Project description:Plastics, in all forms, are a ubiquitous cornerstone of modern civilization. Although humanity undoubtedly benefits from the versatility and durability of plastics, they also cause a tremendous burden for the environment. Bio-upcycling is a promising approach to reduce this burden, especially for polymers that are currently not amenable to mechanical recycling. Wildtype P. putida KT2440 is able to grow on 1,4-butanediol as sole carbon source, but only very slowly. Adaptive laboratory evolution (ALE) led to the isolation of several strains with significantly enhanced growth rate and yield. Genome re-sequencing and proteomic analysis were applied to characterize the genomic and metabolic basis of efficient 1,4-butanediol metabolism. Initially, 1,4-butanediol is oxidized to 4-hydroxybutyrate, in which the highly expressed dehydrogenase enzymes encoded within the PP_2674-2680 ped gene cluster play an essential role. The resulting 4-hydroxybutyrate can be metabolized through three possible pathways: (i) oxidation to succinate, (ii) CoA activation and subsequent oxidation to succinyl-CoA, and (iii) beta oxidation to glycolyl-CoA and acetyl-CoA. The evolved strains were both mutated in a transcriptional regulator (PP_2046) of an operon encoding both beta-oxidation related genes and an alcohol dehydrogenase. When either the regulator or the alcohol dehydrogenase is deleted, no 1,4-butanediol uptake or growth could be detected. Using a reverse engineering approach, PP_2046 was replaced by a synthetic promotor (14g) to overexpress the downstream operon (PP_2047-2051), thereby enhancing growth on 1,4-butanediol. This work provides a deeper understanding of microbial 1,4-butanediol metabolism in P. putida, which is also expandable to other aliphatic alpha-omega diols. It enables the more efficient metabolism of these diols, thereby enabling biotechnological valorization of plastic monomers in a bio-upcycling approach.

Project description:In the title compound, C(17)H(18)N(2)S, the thia-zepine ring adopts a boat conformation and the dihedral angle between the benzene rings is 75.92?(5)°, resulting in a butterfly-like conformation. In the crystal, mol-ecules are connected via weak C(aromatic)-H?N contacts involving the imine N atom as acceptor and through a quite short C-H?? inter-action. The resulting mol-ecular chains propagate along the c-axis direction.

Project description:Two bacterial strains phylogenetically identified as Pseudomonas aeruginosa strains RM1 and SK1 displayed extensive degradation ability on waste engine oil (SAE 40W) in batch cultures. Spectrophotometric analysis revealed the presence of various heavy metals such as lead, chromium and nickel in the waste engine oil. The rate of degradation of waste engine oil by the isolates, for the first 12 days and the last 9 days were 66.3, 31.6 mg l-1 day-1 and 69.6, 40.0 mg l-1 day-1 for strains RM1 and SK1, respectively. Gas chromatographic (GC) analyses of residual waste engine oil, revealed that 66.58, 89.06 % and 63.40, 90.75 % of the initial concentration of the waste engine oil were degraded by strains RM1 and SK1 within 12 and 21 days. GC fingerprints of the waste engine oil after 12 days of incubation of strains RM1 and SK1 showed total disappearance of C15, C23, C24, C25 and C26 hydrocarbon fractions as well as drastic reductions of C13, C14, C16 and PAHs fractions such as C19-anthracene and C22-pyrene. At the end of 21 days incubation, total disappearance of C17-pristane, C22-pyrene, one of the C19-anthracene and significant reduction of C18-phytane (97.2 %, strain RM1; 95.1 %, strain SK1) fractions were observed. In addition, <10 % of Day 0 values of medium fraction ranges C13, and C16 were discernible after 21 days. This study has established the potentials of P. aeruginosa strains RM1 and SK1 in the degradation of aliphatic, aromatic and branched alkane components of waste engine oils.

Project description:Fermentation of sitosterol by a Pseudomonas species (SK-25) resulted in the formation of 5-stigmastene-3 beta, 7 alpha-diol; 5,6 alpha-epoxy-5 alpha-stigmastan-3 beta-ol; 5,6 beta-epoxy-5 beta-stigmastan-3 beta-ol and 5 alpha-stigmastan-3 beta, 5,6 beta-triol. The metabolites were characterized by a variety of conventional chemical and spectrometric techniques.

