Project description:Organic-inorganic xerogel networks were synthesized from bacterial poly (3-hydroxybutyrate) (PHB) for potential biomedical applications. Since silane-based networks usually demonstrate increased biocompatibility and mechanical properties, siloxane groups have been added onto polyurethane (PU) architectures. In this work, a diol oligomer (oligoPHB-diol) was first prepared from bacterial poly(3-hydroxybutyrate) (PHB) with an environmentally friendly method. Then, hexamethylene diisocyanate or biobased dimeryl diisocyanate was used as diisocyanate to react with the short oligoPHB-diol for the synthesis of different NCO-terminated PU systems in a bulk process and without catalyst. Various PU systems containing increasing NCO/OH molar ratios were prepared. Siloxane precursors were then obtained after reaction of the NCO-terminated PUs with (3-aminopropyl)triethoxysilane, resulting in silane-terminated polymers. These structures were confirmed by different analytical techniques. Finally, four series of xerogels were prepared via a sol-gel process from the siloxane precursors, and their properties were evaluated depending on varying parameters such as the inorganic network crosslinking density. The final xerogels exhibited adequate properties in connection with biomedical applications such as a high in vitro degradation up to 15 wt% after 12 weeks.

Project description:Glycerol (Gly) is a well-known, FDA-approved molecule posing three hydroxyl groups. Since Gly is biocompatible, here, it was aimed to prepare poly(Glycerol) (p(Gly)) particles directly for the first time for the delivery of therapeutic agents. Micrometer-sized particles of p(Gly) were successfully synthesized via the micro-emulsion method with an average size of 14.5 ± 5.6 µm. P(Gly) microparticles up to 1.0 g/mL concentrations were found biocompatible with 85 ± 1% cell viability against L929 fibroblasts. Moreover, p(Gly) microparticles were tested for hemocompatibility, and it was found that up to 1.0 mg/mL concentrations the particles were non-hemolytic with 0.4 ± 0.1% hemolysis ratios. In addition, the blood compatibility index values of the prepared p(Gly) particles were found as 95 ± 2%, indicating that these microparticles are both bio- and hemocompatible. Furthermore, Quercetin (QC) flavonoid, which possessed high antioxidant properties, was loaded into p(Gly) microparticles to demonstrate drug-carrying properties of the particles with improved bioavailability, non-toxicity, and high biocompatibility. The results of this study evidently revealed that p(Gly) particles can be directly prepared from a cost-effective and easily accessible glycerol molecule and the prepared particles exhibited good biocompatibility, hemocompatibility, and non-toxicity. Therefore, p(Gly) particles were found as promising vehicles for drug delivery systems in terms of their higher loading and release capability as well as for sustained long term release profiles.

Project description:In this paper we describe the mechanical and biological features of a thermosetting polyester synthesized from glycerol and dodecanedioic acid named Poly-Glycerol-Dodecanoate (PGD). This polymer shows a glass transition temperature (T(g)) around 32 degrees C, and this accounts for its mechanical properties. At room temperature (21 degrees ) PGD behaves like a stiff elastic-plastic material, while at body temperature (37 degrees C), it shows a compliant non-linear elastic behavior. Together with biodegradability and biocompatibility PGD has distinct shape memory features. After the polymer is cured, no matter what the final configuration is, we can recover the original shape by heating PGD to temperatures of 32 degrees C and higher. The mechanical properties together with biocompatibility/biodegradability and shape memory features make PGD an attractive polymer for biomedical applications.

Project description:Utilisation of bio-derived platform molecules in polymer synthesis has advantages which are, broadly, twofold; to digress from crude oil dependence of the polymer industry and secondly to reduce the environmental impact of the polymer synthesis through the inherent functionality of the bio-derived platform molecules. Bulk polymerisation of bio-derived unsaturated di-acids has been employed to produce unsaturated polyester (UPEs) which have been analysed by GPC, TGA, DSC and NMR spectroscopy, advancing on the analysis previously reported. UPEs from the diesters of itaconic, succinic, and fumaric acids were successfully synthesised with various diols and polyols to afford resins of MN 480-477,000 and Tg of -30.1 to -16.6 °C with solubilities differing based on starting monomers. This range of properties allows for many applications and importantly due to the surviving Michael acceptor moieties, solubility and cross-linking can be specifically tailored, post polymerisation, to the desired function. An improved synthesis of itaconate and succinate co-polymers, via the initial formation of an itaconate bis-diol, is also demonstrated for the first time, resulting in significantly improved itaconate incorporation.

Project description:Poly(glycerol sebacate) (PGS), produced from renewable monomers such as sebacic acid and glycerol, has been explored extensively for various biomedical applications. However, relatively less attention has been paid to explore PGS as sustainable materials in applications such as elastomers and rigid plastics, primarily because of serious deficiencies in physical properties of PGS. Here, we present two new approaches for enhancing the properties of PGS; (i) synthesizing block copolymers of PGS with poly(tetramethylene oxide)glycol (PTMO) and (ii) preparing a blend of PGS-b-PTMO with a poly(ester-ether) thermoplastic elastomer. The consequence of molar ratio (hard and soft segments) and M n of soft segment on tensile properties of the material was investigated. The PGS-b-PTMO with 25:75 mole ratios of hard and soft segments and having a medium M n soft segment (5350 g mol-1) exhibits an appreciable increase in percentage of elongation that is from 32% for PGS to 737%. Blends of PGS-b-PTMO and a thermoplastic polyester elastomer, Hytrel 3078, form a semi-interpenetrated polymer network, which exhibits increased tensile strength to 2.11 MPa and percentage of elongation to 2574. An elongation of such magnitude is unprecedented in the literature for predominantly aliphatic polyesters and demonstrates that the simple polyester can be tailored for superior performance.

