ABSTRACT: Data in this article are acquired from 46 naturally distilled whiskey. Whiskey samples were produced in the United Kingdom, United States, Ireland, Scotland, and Canada. Samples differ with their type of distillery including (Blended, finest blended, Malted, single Malted, rye, straight Rye, special blended, and special reserved), years of aging, and ethanol percentages. The contents of beneficial bioactive components including total phenolic contents and total anti-oxidants activity and the correlation between them were addressed.

Unexploited Antineoplastic Effects of Commercially Available Anti-Diabetic Drugs.

Project description:The development of efficacious antitumor compounds with minimal toxicity is a hot research topic. Numerous cancer cell targeted agents are evaluated daily in laboratories for their antitumorigenicity at the pre-clinical level, but the process of their introduction into the market is costly and time-consuming. More importantly, even if these new antitumor agents manage to gain approval, clinicians have no former experience with them. Accruing evidence supports the idea that several medications already used to treat pathologies other than cancer display pleiotropic effects, exhibiting multi-level anti-cancer activity and chemosensitizing properties. This review aims to present the anticancer properties of marketed drugs (i.e., metformin and pioglitazone) used for the management of diabetes mellitus (DM) type II. Mode of action, pre-clinical in vitro and in vivo or clinical data as well as clinical applicability are discussed here. Given the precious multi-year clinical experience with these non-antineoplastic drugs their repurposing in oncology is a challenging alternative that would aid towards the development of therapeutic schemes with less toxicity than those of conventional chemotherapeutic agents. More importantly, harnessing the antitumor function of these agents would save precious time from bench to bedside to aid the fight in the arena of cancer.

Catechin Composition, Phenolic Content, and Antioxidant Properties of Commercially-Available Bagged, Gunpowder, and Matcha Green Teas.

Project description:The leaves of the Camellia sinensis plant are used to produce many different types of teas, and green tea is particularly noted for its health promoting properties which are attributed to high concentrations of phenolic compounds known as catechins. Green tea is available in forms such as bagged, gunpowder (rolled leaf), and matcha, a fine powder. This study evaluated the phenolic content, catechin composition, caffeine concentration, and antioxidant properties of fifteen commercially-available green teas with an emphasis on identifying differences between ceremonial and culinary matcha. Gunpowder and bagged green teas had higher total phenolic contents and greater CUPRAC (cupric ion reducing antioxidant capacity) and ORAC (oxygen radical absorbance capacity) antioxidant capacities than matcha teas. Among matcha samples, less expensive culinary teas had greater total phenolic contents and higher antioxidant capacities than ceremonial products. Hierarchical cluster analysis was used to determine similarities and differences in catechin composition and revealed that bagged and gunpowder green teas had the highest levels of the stereoisomers epi/catechin gallate, which correlated with their strong antioxidant properties. Bagged and gunpowder teas were the lowest cost products in this study, indicating that they are an economical choice for consumers who want a green tea beverage with high catechin content, although they generally had lower caffeine levels than matcha.

Project description:Phytochemicals in red and purple bran rice have potential health benefit to humans. We determined the phytochemicals in brans of 32 red and purple global rice varieties. The description of the origin and physical traits of the whole grain (color, length, width, thickness and 100-kernel weight) of this germplasm collection are provided along with data of total flavonoid and total phenolic contents, oxygen radical absorbance capacity and total proanthocyanidin contents. The contents and proportions of individual oligomers, from degree of polymerization of monomers to 14-mers, and polymers in bran of these 32 rice varieties are presented (DOI: http://dx.doi.org/10.1016/j.foodchem.2016.04.004) [1].

Total Phenolic Contents and Antioxidant Potential of Herbs Used for Medical and Culinary Purposes.

Project description:Herbs used for medical purposes are required to meet high pharmacopoeial quality standards, whereas spices used as additives to dishes and food products do not have to meet such rigorous standards. Therefore, the aim of this paper was to compare medicinal herbs and spices of the same plant species that are applied in different areas with regard to the total phenolic content (TPC) and total antioxidant capacity (TAC) of their methanolic and water extracts. This study showed that the TPC values of both extracts prepared from medicinal herbs and spices as quantified using the Folin-Ciocalteu reagent did not differ significantly; however, the TAC values obtained by the DPPH assays of the methanol and water extracts differed significantly. No such differences were found for the same extracts when the FRAP assay was used for analysis. This study shows that the level of antioxidants is dependent on the plant species and the botanical family. Plants belonging to the Lamiaceae and Asteraceae families are richer in phenolic compounds and have stronger antioxidant potentials than those originating from the Apiaceae family. Multivariate statistical analysis indicates that the majority of medicinal herbs and spices of the same plant species are found in the same cluster. In addition, the extraction solvent was identified as one of the factors influencing the clustering of the plants on the PCA scatterplot and CA dendrogram.

Total phenolic content and total antioxidant capacity of common Indian pulses and split pulses.

Project description:Pulses are valued for their protein content and are good source of bioactive phytochemicals. The objectives of this study were to estimate total phenolic content (TPC), flavonoid content and total antioxidant capacity (TAC) through six different assay methods from twenty seven different varieties of pulses and split pulses commonly consumed in India. TPC and flavonoid content ranged between 38.6-542.7 mg GAE/100 g and 18.3-344.7 mg RE/100 g respectively. TAC measured by FRAP (90.6-2773.5 mg TE/100 g), RPA (190.8-2128.4 mg TE/100 g), DPPH radical scavenging assay (RSA) (42.9-571.1 mg TE/100 g), ABTS RSA (4.5-194.9 mg TE/100 g), hydroxyl RSA (0.0-106.6 mg AAE/g) and nitric oxide (NO) RSA (11.9-43.7 mg AAE/100 g) varied widely among pulses and split pulses analysed in the present study. TAC measured using different methods showed positively strong and significant correlation with TPC as well as flavonoid content except NORSA, indicating that phenolic compounds are the major contributors to the antioxidant properties of these plants. Food composition data obtained in this study would enrich the composition database at global level, which in turn will guide in improving food and nutrition security of the population. These can be used for further research, diet formulation and epidemiological studies. From the results it can be concluded that cow pea (red, small) had the highest TPC and TAC measured by all the parameters except NORSA.