Project description:1. A comparison of rates of oxidation of various compounds by whole cells indicated that protocatechuate was a reaction intermediate when a non-fluorescent species of Pseudomonas oxidized p-cresol. In contrast, a fluorescent Pseudomonas oxidized 3-methylcatechol and 4-methylcatechol when grown with p-cresol, but did not oxidize protocatechuate. 2. Heat-treated extracts of the fluorescent Pseudomonas oxidized catechol, 3-methylcatechol and 4-methylcatechol to ring-fission products, the spectroscopic properties of which were recorded. Rates of enzymic degradation of these products were also measured. 3. Acetic acid and formic acid were obtained by the action of a Sephadex-treated extract on 3-methylcatechol and 4-methylcatechol respectively. In each case 0.8mol. of the carboxylic acid was formed from 1.0mol. of substrate. 4. Dialysed extracts converted 3-methylcatechol into acetaldehyde and pyruvate, with 4-hydroxy-2-oxovalerate as a reaction intermediate. 4-Methylcatechol was converted first into 4-hydroxy-2-oxohexanoate and then into propionaldehyde and pyruvate. 5. The ring-fission product of catechol was formed from phenol by a fluorescent Pseudomonas, that of 3-methylcatechol was formed from o-cresol and m-cresol, and the ring-fission product of 4-methylcatechol was given from p-cresol. Propionate was readily oxidized by these cells after growth with p-cresol, but this compound was not attacked when phenol, o-cresol or m-cresol served as source of carbon. 6. Cell extracts appeared to attack only one enantiomer of synthetic 4-hydroxy-2-oxohexanoate.

The reaction pathways were controlled by varying enaminones with different substituted patterns to give a series of new fused pyrroles and dibenzo[b,e][1,4]diazepin-1-ones selectively. The configuration of the newly formed stereogenic center at the point of attachment of the 1,4-dioxane ring to the aziridine ring is S. The configurations of the pre-existing sites 2-, 5-, and 6-positions of the 1,4-dioxane ring prior to reaction of sulfinyl imine with the sulfur ylide are S, R, and R, respectively. The C-N bond lengths of the aziridine are 1.478?(2) and 1.486?(2)?Å.

Project description:1. 1,4-Epoxy-1,4-dihydronaphthalene is converted by rats into 1,4:2,3-diepoxy-1,2,3,4-tetrahydronaphthalene, which was isolated from the urine. The synthesis of the diepoxide is described. 2. The monoepoxide also yielded a compound that is believed to be 1,4-dihydro-1,4-dihydroxynaphthalene, but no corresponding mercapturic acid was detected. A number of unidentified metabolites of the monoepoxide were detected that appear to arise by the hydroxylation of the diepoxide. 3. The monoepoxide is converted into the diepoxide by a rat-liver microsomal system. 4. 1,4-Epoxy-1,4-dihydronaphthalene does not appear to be an intermediate in naphthalene metabolism.

When NAD(+) was present, cell extracts of Pseudomonas (A) grown with d-glucarate or galactarate converted 1mol. of either substrate into 1mol. each of 2-oxoglutarate and carbon dioxide; 70-80% of the gas originated from C-1 of the hexarate. 2. The enzyme system that liberated carbon dioxide from galactarate was inactive in air and was stabilized by galactarate or Fe(2+) ions; the system that acted on d-glucarate was more stable and was stimulated by Mg(2+) ions. 3. When NAD(+) was not added, 2-oxoglutarate semialdehyde accumulated from either substrate. This compound was isolated as its bis-2,4-dinitrophenylhydrazone, and several properties of the derivative were compared with those of the chemically synthesized material. Methods were developed for the determination of 2-oxoglutarate semialdehyde. 4. Synthetic 2-oxoglutarate semialdehyde was converted into 2-oxoglutarate by an enzyme that required NAD(+); the reaction rate with NADP(+) was about one-sixth of that with NAD(+). 5. For extracts of Pseudomonas (A) grown with d-glucarate or galactarate, or for those of Pseudomonas fragi grown with l-arabinose or d-xylose, specific activities of 2-oxoglutarate semialdehyde-NAD oxidoreductase were much higher than for extracts of the organisms grown with (+)-tartrate and d-glucose respectively. 6. Extracts of Pseudomonas fragi grown with l-arabinose or d-xylose converted l-arabonate or d-xylonate into 2-oxoglutarate when NAD(+) was added to reaction mixtures and into 2-oxoglutarate semialdehyde when NAD(+) was omitted.
