Project description:Two major considerations when encoding pseudo-Boolean (PB) constraints into SAT are the size of the encoding and its propagation strength, that is, the guarantee that it has a good behaviour under unit propagation. Several encodings with propagation strength guarantees rely upon prior compilation of the constraints into DNNF (decomposable negation normal form), BDD (binary decision diagram), or some other sub-variants. However it has been shown that there exist PB-constraints whose ordered BDD (OBDD) representations, and thus the inferred CNF encodings, all have exponential size. Since DNNFs are more succinct than OBDDs, preferring encodings via DNNF to avoid size explosion seems a legitimate choice. Yet in this paper, we prove the existence of PB-constraints whose DNNFs all require exponential size.

Project description:How early does the brain decode object categories? Addressing this question is critical to constrain the type of neuronal architecture supporting object categorization. In this context, much effort has been devoted to estimating face processing speed. With onsets estimated from 50 to 150 ms, the timing of the first face-sensitive responses in humans remains controversial. This controversy is due partially to the susceptibility of dynamic brain measurements to filtering distortions and analysis issues. Here, using distributions of single-trial event-related potentials (ERPs), causal filtering, statistical analyses at all electrodes and time points, and effective correction for multiple comparisons, we present evidence that the earliest categorical differences start around 90 ms following stimulus presentation. These results were obtained from a representative group of 120 participants, aged 18-81, who categorized images of faces and noise textures. The results were reliable across testing days, as determined by test-retest assessment in 74 of the participants. Furthermore, a control experiment showed similar ERP onsets for contrasts involving images of houses or white noise. Face onsets did not change with age, suggesting that face sensitivity occurs within 100 ms across the adult lifespan. Finally, the simplicity of the face-texture contrast, and the dominant midline distribution of the effects, suggest the face responses were evoked by relatively simple image properties and are not face specific. Our results provide a new lower benchmark for the earliest neuronal responses to complex objects in the human visual system.

Project description:We use simple models of the costs and benefits of microbial gene expression to show that changing a protein's expression away from its optimum by 2-fold should reduce fitness by at least [Formula: see text], where P is the fraction the cell's protein that the gene accounts for. As microbial genes are usually expressed at above 5 parts per million, and effective population sizes are likely to be above 10(6), this implies that 2-fold changes to gene expression levels are under strong selection, as [Formula: see text], where Ne is the effective population size and s is the selection coefficient. Thus, most gene duplications should be selected against. On the other hand, we predict that for most genes, small changes in the expression will be effectively neutral.

Project description:PURPOSE:The Cramér-Rao lower bound (CRLB) is widely used in the design of magnetic resonance (MR) experiments for parameter estimation. Previous work has considered only Gaussian or Rician noise distributions in this calculation. However, the noise distribution for multi-coil acquisitions, such as in parallel imaging, obeys the noncentral ?-distribution under many circumstances. The purpose of this paper is to present the CRLB calculation for parameter estimation from multi-coil acquisitions. THEORY AND METHODS:We perform explicit calculations of Fisher matrix elements and the associated CRLB for noise distributions following the noncentral ?-distribution. The special case of diffusion kurtosis is examined as an important example. For comparison with analytic results, Monte Carlo (MC) simulations were conducted to evaluate experimental minimum standard deviations (SDs) in the estimation of diffusion kurtosis model parameters. Results were obtained for a range of signal-to-noise ratios (SNRs), and for both the conventional case of Gaussian noise distribution and noncentral ?-distribution with different numbers of coils, m. RESULTS:At low-to-moderate SNR, the noncentral ?-distribution deviates substantially from the Gaussian distribution. Our results indicate that this departure is more pronounced for larger values of m. As expected, the minimum SDs (i.e., CRLB) in derived diffusion kurtosis model parameters assuming a noncentral ?-distribution provided a closer match to the MC simulations as compared to the Gaussian results. CONCLUSION:Estimates of minimum variance for parameter estimation and experimental design provided by the CRLB must account for the noncentral ?-distribution of noise in multi-coil acquisitions, especially in the low-to-moderate SNR regime. Project description:We report a microfluidic system that physically separates nuclear RNA (nucRNA) and cytoplasmic RNA (cytRNA) from a single cell and enables single-cell integrated nucRNA and cytRNA-sequencing (SINC-seq). SINC-seq constructs two individual RNA-seq libraries, nucRNA and cytRNA, per cell, quantifies gene expression in the subcellular compartments, and combines them to create novel single-cell RNA-seq data. Leveraging SINC-seq, we discover distinct natures of correlation among cytRNA and nucRNA that reflect the transient physiological state of single cells. These data provide unique insights into the regulatory network of messenger RNA from the nucleus toward the cytoplasm at the single-cell level.