Evaluation of commercially available small RNA methods for plant samples

Project description:Plant small RNAs are a diverse and complex set of molecules, ranging in length from 21 to 24 nt, involved in a wide range of essential biological processes. Nowadays, high-throughput sequencing is the most commonly used method for the discovery and quantification of small RNAs. However, it is known that several biases can occur during the preparation of small RNA libraries, especially using low input RNA. We used two types of plant biological samples to evaluate the performance of seven commercially available methods for small RNA library construction, using different RNA input amounts. We show that when working with plant material, library construction methods have differing capabilities to capture small RNAs, and that different library construction methods provide better results when applied to the detection of microRNAs, phased small RNAs, or tRNA-derived fragments.

Anti-Hypochlorite and Catalytic Activity of Commercially Available Moringa oleifera Diet Supplement.

Project description:Aiming at the assessment of the pro-health, and especially anti-hypochlorite properties of Moringa oleifera species a representative, commercially available Moringa oleifera dietary supplement was used as a substrate for the preparation of aqueous Moringa extract. The anti-hypochlorite activity of the extract was assessed using the hypochlorite-specific coumarin-based fluorescence turn-off sensor, namely 7-diethylamino-coumarin-3-carboxylic acid (7-DCCA). This compound was synthesized via the Knoevenagel condensation of 4-diethylamino-2-hydroxybenzaldehyde with Meldrum's acid and the Moringa extract was employed as a medium and catalyst. Moreover, the total phenolic content (TPC) as well as the reactive oxygen species (ROS)-scavenging ability of the aqueous Moringa extract were determined. The results obtained demonstrated the applicability of Moringa extract as an anti-hypochlorite agent. Additionally, the satisfactory yield of the 7-DCCA obtained suggests the usefulness of the extract as a catalyst and the reaction medium. The antioxidative potential of the extract was notably lower than that of the standard (TROLOX). Determination of TPC in 100 g of the dry weight (DW) of studied material revealed a high number of polyphones present.

Anti-Hyperglycemic Effects of Green Crab Hydrolysates Derived by Commercially Available Enzymes.

Project description:The predation and burrowing activity of invasive green crabs have had detrimental effects on important marine resources and habitats. Our objective is to develop bioactive hydrolysates by enzymatic proteolysis of underutilized green crab. Mechanically separated mince was hydrolyzed with Alcalase, Protamex, Flavourzyme, and Papain (1%) for 60 min. Subsequently, the hydrolysates were introduced to a simulated gastrointestinal digestion model. Selected samples were fractionated by ultrafiltration, and their anti-hyperglycemic effects including α-glucosidase, α-amylase, and dipeptidyl peptidase-IV (DPP-IV) inhibitory activities and glucagon-like 1 (GLP-1) secretory activity were evaluated. The Protamex treatment showed the highest α-glucosidase inhibitory activity (IC50 1.38 ± 0.19 mg/mL) compared to other enzyme treatments and the crab mince control, and its α-amylase inhibitory activity (IC50 11.02 ± 0.69 mg/mL) was lower than its α-glucosidase inhibitory activity. Its GLP-1 secretory activity was approximately four times higher than the positive control (10 mM glutamine). The <3 kD fraction contributed significantly to the anti-hyperglycemic activity of Protamex-derived hydrolysates, and this activity was stable after simulated digestion. Our results suggest that green crab hydrolysates obtained by Protamex treatment have the potential for type 2 diabetes management and could be incorporated in food products as a health-promoting ingredient.

Alternatives based on liquid oils, fully hydrogenated fats, and exotic fats like shea and sal etc., have been developed by the research groups of several specialty oils and fats suppliers. This article describes the advantages and disadvantages of those products and compares them to similar products based on palm oil. It is also discussed how reasonable the replacement of palm products would be, since sustainable and 3-MCPD/glycidolester-reduced palm based specialty oils are also available on the market.

Comparative Assessment of Anti-HLA Antibodies Using Two Commercially Available Luminex-Based Assays.

Project description:BackgroundAllospecific anti-HLA antibodies (Abs) are associated with rejection of solid organ grafts. The 2 main kits to detect anti-HLA Ab in patient serum are commercialized by Immucor and One Lambda/ThermoFisher. We sought to compare the performance of both platforms.MethodsBackground-adjusted mean fluorescence intensity (MFI) values were used from both platforms to compare sera collected from 125 pretransplant and posttransplant heart and lung transplant recipients.ResultsMost HLA class I (94.5%) and HLA class II (89%) Abs with moderate to high MFI titer (≥4000) were detected by both assays. A modest correlation was observed between MFI values obtained from the 2 assays for both class I (r = 0.3, r2 = 0.09, P < 0.0001) and class II Ab (r = 0.707, r2 = 0.5, P < 0.0001). Both assays detected anti-class I and II Ab that the other did not; however, no specific HLA allele was detected preferentially by either of the 2 assays. For a limited number of discrepant sera, dilution resulted in comparable reactivity profiles between the 2 platforms.ConclusionsImmucor and One Lambda/ThermoFisher assays have a similar, albeit nonidentical, ability to detect anti-HLA Ab. Although the correlation between the assays was present, significant variances exist, some of which can be explained by a dilution-sensitive "prozone" effect.
