Effect of over-expression of AR-V7 in MDA-MB-453 breast cancer cells

ABSTRACT: The effect of transient transfection of a construct designed to over-express the androgen receptor (AR) variant AR-V7 on gene expression in MDA-MB-453 cells was assessed using Affymetrix Gene 2.0 ST arrays. Transfection of an AR-expressing construct or an empty construct served as controls. AR-V7, AR or empty vector was transfected into MDA-MB-453 cells. Cells were treated with vehicle control or DHT.

Project description:Analysis of MDA-MB-453 breast cancer cells treated with the androgen 5a-dihydrotestosterone (DHT) for 6h, 16h and 48h to define the genes that are differentially regulated in response to DHT. MDA-MB-453 breast cancer cells were treated with 5a-dihydrotestosterone (DHT) for time course, followed by RNA extraction and hybridization on Affymetrix microarrays, in order to obtain the gene expression profiles at three time points. The vehicle treated samples are used as control.

AR heterogeneity in TNBC MDA-MB-453 cells

Project description:Triple-negative breast cancer (TNBC) is aggressive and difficult, and few targeted therapies are available to treat this patient population. The androgen receptor (AR) has emerged as a potential target in breast cancer. Newer generation AR inhibitors, such as Seviteronel (Sevi), are unique in their ability to inhibit AR both directly and by blocking upstream androgen synthesis. The purpose of this study was to investigate the pre-clinical activity of Sevi in TNBC and further explore the effectiveness of targeting both androgen biosynthesis and AR activity in combination with other downstream acting agents. AR overexpressing (AR+) TNBC cell lines, xenografts and androgen responsive patient-derived xenograft (PDX) models were using to show how effectively Sevi inhibits AR activity and cell/tumor proliferation. Single cell RNA-sequencing of the AR+ cell line MDA-MB-453 illustrated heterogeneity in AR levels and identified cell cycle pathway activation in ARHigh versus ARLow expressing cells. Combination treatment with the cell cycle CDK4/6 inhibitor abemaciclib and Sevi showed synergy in AR+ TNBC cells and increased effectiveness, compared to each drug alone, in an AR+ TNBC xenograft model. While cell cycle inhibitors have been approved for use in hormone-receptor positive breast cancer, our studies suggest that these drugs may be equally effective in AR+ TNBC cells, especially when combined with AR antagonists. Implications. These data suggest that targeting AR signaling at multiple points throughout the pathway may be an effective was to treat patients with AR+ TNBC.

Effect of Selective Androgen Receptor Modulator (SARM) on Gene Expression in Triple-Negative Breast Cancer Cells, MDA-MB-231-AR

Project description:RNA from tumors treated with vehicle or 30 mg/kg GTx-027 were pooled and subjected to microarray analysis. Genes that were increased or decreased by 2-fold or more were considered for further analyses. Unlike in prostate cancer, where AR agonists induce more genes than they repress, in MDA-MB-231-AR tumors, GTx-027 inhibited 2.5X the number of genes (1092 vs. 456) than it activated. Functional clustering of the genes indicated that GTx-027 modified more breast cancer genes than other pathway genes. Genes that regulate the function of others cancers, such as colorectal, lung, and oral, and metabolic diseases were also favorably altered by GTx-027. Breast cancer proliferative genes, such as aurora kinase, ERCC1, IGFBP3 were inhibited and growth inhibitory genes, such as NQO1, PTPRJ were activated by GTx-027. Many of the established androgen responsive-genes were also activated by GTx-027, indicating that breast cancer growth inhibitory role of GTx-027 evolved from its agonistic activity. A mixture of cells was suspended in 0.05 ml RPMI+10% FBS and 0.05 ml Matrigel/animal and was injected subcutaneously. Once the tumor size reached 200-300 mm3, the animals were randomized and treated orally with the indicated drugs formulated in Tween 80:Captex 200:water (0.8:0.2:9). Tumor volume and body weight were measured thrice weekly. At the end of study, animals were sacrificed, tumors excised, weighed, and stored for various analyses. RNA from tumors was isolated and verified qualitatively and quantitatively. Samples from each group (n=8; 100 ng/µl; total 1000 ng) were pooled and hybridized to Affymetrix human ST2.0 gene array. Data from the array were analyzed using Ingenuity Pathway Analysis software (IPA3).

Elevated Production of Mitochondrial Reactive Oxygen Species via Hyperthermia Enhanced Cytotoxic Effect of Doxorubicin in Human Breast Cancer Cell Lines MDA-MB-453 and MCF-7.

Project description:Hyperthermia (HT) treatment is a noninvasive cancer therapy, often used with radiation therapy and chemotherapy. Compared with 37 °C, 42 °C is mild heat stress for cells and produces reactive oxygen species (ROS) from mitochondria. To involve subsequent intracellular accumulation of DOX, we have previously reported that the expression of ATP-binding cassette sub-family G member 2 (ABCG2), an exporter of doxorubicin (DOX), was suppressed by a larger amount of intracellular mitochondrial ROS. We then hypothesized that the additive effect of HT and chemotherapy would be induced by the downregulation of ABCG2 expression via intracellular ROS increase. We used human breast cancer cell lines, MCF-7 and MDA-MB-453, incubated at 37 °C or 42 °C for 1 h to clarify this hypothesis. Intracellular ROS production after HT was detected via electron spin resonance (ESR), and DOX cytotoxicity was calculated. Additionally, ABCG2 expression in whole cells was analyzed using Western blotting. We confirmed that the ESR signal peak with HT became higher than that without HT, indicating that the intracellular ROS level was increased by HT. ABCG2 expression was downregulated by HT, and cells were injured after DOX treatment. DOX cytotoxicity enhancement with HT was considered a result of ABCG2 expression downregulation via the increase of ROS production. HT increased intracellular ROS production and downregulated ABCG2 protein expression, leading to cell damage enhancement via DOX.
