(+)-n-Propylmercapturic acid sulphoxide, i.e. (+)-N-acetyl-S-n-propyl-l-cysteine S-oxide, was prepared as the dicyclohexylammonium salt, (-)-n-propyl-mercapturic acid sulphoxide was prepared as the free acid, and S-isopropyl-l-cysteine and isopropylmercapturic acid were also prepared. 2. The metabolism of 1- and 2-bromopropane was studied by radiochromatographic examination of the urine excreted by rats that had been fed with a diet containing (35)S-labelled yeast and then injected subcutaneously with these compounds. In addition to n-propyl-mercapturic acid and 2-hydroxypropylmercapturic acid, the excretion of which has already been reported, n-propylmercapturic acid sulphoxide was shown to be a metabolite of 1-bromopropane. Sulphur-containing metabolites of 2-bromopropane, if present in the urine at all, were there in very small amounts. 3. n-Propylmercapturic acid and isopropylmercapturic acid were isolated from the urine of rats that had been injected subcutaneously with S-n-propyl-l-cysteine and S-isopropyl-l-cysteine respectively.

Project description:1. Various derivatives of 1- and 2-methylnaphthalene, of the type C(10)H(7).CH(2)X, were administered to rats and the urines of the dosed animals were examined for the presence of 1- and 2-menaphthylmercapturic acid by chromatographic and isolation procedures. A similar, but more limited, series of experiments was carried out with rabbits. 2. All the compounds were administered by subcutaneous injection with the exception of S-(1- and 2-menaphthyl)-l-cysteine, which were added to the food. 3. 1-Menaphthylmercapturic acid was isolated from the urine of rats after the administration of 1-menaphthyl chloride, bromide, alcohol, acetate and benzoate, S-(1-menaphthyl)-l-cysteine and S-(1-menaphthyl)glutathione. 4. 2-Menaphthylmercapturic acid was isolated from rat urine after administration of 2-menaphthyl chloride, S-(2-menaphthyl)-l-cysteine and S-(2-menaphthyl)-glutathione, and was detected chromatographically after injecting 2-menaphthyl bromide. 5. The corresponding mercapturic acids were isolated after administering 1- and 2-menaphthyl chloride and 1-menaphthyl acetate to rabbits, but not after giving 1- and 2-menaphthyl bromide and 1-menaphthyl alcohol, although chromatographic evidence of mercapturic acid excretion was obtained after injecting these compounds.

The metabolism of cis- and trans-acenaphthene-1,2-diol has been studied after the administration of these compounds to rats by subcutaneous injection and by stomach tube. 2. 1,8-Naphthalic acid has been isolated as its anhydride from the urine of the dosed animals. 3. A spectrophotometric method for the determination of free and conjugated 1,8-naphthalic acid in urine has been developed and has been used in the study of the metabolism of the acenaphthene-1,2-diols. 4. The urine of rats dosed with cis-acenaphthene-1,2-diol by subcutaneous injection was shown by paper chromatography to contain both cis- and trans-acenaphthene-1,2-diol. Similar findings were obtained after the subcutaneous injection of trans-acenaphthene-1,2-diol.

A premercapturic acid, i.e. a compound that yields a mercapturic acid when decomposed by acid, was isolated as a dicyclohexylammonium salt from the urine of rats and rabbits that had been dosed with bromobenzene. 2. Another premercapturic acid was isolated as its dicyclohexylammonium salt from the urine of rats that had been dosed with chlorobenzene. 3. When decomposed by acid, the premercapturic acid from the urine of animals dosed with bromobenzene gave p-bromophenylmercapturic acid, m-and p-bromophenol and NN'-diacetylcystine. 4. The premercapturic acid derived from chlorobenzene gave the corresponding chloro compounds together with NN'-diacetylcystine when decomposed by acid. 5. On the basis of these and other observations it is suggested that the premercapturic acid formed in the metabolism of bromobenzene is N-acetyl-S-(4-bromo-1,2-dihydro-2-hydroxyphenyl)-l-cysteine. 6. It is also suggested that the premercapturic acid derived from chlorobenzene has an analogous structure.

Project description:BackgroundLauric acid (LA), a common constituent of coconut oil, is used as food additives and supplements in various formulations. Despite various potential pharmacological properties, no scientific evidence on its dose-related toxicity and safety is available till date.ObjectiveThe current study was conducted to evaluate acute oral toxicity of LA on normal rats.MethodsThe study was conducted in accordance with the Organization for Economic Co-operation and Development guidelines (OECD 423) with slight modifications. LA was administered orally to female Sprague Dawley (SD) rats (n = 6/group) at a single dose of 300 and 2,000 mg/kg body weight, respectively, while normal control received vehicle only. Animals from all the three groups were monitored for any behavioural and toxicological changes and mortality for two weeks. Food and fluid consumption, body weight was monitored on daily basis. At the end (on day 15th) of the experimental period, blood was collected for haematological and biochemical analysis. Further, all the animals were euthanized, and internal organs were harvested for histopathological investigation using four different stainings; haematoxylin and eosin, Masson trichrome, Periodic Acid Schiff and Picro Sirius Red for gross pathology through microscopical observation.ResultsThe study results showed no LA treatment-related mortality and morbidity at two different dosages. Daily food and water consumption, body weight, relative organ weight, haematological, and biochemical analysis were observed to be normal with no severe alterations to the internal tissues.ConclusionThe current finding suggests that single oral administration of LA, even up to 2,000 mg/kg body weight, did not exhibit any signs of toxicity in SD rats; thus, it was safe to be used on disease models in animals.

