Project description:High levels of undiagnosed and untreated HCV infection exist in prison populations globally. Prisons are a key location to identify, treat and prevent HCV infection among people who inject drugs (PWID). Understanding prisoners' lived experiences of the HCV continuum of care informs how HCV care can be effectively delivered to this marginalised and high-risk population. This study aimed to explore Irish prisoners' experience of prison and community-based HCV care. We conducted one-to-one interviews with 25 male prisoners with chronic HCV infection. Data collection and analysis was informed by grounded theory. The mean age of participants and first incarceration was 39.5 and 18.3 years respectively. The mean number of incarcerations was eight. The following themes were identified: medical and social factors influencing engagement (fear of treatment and lack of knowledge, HCV relevance and competing priorities), adverse impact of HCV on health and wellness, positive experience of prison life and health care and the transformative clinical and non-clinical changes associated with HCV treatment and cure. Findings suggest that prison release was associated with multiple stressors including homelessness and drug dependence which quickly eroded the health benefits gained during incarceration. The study generated a substantive theory of the need to increase the importance of HCV care among the routine competing priorities associated with the lives of PWID. HCV infected prisoners often lead complex lives and understanding their journeys through the HCV continuum can inform the development of meaningful HCV care pathways. Many challenges exist to optimising HCV treatment uptake in this group and incarceration is an opportunity to successfully engage HCV infected prisoners who underutilise and are underserved by community-based medical services. Support and linkage to care on release is essential to optimising HCV management.

Project description:Challenges particular to second-year students have been identified that can impact persistence in science, technology, engineering, and mathematics (STEM) fields. We implemented a program to improve student success in intermediate-level science courses by helping students to feel they belonged and could succeed in STEM. We used survey measures of perceptions and attitudes and then qualitative measures to characterize the impact of support strategies, including peer mentoring, a second-year science student retreat, learning and advising support resources, and department-specific activities. Analysis of registration and transcript information revealed underperformance by students of color (SOC) and first-generation (FG) students in 200-level science courses. Comparison of these data before and during programming revealed significant improvement in success rates of these students in 200-level biology and chemistry courses, but success rates of SOC and FG students remain lower than the overall rate for 200-level science courses. Contemporaneous with the program, qualitative and quantitative measures of student attitudes revealed a high level of belongingness and support. The results suggest that a focus on students' metacognition about their own abilities and strategic knowledge of how to succeed may be a fruitful direction for future research.

Project description:Supramolecular systems have applications in areas as diverse as materials science, biochemistry, analytical chemistry, and nanomedicine. However, analyzing such systems can be challenging due to the wide range of time scales, binding strengths, distances, and concentrations at which non-covalent phenomena take place. Due to their versatility and sensitivity, Förster resonance energy transfer (FRET)-based techniques are excellently suited to meet such challenges. Here, we detail the ways in which FRET has been used to study non-covalent interactions in both synthetic and biological supramolecular systems. Among other topics, we examine methods to measure molecular forces, determine protein conformations, monitor assembly kinetics, and visualize in vivo drug release from nanoparticles. Furthermore, we highlight multiplex FRET techniques, discuss the field's limitations, and provide a perspective on new developments.

Project description:The changes of the morphology during heating and the degree of crystallinity of dry and hydrated starch granules are investigated using second harmonic generation (SHG) based Stokes polarimetry. A spatial distribution of various polarization parameters, such as the degree of polarization (DOP), the degree of linear polarization (DOLP), and the degree of circular polarization (DOCP) are extracted and compared with the two dimensional second harmonic (SH) Stokes images of starch granules. The SH signal from hydrated and dry starch on heating differed significantly in DOLP and DOCP values, indicating that hydrated starch has a greater degree of ultrastructural amylopectin disorder. The detail of denaturation and the phase transition of hydrated starch demonstrate the significant influence of thermal processing.

Project description:While research on metacognitive knowledge and metacognitive strategies in second language (L2) writing has proliferated, little attention has been paid to metacognitive experiences in learning to write. This study contributes a novel 6-point Likert scale questionnaire, EFL Learners' Writing Metacognitive Experiences Questionnaire (EFLLWMEQ), and reports insights into learners' metacognitive experiences gathered from its use. The questionnaire was designed to investigate, first, the nature of students' metacognitive experiences when they learn to write in English as a foreign language (EFL) and, secondly, the relationship between students' metacognitive experiences and their writing performance. To this end, the questionnaire was developed and validated with two independent samples of 340 and 540 Chinese undergraduates whose metacognitive experiences were measured as they learned to write in EFL. Data were subjected to exploratory factor analysis (EFA) and confirmatory factor analysis (CFA), respectively. Findings of EFA and CFA revealed a four-factor structure of students' metacognitive experiences of EFL writing: Metacognitive estimates, metacognitive feelings, online task-specific metacognitive knowledge, and online task-specific metacognitive strategies. Results showed that students' metacognitive experiences had positive correlations with their EFL writing test scores. Importantly, the CFA results from the sample of 540 students supported the four-factor correlated model with the best model fit, confirming the validity and reliability of the EFLLWMEQ. This study has theoretical and pedagogical implications for how learners' metacognitive experiences can be managed in learning to write, particularly in EFL classrooms.