Project description:Nonexcisional facial skin tightening has long been an elusive goal in aesthetic surgery. The "treatment gap" includes cases who are not "severe" enough for excisions surgery but not "mild" enough for most traditional noninvasive aesthetic modalities. In this retrospective review, we present the largest evaluation to date of radiofrequency (RF) skin tightening technology combination including bipolar RF (FaceTite; InMode) and fractional bipolar RF (Fractora modified to Morpheus8; InMode).MethodsA multicenter retrospective study was conducted between January 2013 and December 2018 using a combination of bipolar RF and fractional bipolar RF for the treatment of facial aging. Data collection included demographic information, Baker Face/Neck Classification, amount of energy used, adverse events, and patient satisfaction. Four cadaver dissections were also conducted to correlate the underlying neuromuscular anatomy with RF treatment of the lower face and neck.ResultsTwo hundred forty-seven patients (234 women and 13 men) were included in the study. Average age was 55.1 years (SD, ±8), body mass index was 24.3 (±2.4), and 9% (23/247) of patients were active smokers at the time of treatment. Patients had an average Baker Face/Neck Classification score of 3.1 (SD, ±1.4). The procedure was performed under local anesthesia in 240/247 cases (97.2%). Patients objectively improved their Baker Face/Neck Classification score by 1.4 points (SD, ±1.1). Ninety-three percent of patients indicated that they were pleased with their results and would undergo the procedure again. Complications recorded for our cohort included prolonged swelling >6 weeks (4.8%, 12/247), hardened area >12 weeks (3.2%, 8/247), and marginal mandibular neuropraxia (1.2%, 3/247), which all resolved without further intervention. When considering possible control variables, age seems to be a significant factor. That is, older patients were more likely to benefit from a larger magnitude of the treatment effect (as demonstrated by a decrease in the Baker rating from pre- to posttreatment) when compared with younger patients. However, both groups did demonstrate significant improvements across time.ConclusionWhile this combination RF treatment (FaceTite bipolar RF and fractional bipolar RF) does not aim to replace a facelift/necklift in appropriate candidates, it does broaden the plastic surgeons' armamentarium to potentially fill a treatment gap.

Project description:BackgroundLow muscle strength and muscle mass are associated with adverse outcomes in older hospitalized patients. The aim of this study was to assess the association between cognitive functioning and muscle strength and muscle mass in hospitalized older patients.MethodsThis prospective inception cohort included 378 patients aged 70 years or older. At admission patients were assessed for cognitive functioning by use of the Six-Item Cognitive Impairment Test (6-CIT). Muscle strength and muscle mass were assessed using handheld dynamometry and segmental multifrequency bioelectrical impedance analysis, within 48 h after admission and on day 7, or earlier on the day of discharge.ResultsThe data of 371 patients (mean age ± standard deviation 80.1 ± 6.4 years, 49.3% female) were available for analyses. The median (interquartile range) 6-CIT score was 4 (0-8) points. At admission, lower cognitive functioning was associated with lower muscle strength, lower skeletal muscle mass (SMM), lower appendicular lean mass, and lower SMM index. Cognitive functioning was not associated with change in muscle strength and muscle mass during hospitalization.ConclusionThis study further strengthens evidence for an association between lower cognitive functioning and lower muscle strength and muscle mass, but without a further decline during hospitalization.

Project description:MOTIVATION:Scaling by sequencing depth is usually the first step of analysis of bulk or single-cell RNA-seq data, but estimating sequencing depth accurately can be difficult, especially for single-cell data, risking the validity of downstream analysis. It is thus of interest to eliminate the use of sequencing depth and analyze the original count data directly. RESULTS:We call an analysis method 'scale-invariant' (SI) if it gives the same result under different estimates of sequencing depth and hence can use the original count data without scaling. For the problem of classifying samples into pre-specified classes, such as normal versus cancerous, we develop a deep-neural-network based SI classifier named scale-invariant deep neural-network classifier (SINC). On nine bulk and single-cell datasets, the classification accuracy of SINC is better than or competitive to the best of eight other classifiers. SINC is easier to use and more reliable on data where proper sequencing depth is hard to determine. Project description:Sinc-shaped Nyquist pulses possess a rectangular spectrum, enabling data to be encoded in a minimum spectral bandwidth and satisfying by essence the Nyquist criterion of zero inter-symbol interference (ISI). This property makes them very attractive for communication systems since data transmission rates can be maximized while the bandwidth usage is minimized. However, most of the pulse-shaping methods reported so far have remained rather complex and none has led to ideal sinc pulses. Here a method to produce sinc-shaped Nyquist pulses of very high quality is proposed based on the direct synthesis of a rectangular-shaped and phase-locked frequency comb. The method is highly flexible and can be easily integrated in communication systems, potentially offering a substantial increase in data transmission rates. Further, the high quality and wide tunability of the reported sinc-shaped pulses can also bring benefits to many other fields, such as microwave photonics, light storage and all-optical sampling.

Project description:Ritz eigenvalues only provide upper bounds for the energy levels, while obtaining lower bounds requires at least the calculation of the variances associated with these eigenvalues. The well-known Weinstein and Temple lower bounds based on the eigenvalues and variances converge very slowly and their quality is considerably worse than that of the Ritz upper bounds. Lehmann presented a method that in principle optimizes Temple's lower bounds with significantly improved results. We have recently formulated a Self-Consistent Lower Bound Theory (SCLBT), which improves upon Temple's results. In this paper, we further improve the SCLBT and compare its quality with Lehmann's theory. The Lánczos algorithm for constructing the Hamiltonian matrix simplifies Lehmann's theory and is essential for the SCLBT method. Using two lattice Hamiltonians, we compared the improved SCLBT (iSCLBT) with its previous implementation as well as with Lehmann's lower bound theory. The novel iSCLBT exhibits a significant improvement over the previous version. Both Lehmann's theory and the SCLBT variants provide significantly better lower bounds than those obtained from Weinstein's and Temple's methods. Compared to each other, the Lehmann and iSCLBT theories exhibit similar performance in terms of the quality and convergence of the lower bounds. By increasing the number of states included in the calculations, the lower bounds are tighter and their quality becomes comparable with that of the Ritz upper bounds. Both methods are suitable for providing lower bounds for low-lying excited states as well. Compared to Lehmann's theory, one of the advantages of the iSCLBT method is that it does not necessarily require the Weinstein lower bound for its initial input, but Ritz eigenvalue estimates can also be used. Especially owing to this property the iSCLBT method sometimes exhibits improved convergence compared to that of Lehmann's lower bounds.
