Project description:Spurred on by recent advances in materials chemistry and drug delivery, a new stimuli-responsive theranostic hybrid platform, based on mechanized monodisperse nano metal-organic frameworks (NMOFs) gated by carboxylatopillar[5]arene (CP5) switches with bio-friendly pH-triggered cargo release capabilities, has been constructed for the first time. This nanoscale smart cargo delivery system showed pH- and/or competitive binding agent-triggered controlled cargo release with negligible premature release, large pore sizes for drug encapsulation, low cytotoxicity, good biodegradability and biocompatibility, and potential application in cell imaging, which offers a new tool in targeted drug delivery and the controlled release of therapeutic agents.

A pH-Responsive Supramolecular Drug Delivery System Constructed by Cationic Pillar[5]arene for Enhancing Antitumor Activity.

Project description:Drug delivery systems have good biocompatibiliy and low side effects for cancer treatment, but overcoming high efficiency of drug-loading and the drug-targeting controlled release still remains challenging. In this work, supramolecular vesicles, with pH-triggering effect, have been successfully constructed for drug delivery, which are fabricated by the complexation between a cationic pillar[5]arene (DAWP5) and a sodium dodecyl sulfonate (SDS) in aqueous solution. Drug-loading and releasing results demonstrated that anticancer drug doxorubicin (DOX) could be loaded efficiently by such cationic vesicles in neutral condition, and the drug release could be controlled in the simulated weak acid environment of tumor cells. Moreover, the vesicles had low cytotoxicity to normal human cell (L02), while the DOX-loaded vesicles could significantly enhance the cytotoxicity of free DOX for normal cell L02 and four tested tumor cells (Hela, HepG2, MGC-803 and T24). Especially for HepG2, after 24 h incubation time, IC50 of DOX-loaded vesicles was only 0.79 μM, about 23% of that of DOX (3.43 μM). These results suggested that such novel vesicles have promising potential to construct nano-drug delivery systems for various biomedical applications.

GOx-assisted synthesis of pillar[5]arene based supramolecular polymeric nanoparticles for targeted/synergistic chemo-chemodynamic cancer therapy.

Project description:BackgroundCancer is the most serious world's health problems on the global level and various strategies have been developed for cancer therapy. Pillar[5]arene-based supramolecular therapeutic nano-platform (SP/GOx NPs) was constructed successfully via orthogonal dynamic covalent bonds and intermolecular H-bonds with the assistance of glucose oxidase (GOx) and exhibited efficient targeted/synergistic chemo-chemodynamic cancer therapy.MethodsThe morphology of SP/GOx NPs was characterized by DLS, TEM, SEM and EDS mapping. The cancer therapy efficinecy was investigated both in vivo and in vitro.ResultsSP/GOx NPs can load drug molecules (Dox) and modify target molecule (FA-Py) on its surface conveniently. When the resultant FA-Py/SP/GOx/Dox NPs enters blood circulation, FA-Py will target it to cancer cells efficiently, where GOx can catalyst the overexpressed glucose to generate H2O2. Subsequently, the generated H2O2 in cancer cells catalyzed by ferrocene unit to form •OH, which can kill cancer cells. Furthermore, the loaded Dox molecules released under acid microenvironment, which can further achieve chemo-therapy.ConclusionAll the experiments showed that the excellent antitumor performance of FA-Py/SP/GOx/Dox NPs, which provided an new method for pillar[5]arene-based supramolecular polymer for biomedical applications.

Photolysis of the BODIPY dye activated by pillar[5]arene.

Project description:Here, a pseudo[3]rotaxane comprising a fluorescent BODIPY derivative and pillar[5]arene was conveniently fabricated via host-guest complexation. Importantly, in this system, the efficient photodecomposition of the BODIPY derivative in the presence of pillar[5]arene was witnessed upon irradiation at 311 nm light, which was demonstrated via UV-Vis absorption, fluorescence emission, NMR and HR-MS spectroscopy techniques, but the only BODIPY dye in the absence of pillar[5]arene couldn't undergo photodegradation. We demonstrated that pillar[5]arene could act as an activator to trigger the photodegradation reaction of BODIPY derivatives via free radical reactions even without supramolecular interactions. The present results provide a new strategy for the efficient photolysis of organic dyes.

Supramolecular Hybrid Material Constructed from Graphene Oxide and Pillar[6]arene-Based Host-Guest Complex as a Ultrasound and Photoacoustic Signals Nanoamplifier.

Project description:Photoacoustic imaging combines the merits of ultrasound imaging and optical imaging that allows a fascinating imaging paradigm with deeper tissue penetration than optical imaging and higher spatial resolution than ultrasound imaging. Herein, we develop a supramolecular hybrid material composed of graphene oxide (GO) and a pillar[6]arene-based host-guest complex (CP6⊃PyN), which can be used as a ultrasound (US) and photoacoustic (PA) signal nanoamplifier. Triggered by the near-infrared (NIR) light mediated photothermal effect, CO2 nanobubbles are generated on the surface of GO@CP6⊃PyN due to the decomposition of bicarbonate counterions, thus strongly amplifying its US and PA performances. Our study, for the first time, demonstrates enhanced US and PA activity in supramolecular hybrid material on the basis of host-guest chemistry as a photoacoustic nanoplatform.

