Project description:Nanothermometry could realize stable, efficient, and noninvasive temperature detection at the nanoscale. Unfortunately, most applications of nanothermometers are still limited due to their intricate synthetic process and low-temperature sensitivity. Herein, we reported a kind of novel bismuth-based upconversion nanomaterial with a fast and facile preparation strategy. The bismuth-based upconversion luminophore was synthesized by the co-precipitation method within 1 minute. By optimizing the doping ratio of the sensitizer Yb ion and the activator Er ion and adjusting the synthetic solvent strategy, the crystallinity of the nanomaterials was increased and the upconversion luminescence intensity was improved. Ratiometric upconversion optical measurements of temperature in the range of 278 K to 358 K can be achieved by ratiometric characteristic emission peaks of thermally sensitive Er ion. This method of rapidly constructing nanometer temperature probes provides a feasible strategy for the construction of novel fluorescent temperature probes.

Project description:For many years, fluorosis has been known as a worldwide disease which seriously diminishes quality of life through skeletal embrittlement and hepatic damage. Aiming to develop novel drugs for simultaneous fluorosis diagnosis and therapy, in this work we explore the feasibility of a novel pyrogallic acid-titanium(iv) complex-modified upconversion nanoprobe (UCNP-PA-Ti) for F- capture and real-time quantification. Utilizing the strong interaction between Ti4+ and F-, the modified PA-Ti decomposes in F--containing solution, which not only weakens the FRET but results in upconversion luminescence (UCL) recovery. Both in vitro and in vivo experiments demonstrate a highly sensitive F- UCL response and therapeutic efficiency, which was promising for successful UCL image monitoring and the therapeutic process. Long blood circulation time and low toxicity ensured their safe application for fluorosis theranostics. Our work provides a new possibility for F- concentration detection within fluorosis therapeutic periods and encourages the development of novel drugs for fluorosis theranostics.

Project description:Intracellular detection is highly desirable for biological research and clinical diagnosis, yet its quantitative analysis with noninvasivity, sensitivity, and accuracy remains challenging. Herein, a near-infrared (NIR) dual-excitation strategy is reported for ratiometric intracellular detection through the design of dye-sensitized upconversion probes and employment of a purpose-built NIR dual-laser confocal microscope. NIR dye IR808, a recognizer of intracellular analyte hypochlorite, is introduced as energy donor and Yb,Er-doped NaGdF4 upconversion nanoparticles are adopted as energy acceptor in the as-designed nanoprobes. The efficient analyte-dependent energy transfer and low background luminescence endow the nanoprobes with ultrahigh sensitivity. In addition, with the nonanalyte-dependent upconversion luminescence (UCL) excited by 980 nm as a self-calibrated signal, the interference from environmental fluctuation can be alleviated. Furthermore, the dual 808/980 nm excited ratiometric UCL is demonstrated for the quantification of the level of intracellular hypochlorite. Particularly, the intrinsic hypochlorite with only nanomolar concentration in live MCF-7 cells in the absence of exogenous stimuli is determined. Such an NIR dual-excitation ratiometric strategy based on dye-sensitized UCL probes can be easily extended to detect various intracellular analytes through tailoring the reactive NIR dyes, which provides a promising tool for probing biochemical processes in live cells and diagnosing diseases.

Project description:In this study, a novel dual-emission ratiometric fluorescent nanoprobe (RFN) was synthesized and ultilized for highly sensitive determination of mercury ions. In this nanoprobe, fluorescein isothiocyanate (FITC) doped silica (SiO2) served as a reference signal, FITC-SiO2 microspheres were synthesized and modified with amino groups, and then Au Nanoclusters (AuNCs) were combined with the amino groups on the surface of the FITC-SiO2 microspheres to obtain the RFN. The selectivity, stability, and pH of the RFN were then optimized, and the determination of mercury ions was performed under optimal conditions. The probe fluorescence intensity ratio (F520 nm/F680 nm) and Hg2+ concentration (1.0 × 10-10 mol/L to 1.0 × 10-8 mol/L) showed a good linear relationship, with a correlation coefficient of R2 = 0.98802 and a detection limit of 1.0 × 10-10 mol/L, respectively. The probe was used for the determination of trace mercury ion in water samples, and the recovery rate was 98.15~100.45%, suggesting a wide range of applications in monitoring pollutants, such as heavy metal ion and in the area of environmental protection.

Project description:We report on the potential application of NIR-to-NIR Nd3+-doped yttrium vanadate nanoparticles with both emission and excitation operating within biological windows as thermal sensors in 123-873 K temperature range. It was demonstrated that thermal sensing could be based on three temperature dependent luminescence parameters: the luminescence intensity ratio, the spectral line position and the line bandwidth. Advantages and limitations of each sensing parameter as well as thermal sensitivity and thermal uncertainty were calculated and discussed. The influence of Nd3+ doping concentration on the sensitivity of luminescent thermometers was also studied.

