Cryptotanshinone and tanshinone IIA promote the expression of tight junction proteins by targeting microRNA-21-5p and contribute the effect of inhibit lung cancer cells

ABSTRACT: Total RNA extraction was done from A549 after different treatments with TRIzol reagent. The mRNA profile of cells in different treatment groups was analyzed by RNA sequencing (RNA-SEQ),Differentially expressed genes were determined based on Q value (Adjusted P-value) <= 0.05. Isolated RNA samples were sent to BGI Co., LTD. in Shenzhen for mRNA-seq using BGISEQ-500. The reference gene sequences were cleaned and sorted by Bowtie2, and then the gene expression level data were calculated by RSEM. Finally, the differentially expressed genes (DEGs) were obtained by DESeq2 analysis (Q value ≤0.05). There were 187 and 75 differentially expressed genes between Control and CPT, and Control and tania, respectively, Venn diagram showed that CPT and TanIIA could simultaneously act on 51 DEGs .
In the CPT group, 109 DEGs were up-regulated and 78 DEGs were down-regulated . Then, we compared the gene expression of A549 cells before and after TanIIA intervention, and found that 35 genes were up-regulated and 40 DEGs were down-regulated after exposure (Figure 2d). Additionally, clustered heatmaps of the top 20 up-regulated and down-regulated DEGs expression were shown in Figure 2E and 2F based on z-scores column direction, respectively.
