Project description:Two bacterial strains phylogenetically identified as Pseudomonas aeruginosa strains RM1 and SK1 displayed extensive degradation ability on waste engine oil (SAE 40W) in batch cultures. Spectrophotometric analysis revealed the presence of various heavy metals such as lead, chromium and nickel in the waste engine oil. The rate of degradation of waste engine oil by the isolates, for the first 12 days and the last 9 days were 66.3, 31.6 mg l-1 day-1 and 69.6, 40.0 mg l-1 day-1 for strains RM1 and SK1, respectively. Gas chromatographic (GC) analyses of residual waste engine oil, revealed that 66.58, 89.06 % and 63.40, 90.75 % of the initial concentration of the waste engine oil were degraded by strains RM1 and SK1 within 12 and 21 days. GC fingerprints of the waste engine oil after 12 days of incubation of strains RM1 and SK1 showed total disappearance of C15, C23, C24, C25 and C26 hydrocarbon fractions as well as drastic reductions of C13, C14, C16 and PAHs fractions such as C19-anthracene and C22-pyrene. At the end of 21 days incubation, total disappearance of C17-pristane, C22-pyrene, one of the C19-anthracene and significant reduction of C18-phytane (97.2 %, strain RM1; 95.1 %, strain SK1) fractions were observed. In addition, <10 % of Day 0 values of medium fraction ranges C13, and C16 were discernible after 21 days. This study has established the potentials of P. aeruginosa strains RM1 and SK1 in the degradation of aliphatic, aromatic and branched alkane components of waste engine oils.

Project description:1. A comparison of rates of oxidation of various compounds by whole cells indicated that protocatechuate was a reaction intermediate when a non-fluorescent species of Pseudomonas oxidized p-cresol. In contrast, a fluorescent Pseudomonas oxidized 3-methylcatechol and 4-methylcatechol when grown with p-cresol, but did not oxidize protocatechuate. 2. Heat-treated extracts of the fluorescent Pseudomonas oxidized catechol, 3-methylcatechol and 4-methylcatechol to ring-fission products, the spectroscopic properties of which were recorded. Rates of enzymic degradation of these products were also measured. 3. Acetic acid and formic acid were obtained by the action of a Sephadex-treated extract on 3-methylcatechol and 4-methylcatechol respectively. In each case 0.8mol. of the carboxylic acid was formed from 1.0mol. of substrate. 4. Dialysed extracts converted 3-methylcatechol into acetaldehyde and pyruvate, with 4-hydroxy-2-oxovalerate as a reaction intermediate. 4-Methylcatechol was converted first into 4-hydroxy-2-oxohexanoate and then into propionaldehyde and pyruvate. 5. The ring-fission product of catechol was formed from phenol by a fluorescent Pseudomonas, that of 3-methylcatechol was formed from o-cresol and m-cresol, and the ring-fission product of 4-methylcatechol was given from p-cresol. Propionate was readily oxidized by these cells after growth with p-cresol, but this compound was not attacked when phenol, o-cresol or m-cresol served as source of carbon. 6. Cell extracts appeared to attack only one enantiomer of synthetic 4-hydroxy-2-oxohexanoate.

Project description:Pseudomonas sp. N.C.I.B. 9816 strain 11, when grown on salicylate in the presence of dibenzo[1,4]dioxan, accumulated cis-1,2-dihydroxy-1,2-dihydrodibenzo[1,4]dioxan and 2-hydroxydibenzo[1,4]dioxan in the culture medium. Each metabolite was isolated in crystalline form and identified by a variety of conventional chemical techniques. Crude cell extracts prepared from the parental strain grown with naphthalene oxidized cis-1,2-dihydroxy-1,2-dihydrodibenzo[1,4]dioxan under both aerobic and anaerobic conditions to 1,2-dihydroxydibenzo[1,4]dioxan. Further degradation of this metabolite was not detected.

Project description:Medicinal plants in genus Lagerstroemia were investigated for phytochemical contents by GC-MS and HPLC with ethanol and hexane extracts and their toxicity MTT and comet assay on human peripheral blood mononuclear cells (PBMCs). ?-Sitosterol is the major component found in all species at 14.70-34.44%. All of the extracts, except for L. speciosa ethanol extract, showed high percentages of cell viability. The IC50 value, 0.24?mg/mL, of ethanol L. speciosa extract predicted an LD50 of 811.78?mg/kg, which belongs to WHO Class III of toxic chemicals. However, in-depth toxicity evaluation by the comet assay showed that the four tested species induced significant (p < 0.05) DNA damage in PBMCs. ?-Sitosterol was previously reported to possess antihyperglycemic activity by increasing insulin secretion in response to glucose. Nonetheless, consumers should consider its toxicity, and the amount of consumption should be of concern.

When NAD(+) was present, cell extracts of Pseudomonas (A) grown with d-glucarate or galactarate converted 1mol. of either substrate into 1mol. each of 2-oxoglutarate and carbon dioxide; 70-80% of the gas originated from C-1 of the hexarate. 2. The enzyme system that liberated carbon dioxide from galactarate was inactive in air and was stabilized by galactarate or Fe(2+) ions; the system that acted on d-glucarate was more stable and was stimulated by Mg(2+) ions. 3. When NAD(+) was not added, 2-oxoglutarate semialdehyde accumulated from either substrate. This compound was isolated as its bis-2,4-dinitrophenylhydrazone, and several properties of the derivative were compared with those of the chemically synthesized material. Methods were developed for the determination of 2-oxoglutarate semialdehyde. 4. Synthetic 2-oxoglutarate semialdehyde was converted into 2-oxoglutarate by an enzyme that required NAD(+); the reaction rate with NADP(+) was about one-sixth of that with NAD(+). 5. For extracts of Pseudomonas (A) grown with d-glucarate or galactarate, or for those of Pseudomonas fragi grown with l-arabinose or d-xylose, specific activities of 2-oxoglutarate semialdehyde-NAD oxidoreductase were much higher than for extracts of the organisms grown with (+)-tartrate and d-glucose respectively. 6. Extracts of Pseudomonas fragi grown with l-arabinose or d-xylose converted l-arabonate or d-xylonate into 2-oxoglutarate when NAD(+) was added to reaction mixtures and into 2-oxoglutarate semialdehyde when NAD(+) was omitted.

Project description:A comparative genome analysis of the global anaerobic regulator Anr regulon in five species of Pseudomonas with different life style was performed. Expression of this regulator was detected in all analyzed Pseudomonas. The predicted Anr regulon (pan-regulon) consisted of 253 genes. However, only 11 Anr-boxes located upstream of qor/hemF, hemN, cioA/PA3931, azu, rpsL, gltP, orthologous to PA2867, cspD, tyrZ, slyD and oprG, were common to all species. Whole genome in silico prediction of metabolic pathways identified genes belonging to heme biosynthesis, cytochromes and Entner-Doudoroff pathway as members of Anr regulon in all strains. Extending genome analysis to 28 Pseudomonas spp. spanning all phylogenetic groups showed Anr-boxes conservation in genes related to these functions. When present, genes related to anaerobic metabolism were predicted to hold Anr-boxes. Focused on the genomes of eight P. aeruginosa isolates of diverse origins, we observed a conserved regulon, sharing nearly 80% of the genes, indicating its key role in this opportunistic pathogen. The results suggest that the core Anr regulon comprises genes involved in central metabolism and aerobic electron transport chain, whereas those genes related to anaerobic metabolism and other functions constitute the accessory Anr-regulon, thereby differentially contributing to the ecological fitness of each Pseudomonas species.
