Characterization of ZIC4 role in choroid plexus carcinoma

ABSTRACT: The study aimed to investigate the role of ZIC4, a transcription factor, in pediatric choroid plexus carcinoma (CPC) using a cell line model (CCHE-45 cell line). We employed RNA sequencing and quantitative proteomics to provide insights into the potential mechanisms underlying its tumor suppressor role in CPC.

Project description:ZIC4 is a transcription factor related to pathways like NOTCH and Sonic Hedgehog (Shh) pathways which are involved in choroid plexus carcinoma (CPC). The current study describes the effect of ZIC4 gene re-expression on a CPC cell line proteome. We performed SILAC-based proteomics of ZIC4-transfected cells versus control cells, as well as empty vector-transfected cells versus control cells. Re-expression of ZIC4 may lead to a better understanding of global gene regulation and identify its role in CPC pathogenesis.

Role of bcatenin in hepatocellular carcinoma

Project description:Hepatocellular carcinoma (HCC) is the second leading cause of cancer-related deaths worldwide. β-catenin is widely thought to be a major oncogene in HCC based on the frequency of mutations associated with aberrant Wnt signaling in HCC patients. Challenging this model, our data reveal that β-catenin nuclear accumulation is restricted to the late stage of the disease. Until then, β-catenin is primarily located at the plasma membrane in complex with multiple cadherin family members where it drives tumor cell survival by enhancing the signaling of growth factor receptors such as EGFR. Therefore, our study reveals the evolving nature of β-catenin in HCC to establish it as a compound tumor promoter during the progression of the disease.
