ABSTRACT: This randomized controlled trial aims to investigate whether an individually tailored exercise program and nutritional guide can affect the development of self-reported peripheral sensory neuropathy and fatigue in patients with colorectal cancer receiving adjuvant chemotherapy.

Project description:Pulses are an important food and are consumed as a sustainable source of plant-based proteins. The demand for pulse proteins is continuously increasing due to their nutritional, economic, and ecological values. Although pulse proteins provide many health benefits, they have limitations in terms of sensory attributes and anti-nutritional factors. To overcome these challenges, fermentation technology has been explored as a natural food processing method, as it has the potential to enhance the techno-functional qualities, sensory attributes, and nutritional value of the products. Spontaneous fermentation is a natural process in which the native microbial population grows in the substrate without the addition of specific microbes or spores. There is a knowledge gap regarding proteomic changes that occur during the spontaneous fermentation of legumes. The current study utilized mass spectrometry-based proteomics to investigate the effects of spontaneous fermentation on three different pulse protein isolates (chickpea, faba bean, and lentils).

Expression Data for Cisplatin Treated Dorsal Root Ganglia from A/J and C57BL/6J Mice

Project description:Peripheral sensory neuropathy is a common complication to cisplatin treatment in cancer patients. Multiple signaling pathways are likely to be implicated in the pathophysiology, there are no explicit treatments and little is known about predictive factors associated with the onset or progression of cisplatin induced peripheral neuropathy. The transcriptional profiles of differentially senesitive strains of dorsal root ganglion across two inbreed strain of mice will promote better understanding of progression and risk factors assocciated with cisplatin induced peripheral neuropathy. Peripheral neuropathy was induced with a bi-weekly treatment of 4mg/kg cisplatin. We used microarrays to detail the transciptional profile underlying cisplatin induced peirperal neuropathy in two differentailly sensitve inbreed strains of mice: A/J and C57BL/6J

Rood’s Approach and Oxaliplatin-induced Peripheral Neuropathy in Colorectal Cancer Patients

Project description:Oxaliplatin-induced chronic peripheral neuropathy is of major concern to oncologists and patients as it has been shown to affect patients’ health-related quality of life. Although a number of interventions have been implicated, none of them can be recommended for clinical use. This therapeutic failure reflects a poor understanding of the real mechanism of oxaliplatin-induced neuropathy. However, oxidative stress is identified to be one of the main biomolecular dysfunctions in this neuropathy. Rood’s approach is a neurophysiological approach that is based on reflexes of the central nervous system in which the sensory stimulation provides desired muscular response and was specially designed for patients with motor control problems. It was developed by Margeret Rood in 1940. According to Rood, sensory stimulation can activate or deactivate the receptor by facilitation or inhibition, which makes it possible to get the desired muscular response.

Effect of endurance training on expression signatures of mouse type I and type IIb myofibers

Project description:Skeletal muscle myofibers, categorized into slow-twitch (type I) and fast-twitch (type II) fibers based on myosin heavy chain (MHC) isoforms, exhibit varying fatigue resistance and metabolic reliance. Type I myofibers are fatigue-resistant with high mitochondrial density and oxidative metabolism, while Type II myofibers fatigue quickly due to glycolytic metabolism and fewer mitochondria. Endurance training induces remodeling of myofiber and mitochondrial, increasing slow-twitch myofibers and enhancing mitochondrial oxidative capacity, improving muscle fitness. In our study, conducted using single-cell techniques, we delved deeply into the transcriptomic differences between type I and type IIb myofibers. In response to endurance training, type I myofibers exhibited heightened signals in essential adaptive responses, such as fatty acid oxidation, mitochondrial biogenesis, and protein synthesis, compared to type IIb myofibers. By analyzing untrained myofibers, we identified specific signaling pathways that explain the differences in their responses to endurance training. These findings provide nuanced insights into the molecular mechanisms governing endurance adaptations in fast and slow-twitch muscles, offering valuable guidance for tailored exercise routines and potential therapeutic interventions.

Loss of Neurological Disease HSAN-I-Associated Gene SPTLC2 Impairs CD8+ T Cell Responses to Infection by Inhibiting T Cell Metabolic Fitness

Project description:Hereditary sensory and autonomic neuropathy type I (HSAN-I) is neurological disorder characterized by distal sensory neuron dysfunction, frequent infections, and ulcerative mutilations. It remains unknown if HSAN-I directly dampens protective immunity. Here we report that HSAN-I-causing mutations of serine palmitoyltransferase long chain base subunit 2 (SPTLC2) affect human T cell responses. T cell antigenic stimulation and inflammation induce SPTLC2 expression. Murine T cell-specific ablation of Sptlc2 fundamentally impairs antiviral T cell survival and effector function. Mechanistically, SPTLC2-deficiency reduces sphingolipid biosynthetic flux and causes a prolonged activation of the mechanistic target of rapamycin complex 1 (mTORC1), endoplasmic reticulum (ER) stress and CD8+ T cell death. Antiviral CD8+ T cell responses are restored by supplementing sphingolipids and pharmacologically inhibiting ER stress-induced cell death. Our study reveals that SPTLC2 underpins protective adaptive immunity by translating extracellular stimuli into intracellular anabolic signals and reducing cellular stress to maintain metabolic reprogramming sustainability

Phosphoproteomics of consecuively active AKT in liver and Insulin stimulated primary Hepatocytes

Project description:Liver mitochondria play a central role in metabolic adaptations to changing nutritional states, yet their dynamic regulation upon anticipated changes in the energy state has remained unaddressed. Here, we show that sensory food perception rapidly induces mitochondrial fission in the liver via protein kinase B/AKT-dependent phosphorylation of serine 131 of the Mitochondrial fission factor (MFFS131), a response mediated via activation of hypothalamic Pro-opiomelanocortin (POMC)-expressing neurons. A non-phosphorylatable MFFS131G knock-in mutation abrogates AKT-induced mitochondrial fragmentation in vitro. In vivo, MFFS131G knock-in mice display altered liver mitochondrial dynamics upon refeeding and impaired insulin stimulated suppression of gluconeogenesis. Collectively, we reveal a critical role for rapid activation of a hypothalamic/liver axis to adapt mitochondrial function to anticipated changes of nutritional state in control of hepatic glucose metabolism. R26-fl-Akt-C mice Mice carrying a conditional myristoylation tagged Akt-C transgene in the ROSA26 locus were used to activate AKT in the liver with a liver specific Cre-dependent virus. The generation of this line has been described previously (V. Kohlhaas et al, 2021) For AAV mediated liver-specific delivery of Cre, R26-fl-Akt-C or control mice were injected with a AAV8-TBG-iCre virus (VB1724, Vector Biolabs). This repository contains two experiments a) Liver of liver active Akt-CA and b) Insulin stimulation of primary heptocytes. Please note that replicate one of the hepatocyte dataset have been removed from the analysis due to the limited number of posphosites compared to others.
