Project description:Fluoroacetate (FA) is a tasteless, odorless, water-soluble metabolic poison with severe toxicological effects. Characterized in the mid-1900s, it has been used as a rodenticide but is comparably lethal to all mammals. Many countries have restricted its use, and modern-day accidental human exposures are rare, but recently, concerns have been raised about its application as a chemical weapon with no known antidote. A combined treatment of methylene blue (MB), an antioxidant, and monosodium glutamate (MSG), a precursor of the citric acid cycle substrate alpha-ketoglutarate, has been recommended as an effective countermeasure; however, no peer-reviewed articles documenting the efficacy of this therapy have been published. Using a rodent model, we assessed the effects of MB and MSG on the neurologic, cardiac, and pulmonary systems. Transcriptomic analysis was used to elucidate inflammatory pathway activation and guide bioassays, which revealed the advantages and disadvantages of these candidate countermeasures. Results show that MB and MSG can reduce neurologic signs observed in rats exposed to sodium FA and improve some effects of intoxication. However, while this strategy resolved some signs of intoxication, ultimately it was unable to significantly reduce lethality.

Project description:Recently, green-prepared oxidized graphenes have attracted huge interest in water purification and wastewater treatment. Herein, reduced graphene oxide (rGO) was prepared by a scalable and eco-friendly method, and its potential use for the removal of methylene blue (MB) from water systems, was explored. The present work includes the green protocol to produce rGO and respective spectroscopical and morphological characterizations, as well as several kinetics, isotherms, and thermodynamic analyses to successfully demonstrate the adsorption of MB. The pseudo-second-order model was appropriated to describe the adsorption kinetics of MB onto rGO, suggesting an equilibrium time of 30 min. Otherwise, the Langmuir model was more suitable to describe the adsorption isotherms, indicating a maximum adsorption capacity of 121.95 mg g-1 at 298 K. In addition, kinetics and thermodynamic analyses demonstrated that the adsorption of MB onto rGO can be treated as a mixed physisorption-chemisorption process described by H-bonding, electrostatic, and ? - ? interactions. These results show the potential of green-prepared rGO to remove cationic dyes from wastewater systems.

Project description:Density functional theory and a semi-empirical quantum chemical approach were used to evaluate the photocatalytic efficiency of ceria (CeO2) combined with reduced graphene oxide (rGO) and graphene (GP) for degrading methylene blue (MB). Two main aspects were examined: the adsorption ability of rGO and GP for MB, and the separation of photogenerated electrons and holes in CeO2/rGO and CeO2/GP. Our results, based on calculations of the adsorption energy, population analysis, bond strength index, and reduced density gradient, show favorable energetics for MB adsorption on both rGO and GP surfaces. The process is driven by weak, non-covalent interactions, with rGO showing better MB adsorption. A detailed analysis involving parameters like fractional occupation density, the centroid distance between molecular orbitals, and the Lewis acid index of the catalysts highlights the effective charge separation in CeO2/rGO compared to CeO2/GP. These findings are crucial for understanding photocatalytic degradation mechanisms of organic dyes and developing efficient photocatalysts.

Project description:Methylene blue (MB) is a synthetic phenothiazine dye that, in the last years, has generated much debate about whether it could be a useful therapeutic drug for tau-related pathologies, such as Alzheimer's disease (AD). However, the molecular mechanism of action is far from clear. Recently we reported that MB activates the plasma membrane Ca2+-ATPase (PMCA) in membranes from human and pig tissues and from cells cultures, and that it could protect against inactivation of PMCA by amyloid ?-peptide (A?). The purpose of the present study is to further examine whether the MB could also modulate the inhibitory effect of tau, another key molecular marker of AD, on PMCA activity. By using kinetic assays in membranes from several tissues and cell cultures, we found that this phenothiazine was able to block and even to completely reverse the inhibitory effect of tau on PMCA. The results of this work point out that MB could mediate the toxic effect of tau related to the deregulation of calcium homeostasis by blocking the impairment of PMCA activity by tau. We then could conclude that MB could interfere with the toxic effects of tau by restoring the function of PMCA pump as a fine tuner of calcium homeostasis.

Project description:The synthesis of new phenothiazine derivatives, analogs of Methylene Blue, is of particular interest in the design of new drugs, as well as in the development of a new generation of agents for photodynamic therapy. In this study, two new derivatives of phenothiazine, i.e., 3,7-bis(4-aminophenylamino)phenothiazin-5-ium chloride dihydrochloride (PTZ1) and 3,7-bis(4-sulfophenylamino)phenothiazin-5-ium chloride (PTZ2), are synthesized for the first time and characterized by NMR, IR spectroscopy, HRMS and elemental analysis. The interaction of the obtained compounds PTZ1 and PTZ2 with salmon sperm DNA is investigated. It is shown by UV-Vis spectroscopy and DFT calculations that substituents in arylamine fragments play a crucial role in dimer formation and interaction with DNA. In the case of PTZ1, two amine groups promote H-aggregate formation and DNA interactions through groove binding and intercalation. In the case of PTZ2, sulfanilic acid fragments prevent any dimer formation and DNA binding due to electrostatic repulsion. DNA interaction mechanisms are studied and confirmed by UV-vis and fluorescence spectroscopy in comparison with Methylene Blue. The obtained results open significant opportunities for the development of new drugs and photodynamic agents.

