Project description:Cadmium (Cd2+) is considered a human carcinogen as it causes oxidative stress and alters DNA repair responses. However, how Cd2+ is taken up by cells remains unclear. We hypothesized that Cd2+ could be transported into cells via a membrane copper (Cu) transporter, CTR1. CTR1 expression was not affected by Cd2+ exposure at the mRNA or protein level. Stable cell lines overexpressing either hCTR1, in the human liver cell line HepG2, or zCTR1, in the zebrafish liver cell line ZFL, were created to study their responses to Cd2+ insult. It was found that both HepG2 and ZFL cells overexpressing CTR1 had higher Cd2+ uptake and thus became sensitive to Cd2+. In contrast, hCTR1 knockdown in HepG2 cells led to a reduced uptake of Cd2+, making the cells relatively resistant to Cd2+. Localization studies revealed that hCTR1 had a clustered pattern after Cd2+ exposure, possibly in an attempt to reduce both Cd2+ uptake and Cd2+-induced toxicity. These in vitro results indicate that CTR1 can transport Cd2+ into the cell, resulting in Cd2+ toxicity.

Project description:Greenhouse hydroponic experiments were conducted using Cd-sensitive (cv. Guiyan1) and Cd-tolerant (cv. Yunyan2) tobacco cultivars to study the ameliorative effects of exogenous glycinebetaine (GB) upon 5 μM Cd stress. The foliar spray of GB markedly reduced Cd concentrations in plants and alleviated Cd-induced soil plant analysis development (SPAD) value, plant height and root length inhibition, with the mitigation effect being more obvious in Yunyan2. External GB markedly reduced Cd-induced malondialdehyde (MDA) accumulation, induced stomatal closure, ameliorated Cd-induced damages on leaf/root ultrastructure, and increased the chlorophyll content and fluorescence parameters of Fo, Fm, and Fv/Fm in both cultivars and Pn in Yunyan2. Exogenous GB counteracted Cd-induced alterations of certain antioxidant enzymes and nutrients uptake, e.g., the depressed Cd-induced increase of superoxide dismutase (SOD) and peroxidase (POD) activities, but significantly elevated the depressed catalase (CAT) and ascorbate peroxidase (APX) activities. The results indicate that alleviated Cd toxicity by GB application is related to the reduced Cd uptake and MDA accumulation, balanced nutrients and antioxidant enzyme activities, improved PSII, and integrated ultrastructure in tobacco plants.

Project description:Cadmium is a toxic heavy metal that may be detected in soils and plants. Wheat, as a food consumed by 60% of the world's population, may uptake a high quantity of Cd through its roots and translocate Cd to the shoots and grains thus posing risks to human health. Therefore, we tried to explore the journey of Cd in wheat via a review of several papers. Cadmium may reach the root cells by some transporters (such as zinc-regulated transporter/iron-regulated transporter-like protein, low-affinity calcium transporters, and natural resistance-associated macrophages), and some cation channels or Cd chelates via yellow stripe 1-like proteins. In addition, some of the effective factors regarding Cd uptake into wheat, such as pH, organic matter, cation exchange capacity (CEC), Fe and Mn oxide content, and soil texture (clay content), were investigated in this paper. Increasing Fe and Mn oxide content and clay minerals may decrease the Cd uptake by plants, whereas reducing pH and CEC may increase it. In addition, the feasibility of methods to diminish Cd accumulation in wheat was studied. Amongst agronomic approaches for decreasing the uptake of Cd by wheat, using organic amendments is most effective. Using biochar might reduce the Cd accumulation in wheat grains by up to 97.8%.

Project description:The uptake of cadmium by isolated liver cells was linearly related to the cadmium concentration to which the cells were exposed in the medium. Cadmium-treated cells synthesized proteins de novo with the characteristics of cadmium-thionein induced in the liver of cadmium-treated animals. Thionein from liver cells incorporated cadmium and [35S]cysteine, had a Ve/Vo (Sephadex G-50) of 1.8-1.9, and was separated into two subfractions by DEAE-cellulose ion-exchange chromatography. Cycloheximide and actinomycin D when added after a cadmium exposure prevented the synthesis of thionein. However, addition of actinomycin D after synthesis had started only decreased the total amount of thionein synthesized. The concentration of cadmium to which the cells were exposed affected the amount of cadmium-thionein synthesized in 6h. The maximum response occurred when cells were exposed to 0.5 microgram of cadmium/ml; at higher metal concentrations the total amount of cadmium-thionein synthesized declined. The system described in the present paper can be used to study the mode of metal toxicity and the mechanism of cadmium-thionein synthesis.

Project description:The metabolism of cadmium was investigated in Wistar-rat liver non-parenchymal cells. Kupffer and endothelial cells, the major cell populations lining the sinusoidal tracts, were isolated by collagenase dispersion and purified by centrifugal elutriation. At 20 h after subcutaneous injection of the metal salt (1.5 mg of Cd/kg body weight), endothelial cells accumulated 2-fold higher concentrations of Cd than did Kupffer or parenchymal cells. Most of the Cd in non-parenchymal cells was associated with cytosolic metallothionein (MT), the low-Mr heavy-metal-binding protein(s). When MT was quantified in cytosols from cells isolated from control rats by a 203Hg competitive-binding assay, low levels were found to be present in Kupffer, endothelial and parenchymal cells. Cd injection significantly increased MT levels in all three cell types. The induction of MT synthesis was investigated in vitro by using primary monolayer cultures. The incorporation of [35S]cysteine into MT increased 47% over constitutive levels in endothelial-cell cultures after the addition of 0.8 microM-Cd2+ to the medium for 10 h. MT synthesis in Kupffer cells was not observed. The lack of MT synthesis by monolayer cultures of Kupffer cells in vitro was associated with a decreased capacity of these cells to accumulate heavy metals from the extracellular medium. This apparent decreased ability to transport metals did not reflect a general defect in either cellular function or metabolic activity, since isolated Kupffer cells incorporated [3H]leucine into protein at rates comparable with those shown by liver parenchymal cells and readily phagocytosed particles.