Nano-Theranostics Constructed from Terpyridine-Modified Pillar [5]arene-Based Supramolecular Amphiphile and Its Application in Both Cell Imaging and Cancer Therapy.

Project description:Theranostics play an important role in cancer treatment due to its realized real-time tracking of therapeutic efficacy in situ. In this work, we have designed and synthesized a terpyridine-modified pillar [5]arenes (TP5). By the coordination of terpyridine and Zn2+, the complex TP5/Zn was obtained. Then, supramolecular amphiphile can be constructed by using host-guest complexation between a polyethylene glycol contained guest (PM) and TP5/Zn. Combining the fluorescence properties from the terpyridine group and the amphiphilicity from the system, the obtained TP5/Zn/PM can further be self-assembled into fluorescent particles with diameters of about 150 nm in water. The obtained particles can effectively load anti-cancer drugs and realize living cell imaging and a precise release of the drugs.

Green, Efficient Detection and Removal of Hg<sup>2+</sup> by Water-Soluble Fluorescent Pillar[5]arene Supramolecular Self-Assembly.

Project description:Developing a water-soluble supramolecular system for the detection and removal of Hg2+ is extremely needed but remains challenging. Herein, we reported the facile construction of a fluorescent supramolecular system (H⊃G) in 100% water through the self-assembly of carboxylatopillar[5]arene sodium salts (H) and diketopyrrolopyrrole-bridged bis(quaternary ammonium) guest (G) by host-guest interaction. With the addition of Hg2+, the fluorescence of H⊃G could be efficiently quenched. Since Hg2+ showed synergistic interactions (coordination and Hg2+- cavity interactions with G and H, respectively), crosslinked networks of H⊃G@Hg2+ were formed. A sensitive response to Hg2+ with excellent selectivity and a low limit of detection (LOD) of 7.17 × 10-7 M was obtained. Significantly, the quenching fluorescence of H⊃G@Hg2+ can be recovered after a simple treatment with Na2S. The reusability of H⊃G for the detection of Hg2+ ions was retained for four cycles, indicating the H⊃G could be efficiently used in a reversible manner. In addition, the H⊃G could efficiently detect Hg2+ concentration in real samples (tap water and lake water). The developed supramolecular system in 100% water provides great potential in the treatment of Hg2+ detection and removal for environmental sustainability.

Nano-hydroxyapatite polymeric hydrogels for dye removal.

Project description:Herein, two kinds of nano-hydroxyapatite were synthesized from Clam and Magellan shell by wet chemical precipitation method. Mainly, carboxymethyl cellulose/acrylamide/nano-hydroxyapatite composite hydrogels were developed via a free-radical polymerization process and investigated as a sorbent for Acid Blue 113 (AB) from aqueous AB solution. The swelled and kinetic behaviours of hydrogels were investigated using a gravimetric method. The swelling properties of the CMC-AM-hydrogels were influenced by the calcium electrolytes (Ca2+) content in nano-hydroxyapatites. The diffusion coefficient value increased with the increase of nano-hydroxyapatite content in the CMC-AM/nHA-CS (0.22353-0.27681 cm2 s-1) and CMC-AM/nHA-MS (0.22377-0.29737 cm2 s-1) hydrogels. The mechanism of water diffusion was found to be anomalous transport. The CMC-AM/nHA-MS hydrogels showed high AB absorption efficiency and adsorption capacities. These results explained that the nano-hydroxyapatites of Magellan shells based hydrogels are attractive nanocomposite hydrogels for the adsorption of dye in the water purification applications.

Size Switchable Supramolecular Nanoparticle Based on Azobenzene Derivative within Anionic Pillar[5]arene.

Project description:A photo/thermal-switchable supramolecular nanoparticles assembly has been constructed based on an inclusion complex between anionic pillar[5]arene 2C-WP5A and azobenzene derivative Azo-py-OMe (G). The novel anionic pillar[5]arene-based host-guest inclusion complexation was investigated by the 1H NMR titration, 2D ROESY and isothermal titration microcalorimetry (ITC) showing high association constant (Ka) of (2.60?±?0.06)?×?104?M-1 with 1:1 binding stoichiometry. Furthermore, the supramolecular nanoparticles assembly can be conveniently obtained from G and a small amount of 2C-WP5A in aqueous solution, which was so-called "host induced aggregating (HIA)". The size and morphology of the supramolecular nanoparticles assembly were characterized by TEM and DLS. As a result of the photo/thermal-isomerization of G included in the cavity of 2C-WP5A, the size of these nanoparticles could reversibly change from ~800?nm to ~250?nm, which could switch the solution of this assembly from turbid to clear.