Project description:Polyphenolic compounds, especially flavonoids, are known as the most common chemical class of phytochemicals, which possess a multiple range of health-promoting effects. Flavonoids are ubiquitous in nature. They are also present in food, providing an essential link between diet and prevention of several diseases.Chrysin (CH), a natural flavonoid, was commonly found in propolis and honey and traditionally used in herbal medicine. A growing body of scientific evidence has shown that CH possesses protective effects against toxic agents in various animal tissues, including brain, heart, liver, kidney, and lung.This study found that CH may be effective in disease management induced by toxic agents. However, due to the lack of information on human, further studies are needed to determine the efficacy of CH as an antidote agent in human.The present article aimed to critically review the available literature data regarding the protective effects of CH against toxic agent-induced toxicities as well as its possible mechanisms.

Project description:In the present study, we report the design and synthesis of new derivatives of the β-keto-enol grafted on pyridine and furan moieties (L 1 -L 11 ). Structures of compounds were fully confirmed by Fourier transform infrared spectroscopy (FT-IR), 1H NMR, 13C NMR, electrospray ionization/liquid chromatography-mass spectrometry (ESI/LC-MS), and elemental analysis. The compounds were screened for antifungal and antibacterial activities (Escherichia coli, Bacillus subtilis, and Micrococcus luteus). In vitro evaluation showed significant fungicidal activity for L 1 , L 4 , and L 5 against fungal strains (Fusarium oxysporum f.sp albedinis) compared to the reference standard. Especially, the exceptional activity has been demonstrated for L 1 with IC50 = 12.83 μg/mL. This compound and the reference benomyl molecule also showed a correlation between experimental antifungal activity and theoretical predictions by Petra/Osiris/Molinspiration (POM) calculations and molecular coupling against the Fgb1 protein. The highest inhibition of bacterial growth for L 1 is due to its strongest binding to the target protein. This report may stimulate the further synthesis of examples of this substance class for the development of new drugs.

Project description:Background4-hydroxy-2-oxoglutarate (HOG) aldolase is a unique enzyme in the hydroxyproline degradation pathway catalyzing the cleavage of HOG to pyruvate and glyoxylate. Mutations in this enzyme are believed to be associated with the excessive production of oxalate in primary hyperoxaluria type 3 (PH3), although no experimental data is available to support this hypothesis. Moreover, the identity, oligomeric state, enzymatic activity, and crystal structure of human HOGA have not been experimentally determined.Methodology/principal findingsIn this study human HOGA (hHOGA) was identified by mass spectrometry of the mitochondrial enzyme purified from bovine kidney. hHOGA performs a retro-aldol cleavage reaction reminiscent of the trimeric 2-keto-3-deoxy-6-phosphogluconate aldolases. Sequence comparisons, however, show that HOGA is related to the tetrameric, bacterial dihydrodipicolinate synthases, but the reaction direction is reversed. The 1.97 Å resolution crystal structure of hHOGA bound to pyruvate was determined and enabled the modeling of the HOG-Schiff base intermediate and the identification of active site residues. Kinetic analyses of site-directed mutants support the importance of Lys196 as the nucleophile, Tyr168 and Ser77 as components of a proton relay, and Asn78 and Ser198 as unique residues that facilitate substrate binding.Conclusions/significanceThe biochemical and structural data presented support that hHOGA utilizes a type I aldolase reaction mechanism, but employs novel residue interactions for substrate binding. A mapping of the PH3 mutations identifies potential rearrangements in either the active site or the tetrameric assembly that would likely cause a loss in activity. Altogether, these data establish a foundation to assess mutant forms of hHOGA and how their activity could be pharmacologically restored.

Project description:Bedaquiline is a novel drug approved in 2012 by the FDA for treatment of drug-resistant tuberculosis (TB). Although it shows high efficacy towards drug-resistant forms of TB, its use has been limited by the potential for significant side effects. In particular, bedaquiline is a very lipophilic compound with an associated long terminal half-life and shows potent inhibition of the cardiac potassium hERG channel, resulting in QTc interval prolongation in humans that may result in cardiac arrhythmia. To address these issues, we carried out a drug discovery programme to develop an improved second generation analogue of bedaquiline. From this medicinal chemistry program, a candidate (TBAJ-876) has been selected to undergo further preclinical evaluation. During this evaluation, three major metabolites arising from TBAJ-876 were observed in several preclinical animal models. We report here our synthetic efforts to unequivocally structurally characterize these three metabolites through their independent directed synthesis.

Project description:Our research sought to determine the molecular and biochemical effects of environmentally relevant exposure to commonly used chloro-s-triazine herbicide terbuthylazine and organophosphate insecticide malathion on zebrafish. To this aim, mature zebrafish were exposed to 2 and 30 µg L-1 terbuthylazine and 5 and 50 µg L-1 malathion alone and in combination for 14 days. Aside from the accumulation of TBARS and protein carbonyls, a decrease in antioxidants and succinate dehydrogenase activity, an increase in oxidized glutathione, and enhanced apoptosis via Caspase-3 and BAX overexpression were observed. Furthermore, terbuthylazine and malathion induced mitochondrial swelling (up to 210% after single exposure and up to 470% after co-exposure) and lactate dehydrogenase leakage (up to 268% after single exposure and up to 570% after co-exposure) in a concentration-dependent manner. Significant upregulation of ubiquitin expression and increased cathepsin D activity were characteristics that appeared only upon terbuthylazine exposure, whereas the induction of IgM was identified as the specific characteristic of malathion toxicity. Meanwhile, no alterations in the zebrafish hypothalamic-pituitary-thyroid axis was observed. Co-exposure increased the adverse effects of individual pesticides on zebrafish. This study should improve the understanding of the mechanisms of pesticide toxicity that lead to fish impairment and biodiversity decline.
