounding solvent. Such dynamic equilibria depend on ligand-solvent interactions, which are more prominent in aliphatic, rather polar solvents that favor the solvation of the ligands and, as a consequence, their displacement from the QD surface. In addition, the ligand-ligand interactions, which are more relevant for larger QDs, contribute to the stabilization of the ligand bonding at the QD surface.

Project description:Single-walled carbon nanotubes (SWCNTs) are a 1D nanomaterial that shows fluorescence in the near-infrared (NIR, >800 nm). In the past, covalent chemistry was less explored to functionalize SWCNTs as it impairs NIR emission. However, certain sp3 defects (quantum defects) in the carbon lattice have emerged that preserve NIR fluorescence and even introduce a new, red-shifted emission peak. Here, we report on quantum defects, introduced using light-driven diazonium chemistry, that serve as anchor points for peptides and proteins. We show that maleimide anchors allow conjugation of cysteine-containing proteins such as a GFP-binding nanobody. In addition, an Fmoc-protected phenylalanine defect serves as a starting point for conjugation of visible fluorophores to create multicolor SWCNTs and in situ peptide synthesis directly on the nanotube. Therefore, these quantum defects are a versatile platform to tailor both the nanotube's photophysical properties as well as their surface chemistry.

Project description:CdSe(ZnS) core(shell) aerogels were prepared from the assembly of quantum dots into mesoporous colloidal networks. The sol-gel method produces inorganic particle interfaces with low resistance to electrical transport while maintaining quantum-confinement. The photoelectrochemical properties of aerogels and their composites with poly(3-hexylthiophene) are reported for the first time.

Project description:Water-soluble nanocomposites based on CdSe/ZnS quantum dots (QDs) and hydrophobic tetraphenylporphyrin (TPP) molecules passivated by chitosan (CS) have been formed. Magnetic circular dichroism (MCD) spectra evidence TPP presence in both monomeric and agglomerated forms in the nanocomposites. The nanocomposites demonstrate more pronounced singlet oxygen generation compared to free TPP in CS at the same concentration due to the intracomplex Förster resonance energy transfer (FRET) with a 45% average efficiency.

Project description:Two novel ternary nanocatalysts, f-MWCNTs-CdS and f-MWCNTs-Ag(2)S were successfully constructed by covalent grafting of fourth generation (G4) hyperbranched, crosslinked poly(amidoamine) (PAMAM) to carboxylated multi-walled carbon nanotubes (MWCNTs-COOH) and subsequent deposition of CdS or Ag(2)S quantum dots (QDs). The structural transformation, surface potential, and morphology of functionalized MWCNTs (f-MWCNTs) and nanocatalysts were characterized by UV-vis spectrophotometer, Fourier transform infrared spectroscopy, powder X-ray diffraction, Raman spectroscopy, thermogravimetric analysis, scanning electron microscopy and energy dispersive spectroscopy. Transmission electron microscopy reveals the effective anchoring of QDs on f-MWCNTs. The catalytic activity of nanocatalysts was evaluated by photodegradation of methyl orange under illumination of UV light. The coupling of MWCNTs, PAMAM and CdS or Ag(2)S QDs significantly enhanced the catalytic efficiency of nanocatalysts. The rate constants for degradation of methyl orange in presence of nanocatalysts were calculated using the Langmuir-Hinshelwood model. Overall, the excellence in photodegradation was accomplished by hybridizing f-MWCNTs with CdS or Ag(2)S.

Project description:Quantum dots have innate advantages as the key component of optoelectronic devices. For white light-emitting diodes (WLEDs), the modulation of the spectrum and color of the device often involves various quantum dots of different emission wavelengths. Here, we fabricate a series of carbon quantum dots (CQDs) through a scalable acid reagent engineering strategy. The growing electron-withdrawing groups on the surface of CQDs that originated from acid reagents boost their photoluminescence wavelength red shift and raise their particle sizes, elucidating the quantum size effect. These CQDs emit bright and remarkably stable full-color fluorescence ranging from blue to red light and even white light. Full-color emissive polymer films and all types of high-color rendering index WLEDs are synthesized by mixing multiple kinds of CQDs in appropriate ratios. The universal electron-donating/withdrawing group engineering approach for synthesizing tunable emissive CQDs will facilitate the progress of carbon-based luminescent materials for manufacturing forward-looking films and devices.

Project description:The influence of carbon multi-walled nanotubes (MWCNTs) and halloysite nanotubes (HNTs) on the physical, thermal, mechanical, and electrical properties of EVA (ethylene vinyl acetate) copolymer was investigated. EVA-based nanocomposites containing MWCNTs or HNTs, as well as hybrid nanocomposites containing both nanofillers were prepared by melt blending. Scanning electron microcopy (SEM) images revealed the presence of good dispersion of both kinds of nanotubes throughout the EVA matrix. The incorporation of nanotubes into the EVA copolymer matrix did not significantly affect the crystallization behavior of the polymer. The tensile strength of EVA-based nanocomposites increased along with the increasing CNTs (carbon nanotubes) content (increased up to approximately 40% at the loading of 8 wt.%). In turn, HNTs increased to a great extent the strain at break. Mechanical cyclic tensile tests demonstrated that nanocomposites with hybrid reinforcement exhibit interesting strengthening behavior. The synergistic effect of hybrid nanofillers on the modulus at 100% and 200% elongation was visible. Moreover, along with the increase of MWCNTs content in EVA/CNTs nanocomposites, an enhancement in electrical conductivity was observed.
