Biochemical studies of toxic agents. Metabolic ring-fission of cis- and trans-acenaphthene-1,2-diol.

ABSTRACT: 1. The metabolism of cis- and trans-acenaphthene-1,2-diol has been studied after the administration of these compounds to rats by subcutaneous injection and by stomach tube. 2. 1,8-Naphthalic acid has been isolated as its anhydride from the urine of the dosed animals. 3. A spectrophotometric method for the determination of free and conjugated 1,8-naphthalic acid in urine has been developed and has been used in the study of the metabolism of the acenaphthene-1,2-diols. 4. The urine of rats dosed with cis-acenaphthene-1,2-diol by subcutaneous injection was shown by paper chromatography to contain both cis- and trans-acenaphthene-1,2-diol. Similar findings were obtained after the subcutaneous injection of trans-acenaphthene-1,2-diol.

Project description:1. The ring-fission of cis- and trans-acenaphthene-1,2-diol by rat liver microsomes was studied. 2. 1,8-Naphthalic acid was detected and isolated after microsomal incubations of the diols. 3. The accompanying reduction of NAD(+) was followed spectrophotometrically. 4. The optimum pH for the microsomal reaction was 9.4 for the oxidation of the cis-diol and 9.8 for that of the trans-diol. 5. p-Chloromercuribenzoate and 2,4-dichlorophenol inhibited the reaction. 6. Possible mechanisms for the microsomal ring-fission, involving 1,8-naphthalic aldehyde, are discussed.

The metabolism of cis- and trans-indane-1,2-diol.

Project description:1. The metabolism of cis-indane-1,2-diol, trans-indane-1,2-diol, indene epoxide and 2-hydroxyindan-1-one in rats has been studied. The substances were administered to the animals by subcutaneous injection. 2. The urine of the dosed animals was examined for the presence of free and conjugated cis- and trans-dihydrodiols, and for each compound it was possible to isolate both cis and trans forms of indane-1,2-diol from the urine. 3. The urines were also examined by paper chromatography for ketones and two ketonic metabolites were detected in the urine of rats dosed separately with cis-indane-1,2-diol, trans-indane-1,2-diol, 2-hydroxyindan-1-one and indene epoxide. The ketones were provisionally identified as (1-oxoindan-2-yl glucosid)uronic acid and 1-oxoindan-2-yl sulphuric acid. 4. (1-Oxoindan-2-yl glucosid)uronic acid was isolated as the 2,4-dinitrophenylhydrazone from the urine of rats dosed separately with cis-indane-1,2-diol and trans-indane-1,2-diol. 5. Possible mechanisms for the interconversion of cis- and trans-indane-1,2-diol are discussed.

cis-Cyclo-heptane-1,2-diol.

Project description:The title compound, C(7)H(14)O(2), is a vicinal diol derived from cyclo-heptane with cis-orientated hydr-oxy groups. The mol-ecules shows no non-crystallographic symmetry. The O-C-C-O torsion angles of both mol-ecules present in the asymmetric unit [-66.4 (2) and -66.9 (2)°] are similar to those in trans-configured cyclo-hexane derivatives (including pyran-oses) as well as rac-trans-cyclo-heptane-1,2-diol, but smaller than those in trans-configured cyclo-pentane derivatives (including furan-oses). In the crystal structure, O-H⋯O hydrogen bonds furnish the formation of sheets parallel to [110].

Biochemical studies of toxic agents. The metabolic formation of 1- and 2-menaphthylmercapturic acid.

