Identification of biomarkers for Miltefosine resistance in Leishmania donovani

Project description:To identify biomarkers specific for Miltefosin resistant parasites, we used DNA microarray to identify differentially expressed genes in resistant parasite in comparision to sensitive and further validated with real time PCR assay and immunoblotting assay Clinically proven miltefosine reistant as well as respondant parasite strains isolated from splenic aspirates and grown in M199 media. For microarray resistant and sensitive samples run in duplicate while for real time PCR, 25 resistant and 33 sensitive strains used. For immunoblotting, confirmed genes were cloned and recombinant protein blotted against miltefosine failure and treated patients sera.

Comprehensive methylome sequencing reveals prognostic epigenetic biomarkers for prostate cancer mortality

Project description:We profiled DNA from primary tumour RP tissue specimens from 15 patients (and adjacent normal tissue specimens from 4 patients) in a discovery cohort using whole-genome bisulphite sequencing (WGBS). We identified a set of differentially methylated regions in the primary tumour samples between patients who died ≤ 10 yrs post-RP and those still alive > 10 yrs post RP (median follow-up time: 19.5 years).

Potential Prognostic Biomarkers for Bone Metastasis from Hepatocellular Carcinoma

Project description:RNA was extracted from 48 pairs of intratumoral and peritumoral formalin-fixed, paraffin-embedded (FFPE) tissue from hepatocellular carcinoma (HCC) patients with and without bone metastases (BM). The Human Cancer Panel DASL Assay containing 502 cancer-related genes was used to identify novel BM-associated genes. A cDNA-mediated annealing, selection, extension, and ligation assay was performed with an array of 502 known cancer-related genes to identify differentially expressed genes in 96 RNA samples. Total RNA was purified from the tissue specimens using the High Pure RNA Paraffin Kit according to the manufacturer’s protocol. DASL experiments were performed to identify genes that were differentially expressed between the BM and NBM groups of matched intratumoral and peritumoral tissues by using A cDNA-mediated annealing, selection, extension, and ligation assay.
