Autohypnosis and Cancerology

ABSTRACT: Therapeutical hypnosis is proven to be an effective medical support to chemotherapy: it was shown that it can reduce the pain, anxiety, fatigue felt by the patient. Yet, hypnosis requires the presence of an hypnotherapist, which is why auto-hypnosis could be an efficient alternative to handle the side effects of chemotherapy. In this study, colorectal cancer and breast cancer patients are either taught auto-hypnosis or are taken in standard care for their chemotherapy. The life quality score (QLQC30) assessed during and after chemotherapies will determine if auto-hypnosis is a good medical support in chemotherapies’ adverse effects management. The proven benefices of auto-hypnosis in the handling of the side effects of chemotherapies could improve the quality of life of cancer affected patients.

Characterization of the Chemotherapy-treated PyMT Matrisome

Project description:Cytotoxic chemotherapy is used to treat many thousands of patients across many cancer types annually. Recent studies have demonstrated that chemotherapy causes systemic response that can be exploited to promote cancer cell survival and dissemination, termed “chemotherapy-induced metastasis. However, there have been no studies investigating how chemotherapy alters the extracellular matrix of breast tumors, or if those changes might help to support metastatic dissemination. Here, we report the first characterization of the chemotherapy-treated breast cancer matrisome using the MMTV-PyMT transgenic mouse model of breast cancer. We identify distinct changes induced by different cytotoxic chemotherapies. In particular, we identify collagen IV as significantly associated with taxane-based chemotherapy treatment. Biological validation confirmed collagen IV as chemotherapy-associated and identified collagen IV-driven Src and FAK signaling as important mediators of invasion in the post-chemotherapy tumor microenvironment.

Sorafenib induces cachexia by impeding transcriptional signaling of the SET1/MLL complex on muscle-specific genes

Project description:Adverse effects of chemotherapies can outweigh the benefits in cancer patients. Various chemotherapeutics are linked to muscle wasting or cachexia, drastically reducing the survival rate of cancer patients. Insights into the molecular basis of chemotherapy-induced cachexia is an unmet need to improve treatment strategies. Here, we demonstrated that Sorafenib- tyrosine kinase inhibitor class of chemotherapeutic agents- induced cachexia. Sorafenib-treated muscle cells show sarcomere disarray, impaired sarcoplasmic reticulum-mediated calcium handling, loss of contractile ability, and reduced mitochondrial oxidative respiratory capacity. To identify the molecular mechanisms underlying Sorafenib-induced cachexia, we performed whole-genome transcriptome profiling or RNA sequencing of muscle cells in response to Sorafenib treatment, compared to DMSO-treated (control) cells. We identified a total of 3030 genes that were downregulated and 2128 genes that were upregulated in Sorafenib-treated cells, as compared to the control. Gene ontology analyses revealed that Sorafenib led to transcriptional downregulation of genes coding for proteins of the sarcomere, sarcoplasmic reticulum, and proteins responsible for muscle contraction, sarcomere organization, and calcium ion handling. Additionally, genes associated with mitochondrial oxidative phosphorylation or electron transport chain were significantly downregulated. In summary, Sorafenib deregulates the transcriptional processes of protein-coding genes associated with sarcomere assembly, calcium ion homeostasis, and mitochondrial oxidative respiration, ultimately leading to impaired muscle function.

Increased glucose availability sensitizes pancreatic cancer to chemotherapy

Project description:Pancreatic cancer (Pancreatic Ductal Adenocarcinoma; PDAC) is highly resistant to chemotherapy. Effective alternative therapies have yet to emerge, and therefore, chemotherapy remains the best available systematic treatment. The discovery of safe and available adjuncts that can improve chemotherapeutic efficacy could potentially improve survival outcomes. We show that a hyperglycemic state enhances the efficacy of conventional single- and multi-agent chemotherapies against PDAC. Molecular analyses of tumors exposed to relatively high glucose levels revealed that GCLC (catalytic subunit of glutamate-cysteine ligase), a key regulator of a metabolic pathway, glutathione biosynthesis, is diminished and underlies chemo-sensitization. Inhibition to GCLC phenocopied the effect of an induced hyperglycemic state in mouse models of PDAC, while rescuing the pathway abrogated the anti-tumor effects observed with chemotherapy treatment under high glucose conditions.

Increased intra-tumoral glucose level is associated with enhanced chemotherapeutic response in pancreatic cancer

Project description:Pancreatic tumors are refractory to current chemotherapies, and there are no effective alternative therapies available for patients with pancreatic cancer (PC). We present an innovative approach to improve chemotherapy efficacy in preclinical models of pancreatic cancer. The effectiveness of various chemotherapies, including standard of care FOLFIRINOX, was remarkably enhanced by inducing a state of hyperglycemia. This was achieved by either providing animals with a high glucose diet or by a pharmacologic method involving streptozotocin coupled with long-acting insulin. Comprehensive integrative analyses revealed a key metabolic pathway, glutathione biosynthesis, mitigated by the high glucose diet. Combining chemotherapies with either the high glucose diet or an inhibitor of glutathione biosynthesis as a feasible approach caused a significant survival benefit in mice bearing orthotopic pancreatic tumors.