Project description:Cyclic GMP-AMP synthase (cGAS) detects cytosolic DNA during virus infection and induces an antiviral state. cGAS signals by synthesis of a second messenger, cyclic GMP-AMP (cGAMP), which activates stimulator of interferon genes (STING). We show that cGAMP is incorporated into viral particles, including lentivirus and herpesvirus virions, when these are produced in cGAS-expressing cells. Virions transferred cGAMP to newly infected cells and triggered a STING-dependent antiviral program. These effects were independent of exosomes and viral nucleic acids. Our results reveal a way by which a signal for innate immunity is transferred between cells, potentially accelerating and broadening antiviral responses. Moreover, infection of dendritic cells with cGAMP-loaded lentiviruses enhanced their activation. Loading viral vectors with cGAMP therefore holds promise for vaccine development.

Project description:Invasive species cause serious environmental and economic harm and threaten global biodiversity. We set out to investigate how quickly invasive plant species are currently spreading in China and how their resulting distribution patterns are linked to socio-economic and environmental conditions. A comparison of the invasive plant species density (log species/log area) reported in 2008 with current data shows that invasive species were originally highly concentrated in the wealthy, southeastern coastal provinces of China, but they are currently rapidly spreading inland. Linear regression models based on the species density and turnover of invasive plants as dependent parameters and principal components representing key socio-economic and environmental parameters as predictors indicate strong positive links between invasive plant density and the overall phytodiversity and associated climatic parameters. Principal components representing socio-economic factors and endemic plant density also show significant positive links with invasive plant density. Urgent control and eradication measures are needed in China's coastal provinces to counteract the rapid inland spread of invasive plants. Strict controls of imports through seaports need to be accompanied by similarly strict controls of the developing horticultural trade and underpinned by awareness campaigns for China's increasingly affluent population to limit the arrival of new invaders. Furthermore, China needs to fully utilize its substantial native phytodiversity, rather than relying on exotics, in current large-scale afforestation projects and in the creation of urban green spaces.

Project description:It is one of the major aims in cancer research to improve our understanding of the underlying mechanisms which initiate and maintain tumor growth and to translate these findings into novel clinical diagnostic and therapeutic concepts with the ultimate goal to improve patient care. One of the greater success stories in this respect has been Waldenström's Macroglobulinemia (WM), which is an incurable B-cell neoplasm characterized by serum monoclonal immunoglobulin M (IgM) and clonal lymphoplasmacytic cells infiltrating the bone marrow. Recent years have succeeded to describe the molecular landscape of WM in detail, highlighting two recurrently mutated genes, the MYD88 and the CXCR4 genes: MYD88 with an almost constant and recurrent point mutation present in over 90% of patients and CXCR4 with over 40 different mutations in the coding region affecting up to 40% of patients. Intriguingly, both mutations are activating mutations leading in the case of CXCR4 to an indelible activation and perpetual signaling of the chemokine receptor. These data have shed light on the essential role of CXCR4 in this disease and have paved the way to use these findings for predicting treatment response to the Bruton tyrosine kinase (BTK) inhibitor ibrutinib and novel therapeutic approaches in WM, which might be transferable to other related CXCR4 positive diseases. Well known for its central role in cancer progression and distribution, CXCR4 is highlighted in this review with regard to its biology, prognostic and predictive relevance and therapeutic implications in WM.

Project description:ContextRecurrent shoulder instability in young athletes can lead to a spectrum of soft tissue and bony lesions that can be bothersome and/or disabling. Coracoid transfer is a treatment option for athletes with recurrent instability.ObjectiveTo report the rate of return to sport for athletes after coracoid transfer.Data sourcesAn electronic search of the literature was performed using the PubMed (MEDLINE) and Cochrane Databases (1966-2018).Study selectionStudies were included if they evaluated return to sport after treatment with coracoid transfer at a minimum 1-year follow-up.Study designSystematic review.Level of evidenceLevel 4.Data extractionData were extracted by 2 authors and included study design, level of evidence, patient demographics (number, age, sex), procedure performed, duration of clinical follow-up, rate of return to sport, patient-reported outcome measures, reoperations, and complications.ResultsFourteen studies met the inclusion criteria. The rate of return to sport at any level ranged from 80% to 100% in all but 1 study (38%), and the rate of return to the previous level of play ranged from 56% to 95% in all but 1 study (16%). Patients returned to sport at an average of 3.2 to 8.1 months. The average patient-reported outcome scores ranged from 78% to 94% (Rowe), 223.6 to 534.3 (Western Ontario Shoulder Instability Index), and 75% to 90% (subjective shoulder value). The rate of postoperative dislocation ranged from 0% to 14%, and the reoperation rate ranged from 1.4% to 13%.ConclusionThere was a high early rate of return to sport in patients who underwent coracoid transfer for anterior shoulder instability, although patients did not reliably return to the same level of play. The procedure had very favorable outcomes for treatment of instability, with low rates of recurrent dislocation and reoperation.