Project description:1. Various derivatives of 1- and 2-methylnaphthalene, of the type C(10)H(7).CH(2)X, were administered to rats and the urines of the dosed animals were examined for the presence of 1- and 2-menaphthylmercapturic acid by chromatographic and isolation procedures. A similar, but more limited, series of experiments was carried out with rabbits. 2. All the compounds were administered by subcutaneous injection with the exception of S-(1- and 2-menaphthyl)-l-cysteine, which were added to the food. 3. 1-Menaphthylmercapturic acid was isolated from the urine of rats after the administration of 1-menaphthyl chloride, bromide, alcohol, acetate and benzoate, S-(1-menaphthyl)-l-cysteine and S-(1-menaphthyl)glutathione. 4. 2-Menaphthylmercapturic acid was isolated from rat urine after administration of 2-menaphthyl chloride, S-(2-menaphthyl)-l-cysteine and S-(2-menaphthyl)-glutathione, and was detected chromatographically after injecting 2-menaphthyl bromide. 5. The corresponding mercapturic acids were isolated after administering 1- and 2-menaphthyl chloride and 1-menaphthyl acetate to rabbits, but not after giving 1- and 2-menaphthyl bromide and 1-menaphthyl alcohol, although chromatographic evidence of mercapturic acid excretion was obtained after injecting these compounds.

Post-pulse addition of trans-cyclohexane-1,2-diol improves electrotransfer mediated gene expression in mammalian cells.

Project description:Electric field mediated gene transfer is facing a problem in expression yield due to the poor transfer across the nuclear envelope. Trans-cyclohexane-1,2-diol (TCHD) was shown to significantly increase chemically mediated transfection by collapsing the permeability barrier of the nuclear pore complex. We indeed observed a significant increase in expression by electrotransfer when cells are treated post pulse by a low non toxic concentration of TCHD. This was obtained for different pulsing conditions, cell strains and plasmid constructs. An interesting improvement in cell viability can be obtained. This can significantly enhance the non-viral gene electrical delivery.

The urinary excretion of S-methylcysteine, methylmercapturic acid, methylthioacetic acid and N-(methylthioacetyl)glycine has been demonstrated by paper chromatography. 3. Methylmercapturic acid and N-(methylthioacetyl)glycine have been isolated from the urine of the dosed animals. 4. The methylthio compounds detected represented about 2% of the iodomethane administered.

Biochemical and physiological properties of rhodopsin regenerated with 11-cis-6-ring- and 7-ring-retinals.

Project description:Phototransduction is initiated by the photoisomerization of rhodopsin (Rho) chromophore 11-cis-retinylidene to all-trans-retinylidene. Here, using Rho regenerated with retinal analogs with different ring sizes, which prevent isomerization around the C(11)=C(12) double bond, the activation mechanism of this G-protein-coupled receptor was investigated. We demonstrate that 11-cis-7-ring-Rho does not activate G-protein in vivo and in vitro, and that it does not isomerize along other double bonds, suggesting that it fits tightly into the binding site of opsin. In contrast, bleaching 11-cis-6-ring-Rho modestly activates phototransduction in vivo and at low pH in vitro. These results reveal that partial activation is caused by isomerization along other double bonds in more rigid 6-locked retinal isomers and protonation of key residues by lowering pH in 11-cis-6-ring-Rhos. Full activation is not achieved, because isomerization does not induce a complete set of conformational rearrangements of Rho. These results with 6- and 7-ring-constrained retinoids provide new insights into Rho activation and suggest a potential use of locked retinals, particularly 11-cis-7-ring-retinal, to inactivate opsin in some retinal degeneration diseases.

The dihedral angle between the two benzene rings of the title compounds is 58.86 (4)°. In the crystal, it displays intra and intermolecular O-H···O hydrogen bonding and intermolecular O-H···Cl hydrogen···chlorine interactions.

Torquoselective ring opening of fused cyclobutenamides: evidence for a cis,trans-cyclooctadienone intermediate.

Project description:Electrocyclic ring opening of 4,6-fused cyclobutenamides 1 under thermal conditions leads to cis,trans-cyclooctadienones 2-E,E as transient intermediates, en route to 5,5-bicyclic products 3. Theoretical calculations predict that 4,5-fused cyclobutenamides should likewise undergo thermal ring opening, giving cis,trans-cycloheptadienones, but in this case conversion to 5,4-bicyclic products is thermodynamically disfavored, and these cyclobutenamides instead rearrange to vinyl cyclopentenones.
