Factors contributing to tourism demand at major Japanese hot springs.

ABSTRACT: Hot springs are a major tourism resource in nature-based tourism, and the hot springs market is one of the biggest sectors in wellness tourism markets. In the present study, we examine factors contributing to tourism demand for major hot spring resorts in Japan using ordinary least squares regression models and generalized linear mixed models, and compare the estimation results. The results show that significant factors in most of our models are quality of accommodations and the degree of dependence on inbound demand. Furthermore, the number of non-Japanese languages supported on websites of hot spring resorts has a significant impact on inbound demand. Since the results of the present study cover more than 80 hot spring sites, the results highlight common important factors for hot spring resorts. Such widely applicable factors have been missing in previous studies, and the present study fills this research gap.

Project description:Community structures of submerged microbial slime streamers (SMSS) in sulfide-containing hot springs at 72 to 80 degrees C at Nakabusa and Yumata, Japan, were investigated by molecular analysis based on the 16S rRNA gene. The SMSS were classified into two consortia; consortium I occurred at lower levels of sulfide in the hot springs (less than 0.1 mM), and consortium II dominated when the sulfide levels were higher (more than 0.1 mM). The dominant cell morphotypes in consortium I were filamentous and small rod-shaped cells. The filamentous cells hybridized with fluorescent oligonucleotide probes for the domain Bacteria, the domain Archaea, and the family Aquificaceae: Our analysis of the denaturing gradient gel electrophoresis (DGGE) bands by using reverse transcription (RT)-PCR amplification with two primer sets (Eub341-F with the GC clamp and Univ907R for the Bacteria and Eub341-F with the GC clamp and Arch915R) indicated that dominant bands were phylogenetically related to microbes in the genus Aquifex: On the other hand, consortium II was dominated by long, small, rod-shaped cells, which hybridized with the oligonucleotide probe S-*-Tdes-0830-a-A-20 developed in this study for the majority of as-yet-uncultivated microbes in the class Thermodesulfobacteria: The dominant DGGE band obtained by PCR and RT-PCR was affiliated with the genus Sulfurihydrogenibium: Moreover, our analysis of dissimilatory sulfite reductase (DSR) gene sequences retrieved from both consortia revealed a high frequency of DSR genes corresponding to the DSR of Thermodesulfobacteria-like microorganisms. Using both sulfide monitoring and (35)SO(4)(2-) tracer experiments, we observed microbial sulfide production and consumption by SMSS, suggesting that there is in situ sulfide production by as-yet-uncultivated Thermodesulfobacteria-like microbes and there is in situ sulfide consumption by Sulfurihydrogenibium-like microbes within the SMSS in the Nakabusa and Yumata hot springs.

Project description:Little is known about the distribution and ecological functions of abundant, intermediate, and rare biospheres and their correlations with environmental factors in hot springs. Here, we explored the microbial community composition of total, abundant, intermediate, and rare biospheres in 66 Tibetan hot springs (pairwise geographic distance 0-610 km, temperature 32-86°C, pH 3.0-9.5, and salinity 0.13-1.32 g/L) with the use of Illumina MiSeq high-throughput sequencing. The results showed that the abundant sub-communities were mainly composed of Chloroflexi, Proteobacteria, Deinococcus-Thermus, Aquificae, Bacteroidetes, and Firmicutes. In contrast, the rare sub-communities mainly consisted of most newly proposed or candidate phyla of Dictyoglomi, Hydrogenedentes, Atribacteria, Hadesarchaea, Aminicenantes, Microgenomates, Calescamantes, Omnitrophica, Altiarchaeales, and Chlamydiae. However, the abundant and rare sub-communities shared some common phyla (e.g., Crenarchaeota, Bathyarchaeota, and Chlorobi), which were composed of different OTUs. The abundant, intermediate, and rare sub-communities were mainly influenced by different environmental variables, which could be ascribed to the fact that they may have different growth and activity and thus respond differently to these variables. Spatial factors showed more contribution to shaping of the intermediate and rare communities than to abundant sub-community, suggesting that the abundant taxa were more easily dispersed than their rare counterparts among hot springs. Microbial ecological function prediction revealed that the abundant and rare sub-communities responded differently to the measured environmental factors, suggesting they may occupy different ecological niches in hot springs. The rare sub-communities may play more important roles in organic matter degradation than their abundant counterparts in hot springs. Collectively, this study provides a better understanding on the microbial community structure and potential ecological functions of the abundant and rare biospheres in hot spring ecosystems. The identified rare taxa provide new opportunities of ecological, taxonomic and genomic discoveries in Tibetan hot springs.
