Comparison of Image Quality Between "Double Low Dose" Liver CT and Standard Liver CT

ABSTRACT: In patients with malignancies, contrast-enhanced abdominal CT (hereafter abdominal CT) plays an important role in detecting carcinoma recurrence and assessing treatment response. In this study, we aim to investigate whether such a "double low" dose CT is feasible in patients with liver metastases of colorectal cancer using a vendor-agnostic artificial intelligence-based noise reduction and contrast enhancement software.

Single-nucleus multiomic mapping of m6A methylome and transcriptome in native populations of cells with sn-m6A-CT [Bulk CUT&Tag]

Project description:N6-methyladenosine (m6A) RNA modification plays important roles in the governance of gene expression and is temporally regulated in different cell states. In contrast to global m6A profiling in bulk sequencing, technologies for analyzing m6A heterogeneity across single-cells are not extensively established. Here, we report single-nucleus m6A CUT-Tag (sn-m6A-CT) for simultaneous profiling of m6A methylome and transcriptome within a single nucleus. m6A-CT is capable of enriching m6A-marked RNA molecules in situ, without isolating RNAs from cells, in a highly efficient manner. We adapted m6A-CT to the droplet-based single-cell omics platform and demonstrated high throughput performance in analyzing nuclei isolated from thousands of cells from various cell types. We show that sn-m6A-CT profiling is sufficient to determine cell identity and allows the generation of cell-type-specific m6A methylome landscapes from heterogeneous populations. Further, we exemplified the robust deconvolution capabilities in m6A heterogeneities with stem cells in multiple distinct pluripotency states. These indicate that sn-m6A-CT provides additional dimensions to multimodal datasets and new insights into epitranscriptomic landscape in defining cell fate identity and states.

4D Positron Emission Tomography/Computerised Tomography (PET/CT) in Liver Surgical Planning

Project description:Interventions: A standard whole body PET/CT scan will be performed first. This involves a patient lying on a scanning table for around 20 mins while breathing normally. The patient will recieve CT and a PET scan from the base of the brain to thighs. Then at the conclusion of the whole body PET/CT scan the patient will be administered with IV contrast and asked to hold their breath while a high quality CT scan of the liver is acquired taking about 30 seconds. Once the contrast CT scan has concluded the patient will be instructed to breath normally again and a 10 minute gated PET scan of the liver will be acquired. Primary outcome(s): To determine the number of positive liver metastases on contrast enhanced respiratory gated 4D PET/CT scanning compared to conventional ungated whole body PET/CT without contrast-enhanced CT.[At the time of PET/CT scan] Study Design: Purpose: Diagnosis; Allocation: Non-randomised trial; Masking: Open (masking not used);Assignment: Single group;Type of endpoint: Efficacy

Assessing How Normal Variations in CT Scanning Affects Its Interpretation

Project description:The purpose of the study is to see how measurements of tumor differences vary with slight changes in CT scan parameters. Reproducible radiomic features can be extracted for abdominal tumors, and specifically colorectal liver metastases, imaged with clinical CT scanners even in the setting of variable scan parameters and variable contrast timing. Participants will be consented to undergo an additional CT of their abdomen.