Project description:Poly(N-isopropylacrylamide) (PNIPAM)-based thermosensitive hydrogels demonstrate great potential in biomedical applications. However, they have inherent drawbacks such as low mechanical strength, limited drug loading capacity and low biodegradability. Formulating PNIPAM with other functional components to form composited hydrogels is an effective strategy to make up for these deficiencies, which can greatly benefit their practical applications. This review seeks to provide a comprehensive observation about the PNIPAM-based composite hydrogels for biomedical applications so as to guide related research. It covers the general principles from the materials choice to the hybridization strategies as well as the performance improvement by focusing on several application areas including drug delivery, tissue engineering and wound dressing. The most effective strategies include incorporation of functional inorganic nanoparticles or self-assembled structures to give composite hydrogels and linking PNIPAM with other polymer blocks of unique properties to produce copolymeric hydrogels, which can improve the properties of the hydrogels by enhancing the mechanical strength, giving higher biocompatibility and biodegradability, introducing multi-stimuli responsibility, enabling higher drug loading capacity as well as controlled release. These aspects will be of great help for promoting the development of PNIPAM-based composite materials for biomedical applications.

Project description:Nanoparticles play an important role in biomedicine. We have developed a method for size-controlled synthesis of photomagnetic Prussian blue nanocubes (PBNCs) using superparamagnetic iron oxide nanoparticles (SPIONs) as precursors. The developed PBNCs have magnetic and optical properties desired in many biomedical diagnostic and therapeutic applications. Specifically, the size-tunable photomagnetic PBNCs exhibit high magnetic saturation, strong optical absorption with a peak at approximately 700 nm, and superior photostability. Our studies demonstrate that PBNCs can be used as MRI and photoacoustic imaging contrast agents in vivo. We also showed the utility of PBNCs for labeling and magnetic manipulation of cells. Dual magnetic and optical properties, together with excellent biocompatibility, render PBNCs an attractive contrast agent for both diagnostic and therapeutic applications. The use SPIONs as precursors for PBNCs provides flexibility and allows researchers to design theranostic agents according to required particle size, optical, and magnetic properties.

Project description:Naphthyridines, also known as diazanaphthalenes, are a group of heterocyclic compounds that include six isomeric bicyclic systems containing two pyridine rings. 1,6-Naphthyridines are one of the members of such a family capable of providing ligands for several receptors in the body. Among such structures, 1,6-naphthyridin-2(1H)-ones (7) are a subfamily that includes more than 17,000 compounds (with a single or double bond between C3 and C4) included in more than 1000 references (most of them patents). This review will cover the analysis of the diversity of the substituents present at positions N1, C3, C4, C5, C7, and C8 of 1,6-naphthyridin-2(1H)-ones, the synthetic methods used for their synthesis (both starting from a preformed pyridine or pyridone ring), and the biomedical applications of such compounds.

Project description:A novel biodegradable poly(amide-imide) (PAI) with good hydrophilicity was synthesized by incorporation of l-glycine into the polymer chain. For comparison purposes, a pure PAI containing no l-glycine was also synthesized with a three-step method. In this study, we evaluated the novel PAI's thermal stability, hydrophilicity, solubility, biodegradability and ability to support bone marrow mesenchymal stem cell (BMSC) adhesion and growth by comparing with the pure PAI. The hydrophilic tests demonstrated that the novel PAI has possible hydrophilicity at a 38° water contact angle on the molecule surface and is about two times more hydrophilic than the pure PAI. Due to an extra unit of l-glycine in the novel PAI, the average degradation rate was about 2.4 times greater than that of the pure PAI. The preliminary biocompatibility studies revealed that all the PAIs are cell compatible, but the pure PAI exhibited much lower cell adhesion than the l-glycine-incorporated novel PAI. The hydrophilic surface of the novel PAI was more suitable for cell adhesion, suggesting that the surface hydrophilicity plays an important role in enhancing cell adhesion and growth.

Project description:The manufacturing of sanitary and household furniture on a large scale with inherently antimicrobial properties is an essential field of research. This work focuses on the synthesis of polymer composites based on the unsaturated polyester of resin loaded with 5 wt.%-Triclosan produced by a co-mixing approach on automated technological complex with a potential for broad applications. According to findings, the polymer composite has a non-porous structure (surface area < 1.97 m2/g) suitable for sanitary applications to reduce the growth of bacteria. The chemical composition confirmed the presence of major elements, and the inclusion of Triclosan was quantitatively confirmed by the appearance of chlorine on XRF (1.67 wt.%) and EDS (1.62 wt.%) analysis. Thermal analysis showed the difference of 5 wt.% in weight loss, which confirms the loading of Triclosan into the polymer matrix. The polymer composite completely inhibited the strains of S. aureus 6538-P, S. aureus 39, S. epidermidis 12228, and Kl. Pneumoniae 10031 after 5-min contact time. The antimicrobial effects against Kl. pneumoniae 700603, Ps. aeruginosa 9027 and Ps. aeruginosa TA2 strains were 92.7%, 85.8% and 18.4%, respectively. The inhibition activity against C. albicans 10231 and C. albicans 2091 was 1.6% and 82.4%, respectively; while the clinical strain of C. albicans was inhibited by 92.2%. The polymer composite loaded with 5 wt.%-Triclosan displayed a stability over the period that illustrates the possibility of washing the composite surface.
