Investigating on the Methodology Effect When Evaluating Lucid Dream.

ABSTRACT: Lucid dreaming (LD) is a state of consciousness in which the dreamer is aware that he or she is dreaming and can possibly control the content of his or her dream. To investigate the LD prevalence among different samples, researchers have used different types of methodologies. With regard to retrospective self-report questionnaire, two ways of proceeding seem to emerge. In one case, a definition of LD is given to participants ("During LD, one is-while dreaming-aware of the fact that one is dreaming. It is possible to deliberately wake up, to control the dream action, or to observe passively the course of the dream with this awareness"), while in the other instances, participants are presented separate questions targeting specific LD indicators (dream awareness and dream control). In the present study, we measured LD frequency in a sample of French student in order to investigate for possible disparities in LD frequency depending on the type of questionnaire as outlined above. Moreover, we also study links between the prevalence of LD as assessed, respectively, by each questionnaire with various factors such as Vividness of Mental Imagery and Parasomnia. Results revealed no significant difference between LD frequencies across questionnaires. For the questionnaire with definition (DefQuest), 81.05% of participants reported experience of LD once or more. Concerning the questionnaire based on LD indicators (AwarContQuest), 73.38% of participants reported having experienced LD once or more. However, with regard to the correlations analysis, links between LD prevalence and factors such as Vividness of Mental Imagery and Parasomnia, varied across questionnaires. This result is an argument suggesting that researchers should be careful when investigating links between LD and other factors. The type of methodology may influence findings on LD research. Further studies are needed to investigate on the methodology effect in LD research namely on the respective weight of awareness and control.

Project description:The genetic underpinnings of most pediatric-cancer cases are unknown. Population-based studies use large sample sizes but have accounted for only a small proportion of the estimated heritability of pediatric cancers. Pedigree-based studies are infeasible for most human populations. One alternative is to collect genetic data from a single nuclear family and use inheritance patterns within the family to filter candidate variants. This approach can be applied to common and rare variants, including those that are private to a given family or to an affected individual. We evaluated this approach using genetic data from three nuclear families with 5, 4, and 7 children, respectively. Only one child in each nuclear family had been diagnosed with cancer, and neither parent had been affected. Diagnoses for the affected children were benign low-grade astrocytoma, Wilms tumor (stage 2), and Burkitt's lymphoma, respectively. We used whole-genome sequencing to profile normal cells from each family member and a linked-read technology for genomic phasing. For initial variant filtering, we used global minor allele frequencies, deleteriousness scores, and functional-impact annotations. Next, we used genetic variation in the unaffected siblings as a guide to filter the remaining variants. As a way to evaluate our ability to detect variant(s) that may be relevant to disease status, the corresponding author blinded the primary author to affected status; the primary author then assigned a risk score to each child. Based on this evidence, the primary author predicted which child had been affected in each family. The primary author's prediction was correct for the child who had been diagnosed with a Wilms tumor; the child with Burkitt's lymphoma had the second-highest risk score among the seven children in that family. This study demonstrates a methodology for filtering and evaluating candidate genomic variants and genes within nuclear families that may merit further exploration.

Project description:We demonstrate improved methods for making valid and accurate comparisons of fluorescence measurement capabilities among instruments tested at different sites and times. We designed a suite of measurements and automated data processing methods to obtain consistent objective results and applied them to a selection of 23 instruments at nine sites to provide a range of instruments as well as multiple instances of similar instruments. As far as we know, this study represents the most accurate methods and results so far demonstrated for this purpose. The first component of the study reporting improved methods for photoelectron scale (Spe) evaluations, which was published previously (Parks, El Khettabi, Chase, Hoffman, Perfetto, Spidlen, Wood, Moore, and Brinkman: Cytometry A 91 (2017) 232-249). Those results which were within themselves are not sufficient for instrument comparisons, so here, we use the Spe scale results for the 23 cytometers and combine them with additional information from the analysis suite to obtain the metrics actually needed for instrument evaluations and comparisons. We adopted what we call the 2+2SD limit of resolution as a maximally informative metric, for evaluating and comparing dye measurement sensitivity among different instruments and measurement channels. Our results demonstrate substantial differences among different classes of instruments in both dye response and detection sensitivity and some surprisingly large differences among similar instruments, even among instruments with nominally identical configurations. On some instruments, we detected defective measurement channels needing service. The system can be applied in shared resource laboratories and other facilities as an aspect of quality assurance, and accurate instrument comparisons can be valuable for selecting instruments for particular purposes and for making informed instrument acquisition decisions. An institutionally supported program could serve the cytometry community by facilitating access to materials, and analysis and maintaining an archive of results. © 2018 International Society for Advancement of Cytometry.
