An Analysis of the Response of Human Tumor Microvascular Endothelium to Ionizing Radiation

ABSTRACT: Doctors will take some tissue from the tissue removed during surgery in order to study how the blood vessels of the tumor respond to radiation therapy. The tissue obtained will be used to determine how these tumor blood vessels respond to radiation therapy delivered to the tumor, after it has been removed. This radiation is delivered in the research lab. This research is being conducted in order to develop new methods to treat tumors by radiation therapy. No additional surgery will be performed to obtain these samples, and only materials that remain after all diagnostic testing has been completed will be used.

Project description:Intraoperative Electron Radiation Therapy (IOERT) is a therapeutic technique that delivers a single high dose of ionizing radiation (IR) directly to the tumor bed during cancer surgery. The main goal of IOERT is to counteract tumor growth by acting on residual cancer cells in the site at high risk for recurrence as well as to preserve healthy surrounding tissue from the side effects of radiation therapy. The high IR dose used during IOERT treatment induces a strong stress response resulting in the activation of pro- and anti-proliferative cell signaling pathways in both tumor and normal cells. The radiobiology of healthy tissue response to IR is a topic of interest which may contribute to avoid the impairment of normal tissue/organ function and to decrease the risks of secondary cancers. In this study, we examined changes in gene expression in MCF10A breast epithelial cell line exposed to 9Gy and 23Gy high single dose of IR delivered by IOERT. Changes in gene expression in MCF10A breast epithelial cell line exposed to 9Gy and 23Gy high single dose of IR (named MCF10A_9Gy and MCF10A_23Gy respectively), were analyzed as two-color hybridizations using Agilent Technologies whole human genome 4x44K microarrays

Project description:RATIONALE: Studying samples of tumor tissue and blood from patients with cancer in the laboratory may help doctors learn more about changes that occur in DNA and identify biomarkers related to cancer. It may also help doctors predict how patients respond to treatment. PURPOSE: This phase I trial is studying biomarkers in tumor tissue and blood samples from patients undergoing chemotherapy and radiation therapy for stage II or stage III rectal cancer that can be removed by surgery.

GEP signatures of primary breast cells radiation treated.

Project description:Breast cancer (BC) recovery has considerably increased thanks to the advances achieved from research in this field. However, despite the important results obtained, BC remains a complex multifactorial disease with distinct subtypes associated with different outcomes.So, many efforts from radiobiological research are needed to help clinicians in understanding molecular features of a specific tumor subtype, in order to better define the most successful treatment plan, including the choice of the best Radiation Teraphy (RT) modality and schedule in clinical practice. The aim of the present study was to analyze the GEPs in primary breast cancer cells following irradiation with doses of 9 Gy and 23 Gy electron beam delivered by IOERT treatment, in order to define gene signatures of response to high doses of IR.

Endothelial cell death after ionizing radiation does not impair vascular structure

Project description:The effect of radiation therapy on tumor vasculature has long been a subject of debate. Increased oxygenation and perfusion have been documented during radiation treatments. Conversely, apoptosis of irradiated tumor endothelial cells has been proposed as a major determinant of tumor control. To approach these contradictions, we used multiphoton microscopy in two murine tumor models: MC38, a highly vascularized and B16F10, a moderately vascularized model, grown in transgenic mice with tdTomato labelled endothelium. These tumors received either a single dose of radiation (15 Gy) or a fractionated dose (5x3 Gy). Unexpectedly, even these high doses led to little structural change of the perfused vasculature. However, non-perfused vessels and blind ends were substantially impaired after radiation accompanied by apoptosis of their endothelium. Additionally, proliferation of endothelium was blocked. Further, RNAseq confirmed the modification of gene expression in apoptotic and cell cycle regulation pathways. Therefore, we show that apoptosis of tumor endothelial cells after radiation does not impair vascular structure.

RNAseq of GL261 tumor endothelial cells from wild type and Eltd1-/- mice

Project description:Eltd1 is widely expressed in endothelial cells. We have reported that the expression of Eltd1 is highly up-regulated in glioma vessels. Other studies have shown that Eltd1 is up-regulated in vessels in other types of tumors, and has been suggested as a vascular target for anti-angiogenesis therapy. However, the role of Eltd1 in vessels is largely unknown. We isolated endothelial cells from healthy brain tissue and GL261 tumor tissue, and analyzed the role of Eltd1 in tumor angiogenesis by analyzing the endothelial gene profile from wild type and Eltd1-/- mice.

Lymphatic-Preserving Treatment Sequencing with Immune Checkpoint Inhibition Unleashes cDC1-Dependent Antitumor Immunity in HNSCC

Project description:Despite the promise of immune checkpoint inhibition (ICI), therapeutic responses remain limited. This raises the possibility that standard of care treatments delivered in concert may compromise the tumor response. To address this, we employed tobacco-signature HNSCC murine models in which we mapped tumor-draining lymphatics and developed models for regional lymphablation with surgery or radiation. Remarkably, we found that lymphablation eliminates the tumor ICI response, significantly worsening overall survival and repolarizing the tumor- and peripheral- immune compartments. Mechanistically, within tumor-draining lymphatics, we observed an upregulation of cDC1 cells and IFN-I signaling and show that both are necessary for the ICI response and lost with lymphablation. Ultimately, we provide a mechanistic understanding of how standard oncologic therapies targeting regional lymphatics impact the tumor response to immune- oncology therapy in order to define rational, lymphatic-preserving treatment sequences that mobilize systemic antitumor immunity, achieve optimal tumor responses, control regional metastatic disease, and confer durable antitumor immunity.
