3D Binary Mesocrystals from Anisotropic Nanoparticles.

ABSTRACT: Binary mesocrystals offer the combination of nanocrystal properties in an ordered superstructure. Here, we demonstrate the simultaneous self-assembly of platinum and iron oxide nanocubes into micrometer-sized 3D mesocrystals using the gas-phase diffusion technique. By the addition of minor amounts of a secondary particle type tailored to nearly identical size, shape and surface chemistry, we were able to promote a random incorporation of foreign particles into a self-assembling host lattice. The random distribution of the binary particle types on the surface and within its bulk has been visualized using advanced transmission and scanning electron microscopy techniques. The 20-40 μm sized binary mesocrystals have been further characterized through wide and small angle scattering techniques to reveal a long-range ordering on the atomic scale throughout the crystal while showing clear evidence that the material consists of individual building blocks. Through careful adjustments of the crystallization parameters, we could further obtain a reverse superstructure, where incorporated particles and host lattice switch roles.

Project description:Pseudomonas aeruginosa was used to synthesize anisotropic gold nanoparticles from the unusually reducible aryldiazonium gold (III) salt of the chemical formula [HOOC-4-C6H4N≡N]AuCl4 (abbreviated as DS-AuCl4). We investigated the effect of bacterial cell density, temperature, and pH on the AuNP synthesis. The bacterial cell density of 6.0 × 108 CFU/mL successfully reduced 0.5 mM DS-AuCl4 salt to AuNPs after incubation at 37 °C (24 h), 42 °C (24 h), and 25 °C (48 h). Transmission electron microscopy (TEM) images revealed the formation of spherical, triangle, star, hexagon, and truncated triangular morphologies for the AuNPs synthesized using P. aeruginosa bacteria. The average size of AuNPs synthesized at 25 °C (48 h), 37 °C (24 h), and 42 °C (24 h) was 39.0 ± 9.1 nm, 26.0 ± 8.1 nm, and 36.7 ± 7.7 nm, respectively. The average size of AuNPs synthesized at pH 3.7, 7.0, and 12.7 was 36.7 ± 7.7 nm, 14.7 ± 3.8 nm, and 7.3 ± 2.5 nm, respectively, with the average size decreasing at a pH of 12.7. The reduction of the DS-AuCl4 salt was confirmed using X-ray photoelectron spectroscopy (XPS). The significant peaks for C1s, Au4f doublet, N1s, and O1s are centered at 285, 84-88, 400, and 532 eV. The ability of inactivated bacteria (autoclave-dead and mechanically lysed bacteria), peptidoglycan, and lipopolysaccharides to reduce the DS-AuCl4 salt to AuNPs was also investigated. Anisotropic AuNPs were synthesized using inactivated bacteria and peptidoglycan but not using lipopolysaccharides. The AuNPs demonstrated biocompatibility with human RBCs and were safe, with no antibacterial activities against Escherichia coli and Staphylococcus aureus. This is the first report demonstrating the synthesis of AuNPs using aryldiazonium gold(III) salts with P. aeruginosa. These AuNPs are promising candidates for exploring potential applications in nanomedicine and drug delivery. Project description:BackgroundSpatial mapping of transcriptional states provides valuable biological insights into cellular functions and interactions in the context of the tissue. Accurate 3D cell segmentation is a critical step in the analysis of this data towards understanding diseases and normal development in situ. Current approaches designed to automate 3D segmentation include stitching masks along one dimension, training a 3D neural network architecture from scratch, and reconstructing a 3D volume from 2D segmentations on all dimensions. However, the applicability of existing methods is hampered by inaccurate segmentations along the non-stitching dimensions, the lack of high-quality diverse 3D training data, and inhomogeneity of image resolution along orthogonal directions due to acquisition constraints; as a result, they have not been widely used in practice.MethodsTo address these challenges, we formulate the problem of finding cell correspondence across layers with a novel optimal transport (OT) approach. We propose CellStitch, a flexible pipeline that segments cells from 3D images without requiring large amounts of 3D training data. We further extend our method to interpolate internal slices from highly anisotropic cell images to recover isotropic cell morphology.ResultsWe evaluated the performance of CellStitch through eight 3D plant microscopic datasets with diverse anisotropic levels and cell shapes. CellStitch substantially outperforms the state-of-the art methods on anisotropic images, and achieves comparable segmentation quality against competing methods in isotropic setting. We benchmarked and reported 3D segmentation results of all the methods with instance-level precision, recall and average precision (AP) metrics.ConclusionsThe proposed OT-based 3D segmentation pipeline outperformed the existing state-of-the-art methods on different datasets with nonzero anisotropy, providing high fidelity recovery of 3D cell morphology from microscopic images.