Project description:Posterior segment ocular diseases are highly prevalent worldwide due to the lack of suitable noninvasive diagnostic and therapeutic tactics. Herein, concerning this predicament, we designed a hybrid retina-targeted photothermal theranostic nanoplatform (UCNPs@Bi@SiO2@GE HP-lips), based on the unique upconversion luminescence (UCL) imaging of upconversion nanoparticles (UCNPs), efficient photothermal conversion ability of Bi nanoparticles, and thermal-induced phase transition properties of the liposomes (lips). The nanoplatform was functionalized with penetratin (PNT) and hyaluronic acid (HA), to obtain retina-targeted liposomes (HP-lips). Lipophilic genistein (GE) was entrapped into the liposomes (GE HP-lips). An in vitro release study showed NIR irradiation could photothermally trigger controlled release of GE from the liposomal platform. Moreover, cellular uptake evaluation via UCL imaging demonstrated UCNPs@Bi@SiO2@GE HP-lips represented the brightest UCL, compared with other formulations, which is beneficial for the accurate evaluation of the prognosis and severity of angiogenesis-related posterior segment disorders. Therefore, UCNPs@Bi@SiO2@GE HP-lips exhibit promising potential as a theranostic nanoplatform for posterior segment ocular diseases.

Project description:Two sensitive and ratiometric fluorescent probes (probe I and probe II) were developed for the detection of fluoride ions. Probe I can detect fluoride ions quantitatively within a range of 0-6 μM and a detection limit of 73 nM, while probe II has a range of 0-40 μM and a detection limit of 138 nM. The test strips from probe I are quickly able to recognize F- (5 min) inside of the F- safety level in drinking water (1.0 mg/L, ∼5 μM) under 254 nm ultraviolet light, and the test strips from probe II quickly recognize F- (12 min) in dangerously high F- levels in water (4.0 mg/L, ∼21 μM) under 254 nm ultraviolet light. This combination of fluorescent paper sensors from probe I and probe II can be used as a simple and convenient tool to determine whether water is safe to drink or dangerous.

Project description:A newly designed cyanide-selective chemosensor based on chromone containing benzothiazole groups [3-(2,3-dihydro-benzothiazol-2-yl)-chromen-4-one (DBTC)] was synthesized and structurally characterized by physico-chemical, spectroscopic, and single-crystal X-ray diffraction analyses. The compound DBTC can detect cyanide anions based on nucleophilic addition as low as 5.76 nM in dimethyl sulfoxide-N-(2-hydroxyethyl)piperazine-N'-ethanesulfonic acid buffer (20 mM, pH 7.4) (v/v = 1:3). The binding mode between receptor DBTC and cyanide nucleophile has also been demonstrated by experimental studies using various spectroscopic tools and theoretical studies, and the experimental work has also been verified by characterizing one supporting compound of similar probable structure of the final product formed between DBTC and cyanide ion (DBTC-CN compound) by single-crystal X-ray analysis for detailed structural analyses. In theoretical study, density functional theory procedures have been used to calculate the molecular structure and the calculation of the Fukui function for evaluation of the electrophilic properties of each individual acceptor atom. Furthermore, the efficacy of the probe (DBTC) to detect the distribution of CN- ions in living cells has been checked by acquiring the fluorescence image using a confocal microscope. Notably, the paper strips with DBTC were prepared, and these could serve as efficient and suitable CN- test kits successfully.

Project description:Upconversion nanophosphors, particles that can absorb low energy radiation and emit high energy light through multi-photon absorption processes, have gained augmented attention in recent years. Due to their admirable optical and chemical properties, these nanoparticles are finding wide range of applications in the field of bioimaging, light emitting devices and security printing. However, for any practical application, it is extremely important that a facile synthetic route is developed that can lead to the generation of nanophosphors exhibiting efficient upconversion luminescence under diverse experimental conditions. Herein, we report a new ligand-free approach for the synthesis of lanthanide-based upconversion nanoparticles by adopting a simple solid-state synthetic route. The reaction conditions such as temperature and time were optimized to obtain nanophosphors exhibiting enhanced upconversion luminescence. The synthesized nanoparticles, due to its ligand-free nature, could be well dispersed in both aqueous and organic media. The nanophosphors retained the upconversion luminescence under varying time, pH and temperature, indicating that the absence of ligand had least effect on their stability. The nanophosphors were found to exhibit good cell viability even under high concentrations, unveiling their potential as bioimaging agents in the biomedical research.

Project description:A highly selective, novel BINOL based sensor BBCN has been developed for the fluorescent ratiometric detection of cyanide ions (CN-). The optical study revealed that BBCN exhibited unique spectral changes only with cyanide ions in the presence of other competing ions. Besides, an apparent fluorescent colour change from green to blue was observed. A clear linear relationship was observed between the fluorescence ratiometric ratio of BBCN and the concentration of CN- with a reasonably low detection limit (LOD) of 189 nM (507 ppb). The optical response was due to the nucleophilic addition of CN- to the dicyanovinyl group of the sensor, which compromises the probe's intramolecular charge transfer. This mechanism was well confirmed by Job's plot, 1H-NMR and ESI-MS studies. BBCN showed immediate spectral response towards (1 second) CN- and detection could be realized in a broad pH window. Furthermore, the practical utility of BBCN was studied by test paper-based analysis and the detection of CN- in various water resources.