Project description:A promising alternative to antibiotics for treatment of Staphylococcus aureus infections is photodynamic inactivation (PDI), which employs a photosensitizer (PS) that produces cytotoxic reactive oxygen species (ROS) when exposed to molecular oxygen and antimicrobial blue light in the spectrum of 400-470 nm. Although the precise mechanistic basis of PDI has not been defined, the formation of ROS and free radicals that oxidize a number of cellular targets, including membrane lipids, damage to proteins and nucleic acids result in inactivation of essential cellular functions and subsequent cell death. Because PDI is non-selective and affects multiple cellular targets, development of resistance or tolerance to PDI has been considered to be unlikely and attempts to induce S. aureus resistance or tolerance upon repeated sub-lethal doses of PDI have not succeeded. However, multiple aspects of PDI suggest that development of tolerance is highly probable, in particular when PDI is used for treatment of infections where the environment at the infection site prevents penetration of PDI at a level sufficient to cause death of all bacteria and with tolerant phenotypes emerging from the surviving bacteria. In this study, we sought to identify the mechanisms that contribute to PDI tolerance in S. aureus. S. aureus HG003 and the isogenic HG003DmutSL strain with defects in DNA mismatch repair were used to evaluate the response of S. aureus and the roles of DNA mismatch repair and gene regulatory networks to repeated sublethal doses of PDI. Global transcriptome and genome analyses were used in an agnostic approach to identify the underlying transcriptional responses and genetic adaptations that occur as a result of repeated PDI and contribute to PDI tolerance. Our results reveal multiple metabolic, transport and cell wall biogenesis pathways that contribute to PDI tolerance, and a S. aureus regulatory gene likely responsible for the adaptive transcriptional response to PDI.

Project description:In this work, reduced graphene oxide (rGO) was fabricated at different reduction temperatures via an environmentally friendly solvothermal approach. The rGO formed at 160 °C clearly showed the partial restoration of the sp2 hybridization brought about by the elimination of oxygenated functionalities from the surface. Owing to the augmented surface area and the band gap reduction, rGO-160 exhibited the best adsorption (29.26%) and photocatalytic activity (32.68%) towards the removal of MB dye. The effects of catalyst loading, initial concentration of dye, light intensity, and initial pH of solution were evaluated. It was demonstrated that rGO-160 could achieve a higher adsorptive removal (87.39%) and photocatalytic degradation (98.57%) of MB dye when 60 mg of catalyst, 50 ppm of dye at pH 11, and 60 W m-2 of UV-C light source were used. The MB photodegradation activity of rGO-160 displayed no obvious decrease after five successive cycles. This study provides a potential metal-free adsorbent-cum-photocatalyst for the decontamination of dyes from wastewater.

Project description:Adhesion is a primary challenge following surgery, and the anti-adhesive effect of methylene blue (MB) has been investigated. This systematic review and meta-analysis aimed to evaluate the effect of MB on postoperative adhesions in experimental studies. We initially searched OVID-MEDLINE, EMBASE, and Google Scholar in February 2021, and then in May 2021. The anti-adhesive efficacy of MB was compared with that of the control (either placebo or nothing) after the surgical procedure. The primary and secondary outcomes were the macroscopic and microscopic adhesion scores, respectively. Traditional meta-analysis, meta-regression, and trial sequential analysis (TSA) were performed to analyze the retrieved outcomes. We included 13 experimental studies of 367 rats (200 rats received MB and 167 rats received placebo or nothing). The macroscopic adhesion scores were significantly lower in the MB-administered group than in the control group (standardized mean difference, 2.313; 95% confidence interval, 1.104 to3.523; I2 = 94.0%, Tau = 2.059). Meta-regression analysis showed that macroscopic adhesion tended to decrease with an increase in MB dose. TSA demonstrated that the cumulative Z curve crossed both the conventional test and trial sequential monitoring boundary for the macroscopic adhesion score. MB had a beneficial effect on intraperitoneal adhesion following laparotomy, and adhesions decreased with increase in dose.

Project description:UNLABELLED: BACKGROUND:We aimed to minimalize operative complications by spraying of methylene blue stain on thyroid glands and the perithyroidal area. MATERIAL AND METHODS:The intra-operative methylene blue spraying technique was used prospectively on a total of 56 patients who had undergone primary (not recurrent) thyroid surgery for a variety of thyroid diseases. Bilateral total thyroidectomy was performed in all cases. After superior but before inferior pole ligation, 0.5ml of methylene blue was sprayed over the thyroid lobe and perilober area. Tissues, especially parathyroides, the recurrent laryngeal nerve, and the inferior thyroid artery, were identified and evaluated. RESULTS:Recurrent laryngeal nerve and arteries were not stained and thus they remained white in all cases while all other tissues were stained blue. Within three minutes parathyroid glands washed out the blue stain and the original yellow color was regained. Thyroid tissue wash-out time was not less than 15 minutes; perithyroideal muscles, tendinous and lipoid structures took no less than 25 minutes. CONCLUSION:The safety of intravascular methylene blue guidance on thyroid surgery is known. This research demonstrates the effectiveness of the spraying technique, a new technique which ensures not only identification of parathyroid glands within three minutes, but also identification of recurrent laryngeal nerves and inferior thyroid arteries.

Project description:S and N co-doped reduced graphene (S-N-rGO) nanohybrids were prepared by a one-step oil bath heating process using glutathione (GSH) as a green and mild co-reduction agent and a S and N source. It can be applied in the field of adsorption for the removal of methylene blue (MB) from aqueous solutions. The efficient adsorption rate of S-N-rGO hybrids for MB (50 mg L-1) was observed with the best even within 2'07'' from blue solutions into colorless (the mass ratio GO : GSH = 60 : 200). Under this mass ratio, the effects of initial solution pH, temperature, initial concentration and contact time on adsorption towards MB were explored systematically. The results indicated that the adsorption capacity at 275 K could reach up to 598.8 mg g-1, the adsorption behavior followed the pseudo-second-order kinetic model and the equilibrium adsorption data fitted the Langmuir isotherm well. Thermodynamic and kinetic analyses revealed that adsorption is an exothermic, spontaneous and physisorption process.