Project description:The application of phosphorus-solubilizing bacteria is an effective method for increasing the available phosphorus content and inhibiting wheat uptake of heavy metals. However, further research is needed on the mechanism by which phosphorus-solubilizing bacteria inhibit cadmium (Cd) uptake in wheat roots and its impact on the expression of root-related genes. Here, the effects of strain Klebsiella aerogenes M2 on Cd absorption in wheat and the expression of root-related Cd detoxification and immobilization genes were determined. Compared with the control, strain M2 reduced (64.1-64.6%) Cd uptake by wheat roots. Cd fluorescence staining revealed that strain M2 blocked the entry of exogenous Cd into the root interior and enhanced the immobilization of Cd by cell walls. Forty-seven genes related to Cd detoxification, including genes encoding peroxidase, chalcone synthase, and naringenin 3-dioxygenase, were upregulated in the Cd+M2 treatment. Strain M2 enhanced the Cd resistance and detoxification activity of wheat roots through the regulation of flavonoid biosynthesis and antioxidant enzyme activity. Moreover, strain M2 regulated the expression of genes related to phenylalanine metabolism and the MAPK signaling pathway to enhance Cd immobilization in roots. These results provide a theoretical basis for the use of phosphorus-solubilizing bacteria to remediate Cd-contaminated fields and reduce Cd uptake in wheat.

Project description:Increasingly cancer is being viewed as a channelopathy because the passage of ions via ion channels and transporters mediate the regulation of tumor cell survival, death, and motility. As a result, a potential targeted therapy for cancer is to use venom peptides that are selective for ion channels and transporters overexpressed in tumor cells. Here we describe the selectivity and mechanism of action of terebrid snail venom peptide, Tv1, for treating the most common type of liver cancer, hepatocellular carcinoma (HCC). Tv1 inhibited the proliferation of murine HCC cells and significantly reduced tumor size in Tv1-treated syngeneic tumor-bearing mice. Tv1's mechanism of action involves binding to overexpressed transient receptor potential (TRP) channels leading to calcium dependent apoptosis resulting from down-regulation of cyclooxygenase-2 (COX-2). Our findings demonstrate the importance of modulating ion channels and the unique potential of venom peptides as tumor specific ligands in the quest for targeted cancer therapies.

Project description:AIM:To explore the biological behaviors of hepatic oval cells after transfused into the circulation of experimental animals. METHODS:Oval cells from male SD rat were transfused into the circulation of a female rat which were treated by a 2-AAF/CCl(4) program, through caudal vein. Sex-determining gene sry which located on Y chromosome was examined by PCR and in situ hybridization technique in liver, kidney and spleen of the experimental animals, respectively. RESULTS:The results of the cell-transplant experiment showed that the sry gene was detectable only in the liver but not in spleen and kidney of the experimental rats, and no signals could be detected in the control animals. It can be also morphologically proved that some exogenous cells had migrated into the parenchyma of the liver and settled there. CONCLUSION:The result means that there are exogenous cells located in the liver of the experimental animal and the localization is specific to the liver. This indicates that some "signal molecules" must exist in the circulation of the rats treated by 2-AAF/CCl(4). These "signal molecules" might play an important role in specific localization and differentiation of transfused oval cells.

Project description:BackgroundConcentrations of cadmium (Cd) in the grain of many durum wheats (Triticum turgidum subsp. durum) grown in North American prairie soils often exceed international trade standards. Genotypic differences in root-to-shoot translocation of Cd are a major determinant of intraspecific variation in the accumulation of Cd in grain. We tested the extent to which changes in whole-plant Cd accumulation and the distribution of Cd between tissues influences Cd accumulation in grain by measuring Cd accumulation throughout the grain filling period in two near-isogenic lines (NILs) of durum wheat that differ in grain Cd accumulation.ResultsRoots absorbed Cd and transported it to the shoots throughout the grain filling period, but the low- and high-Cd NILs did not differ in whole-plant Cd uptake. Although the majority of Cd accumulation was retained in the roots, the low- and high-Cd NILs differed substantively in root-to-shoot translocation of Cd. At grain maturity, accumulation of Cd in the shoots was 13% (low-Cd NIL) or 37% (high-Cd NIL) of whole-plant Cd accumulation. Accumulation of Cd in all shoot tissue, including grain, was at least 2-fold greater in the high-Cd NIL at all harvests. There was no net remobilization of shoot Cd pools during grain filling. The timing of Cd accumulation in grain was positively correlated with grain biomass accumulation, and the rate of grain filling peaked between 14 and 28 days post-anthesis, when both NILs accumulated 60% of total grain biomass and 61-66% of total grain Cd content.ConclusionsThese results show that genotypic variation in root-to-shoot translocation of Cd controls accumulation of Cd in durum wheat grain. Continued uptake of Cd by roots and the absence of net remobilization of Cd from leaves during grain filling support a direct pathway of Cd transport from roots to grain via xylem-to-phloem transfer in the stem.
